Analysis of the fungicidal efficacy, environmental fate, and safety of the application of a mefentrifluconazole and pyraclostrobin mixture to control mango anthracnose

Siwei Wang,Xiaonan Wang,Qiang He,Haidan Lin,Hong Chang,Yanping Liu,Haibin Sun,Xiaobing Song

BACKGROUND Mango anthracnose is among the most severe diseases impacting mango yields and quality. While this disease can be effectively controlled through chemical means, it is vital that appropriate field efficacy and fate determination studies be conducted when applying pesticides to crops in order to appropriately gauge the ecological and health risks associated with the use of these agents. RESULTS GAP field trials were conducted to explore the efficacy, dissipation, and terminal residues associated with the application of mefentrifluconazole and pyraclostrobin to mango crops in six locations throughout China. These analyses revealed that three applications of mefentrifluconazole [160 mg active ingredient (a.i.) kg −1 ] in combination with pyraclostrobin mixture achieved satisfactory disease control efficacy. To simultaneously detect mefentrifluconazole and pyraclostrobin residues on mangoes, a ‘quick, easy, cheap, effective, rugged and safe’ (QuEChERS) high-performance liquid chromatography–tandem mass spectrometry (HPLC-MS/MS)-based approach was established. The initial mefentrifluconazole and pyraclostrobin concentrations ranged from 0.18 to 0.34 mg kg −1 , and these two compounds exhibited respective half-lives of 5.6 to 10.8 days and 5.5 to 9.0 days. At 21 days following foliage application, the terminal mefentrifluconazole and pyraclostrobin residue concentrations were 0.02–0.04 and 0.01–0.04 mg kg −1 , with these concentrations being below the maximum residue limit (MRL) established for pyraclostrobin. Both short-term [acute reference dose percent (ARfD%) 0.78–2.36% and 2.0–6.08%] and chronic [acceptable daily intake percent (ADI%) 0.08–0.47% and 0.09–0.55%] dietary intake risk assessments for mefentrifluconazole and pyraclostrobin indicated that these terminal residue concentrations are acceptable for the general population. CONCLUSION Mefentrifluconazole and pyraclostrobin in mango was rapidly degraded following first-order kinetics models. The dietary risk of mefentrifluconazole and pyraclostrobin through mango was negligible to consumers. The application of a 400 g L −1 mefentrifluconazole–pyraclostrobin suspension concentrate mixture represents a highly efficacious fungicidal approach to controlling mango anthracnose that exhibits significant potential for development as it is easily degraded and associated with low residual concentrations after application. © 2022 Society of Chemical Industry.

Bingna Huang,Feifei Chen,Yue Shen,Changcheng An,Ningjun Li,Jiajun Jiang,Chong Wang,Changjiao Sun,Xiang Zhao,Bo Cui,Zhanghua Zeng,Haixin Cui,Yan Wang

In this study, pyraclostrobin nanocapsules were prepared by in situ polymerization with urea–formaldehyde resin as a wall material. The effects of different emulsifiers, emulsifier concentrations, and solvents on the physicochemical properties of pyraclostrobin nanocapsules were investigated. Solvesso™ 100 was selected as the solvent, and Emulsifier 600# was used as the emulsifier, which accounted for 5% of the aqueous phase system, to prepare pyraclostrobin nanocapsules with excellent physical and chemical properties. The particle size, ζ potential, and morphology of the nanocapsules were characterized by a particle size analyzer and transmission electron microscope. The nanocapsules were analyzed by Fourier-transform infrared spectroscopy, and the loading content and sustained release properties of the nanocapsules were measured. The results show that the size of the prepared nanocapsules was 261.87 nm, and the polydispersity index (PDI) was 0.12, presenting a uniform spherical appearance. The loading content of the pyraclostrobin nanocapsules was 14.3%, and their cumulative release rate was 70.99% at 250 h, providing better efficacy and sustainability compared with the pyraclostrobin commercial formulation. Keywords:pyraclostrobin;nanotechnology;pesticide nanocapsules;in situ polymerization

A pH-responsive vanillin-Schiff base microcapsule for controlled release of pyraclostrobin

Wenneng Wu,Chang Yan,Peng Geng,Peiyi Wang,Hongguo Wu

BACKGROUND Environmentally responsive microcapsules hold great promise for sustainable pest management by improving efficacy, lowering dosage and enhancing environmental compatibility. Herein, a pH-responsive pyraclostrobin (Pyr)-loaded microcapsule (Pyr@PU) with an isocyanate core and vanillin-Schiff base polyurethane shell were successfully fabricated via interfacial polymerization. RESULTS The obtained microcapsules exhibited high loading capacity (80.89%) and encapsulation efficiency (96.17%), indicating excellent drug loading and encapsulation performance. In vitro release experiments confirmed that the Pyr@PU microcapsules exhibited prominent pH-responsive release behavior, with a higher cumulative Pyr release (82.81%) at pH 4.0 after 144 h. Meanwhile, the prepared Pyr@PU microcapsules possessed excellent photostability, favorable leaf wettability and superior rainfastness. Bioactivity assays revealed that the microcapsules exerted favorable antifungal effects. At 200 μg mL −1 , Pyr@PU provided protective and curative efficacy of 79.62% and 67.59% against kiwifruit pathogens, as well as 57.90% and 37.89% on rice leaves, respectively. Biosafety evaluation showed that Pyr@PU reduced the 96-h acute toxicity to zebrafish by 2.3-fold and displayed no obvious adverse effects on rice seed germination. CONCLUSION The fabricated pH-responsive Pyr@PU microcapsule exhibits superior UV resistance, leaf adhesion, rainfastness, antifungal efficacy, and biocompatibility. As a promising intelligent controlled-release platform for pesticides, Pyr@PU provides both control efficacy and environmental compatibility, showing great potential for application in sustainable agriculture. © 2026 Society of Chemical Industry.

Dissipation, residues and risk assessment of oxine-copper and pyraclostrobin in citrus

Xiangwu Liu,Ya Yang,Ya Chen,Qingtao Zhang,Ping Lu,Deyu Hu

An efficient, sensitive, simple and fast method for the simultaneous determination of oxine-copper and pyraclostrobin in citrus fruit was developed and validated. The method uses ethylene diamine tetra-acetic acid as a competitive ligand to convert oxine-copper to soluble 8-hydroxyquinoline for analysis by QuEChERS and LC-MS/MS. Linear relationships were determined with correlation coefficients ranging from 0.9904 to 0.9998. The limits of detection for the analytes were 0.012–0.8 μg kg−1, and the limits of quantitation were 0.04–2.6 μg kg−1 in citrus. The average recoveries were 79.1–114.9% with relative standard deviations of less than 7.4%. The analyses of dissipation indicated that the half-lives of oxine-copper and pyraclostrobin were 1.94–3.67 and 1.79–2.48 days and the terminal residues were <0.08–8.99 and <0.02–1.90 mg kg−1, respectively. The risk quotients of oxine-copper and pyraclostrobin were 0.026–0.199 and 0.003–0.022, respectively. This risk assessment provides a reference for the safe and reasonable use of oxine-copper and pyraclostrobin and may help to establish maximum residue limits for these pesticides in China.

Ecotoxicological Effects of Fungicides Azoxystrobin and Pyraclostrobin on Freshwater Aquatic Bacterial Communities

Lu Tao,Zhou Zhigao,Zhang Qi,Zhang Zhenyan,Qian Haifeng

Extensive use of the fungicides azoxystrobin (AZ) and pyraclostrobin (PYR) can have negative effects on aquatic environments, but comprehensive studies on their effect on aquatic microbial communities are still lacking. We found that AZ and PYR could both inhibit the growth of Chlorella vulgaris , but PYR also inhibited Microcystis aeruginosa more strongly than did AZ. High-throughput sequencing analysis showed that AZ promoted the growth of Cyanobacteria in microcosms, and both PYR and AZ disturbed the ecological balance in the aquatic bacterial community and created distinct ecological risks. Our study suggests that the ecological risk of fungicides is complex, and fungicide use should be better managed to reduce potential risks to the environment.

Mechanistic insights into pyraclostrobin-induced hepatotoxicity in zebrafish via the gut-liver axis

Liruiyuan Gao,Zihao Fu,Juncheng Li,Weiguang Wang,Xia Luo,Liqiang Chen

Pyraclostrobin dysregulates bile acid homeostasis via the gut-liver axis. • Pyraclostrobin disrupts hepatic arachidonic acid steroid and metabolism. • Gut microbiota dysbiosis exacerbates pyraclostrobin hepatotoxicity. • Docking results confirm the inhibition of CYP7A1 and BSEP by pyraclostrobin. Pyraclostrobin is one of the most widely used fungicides globally; however, the mechanisms underlying its hepatotoxicity at environmentally relevant concentrations remain poorly understood. This study demonstrates that exposure to pyraclostrobin at environmentally relevant concentrations (15 and 30 μg/L) induces liver injury in zebrafish by disrupting the homeostasis of the gut-liver axis and bile acid metabolism. The results showed that aquatic exposure to pyraclostrobin at these concentrations leads to the substantial accumulation in the liver and gut tissues of zebrafish. This accumulation caused hepatic vacuolation and dysfunction, intestinal villi defects, impaired gut barrier function, and a significant upregulation of proinflammatory cytokines, including TNF-alpha and IL-1 beta. Integrated multi-omics analysis revealed that pyraclostrobin exposure disrupted arachidonic acid metabolism and steroid hormone biosynthesis in the liver, induced gut microbiota dysbiosis, and exacerbated hepatic immune and metabolic dysfunction by modulating bile acid cycling through the gut-liver axis. Molecular docking studies suggested possible interactions between pyraclostrobin and both the rate-limiting enzyme in bile acid synthesis (cholesterol 7α-hydroxylase; CYP7A1) and the bile salt export pump (BSEP), potentially contributing to bile acid metabolic disorders. This study identifies a novel mechanism through which environmentally relevant concentrations of pyraclostrobin induce liver injury via bile acid accumulation and inflammatory responses through the gut-liver axis. It highlights the necessity of assessing the long-term exposure risks of pyraclostrobin to nontarget organisms at realistic environmental concentrations. Biodegradation of pyraclostrobin by two microbial communities from Hawaiian soils and metabolic mechanism

Xiaoxin Chen,Sheng He,Zhibin Liang,Qing X. Li,Hai Yan,Jiye Hu,Xiaolu Liu

Pyraclostrobin has been widely and long-termly applicated to agricultural fields. The removal of pyraclostrobin from ecological environment has received wide attention. In this study, using sequential enrichments with pyraclostrobin as a sole carbon source, two microbial communities (HI2 and HI6) capable of catabolizing pyraclostrobin were obtained from Hawaiian soils. The microfloras analysis indicated that only Proteobacteria and Bacteroides could survive in HI2-soil after acclimatization, whereas the number of Proteobacteria in HI6-soil accounted for more than 99%. The percentages of Pseudomonas in the HI2 and HI6 microfloras were 69.3% and 59.3%, respectively. More than 99% of pyraclostrobin (C 0  = 100 mg L −1 ) was degraded by the HI2 and HI6 microorganisms within five days. A unique metabolite was identified by high performance liquid chromatography tandem quadrupole time-of-flight mass spectrometry (HPLC-QTOF-MS/MS). A metabolic pathway involving carbamate hydrolysis was proposed. The tertiary amine group of pyraclostrobin was hydrolyzed to primary amine group with the decarboxylation, which facilitated pyraclostrobin detoxification because carboxylester was an important functional group. The metabolic mechanism suggested that Pseudomonas expressing carboxylesterase might be able to degrade carbamate chemicals. Therefore, Pseudomonas might be an ideal candidate for expression and cloning of carbamate-degrading gene in genomics studies. The current study would have important implications in detoxification and bioremediation of carbamates through the C N bond cleavage of methyl carbamate.

Dissipation and Residues of Pyraclostrobin in Rosa roxburghii and Soil under Field Conditions

Lei Han,Qiong Wu,Xiaomao Wu

Rosa roxburghiihas been widely planted in China. Powdery mildew is the most serious disease ofR. roxburghiicultivation. Pyraclostrobin was widely used as a novel fungicide to control powdery mildew ofR. roxburghii. To assess the safety of pyraclostrobin for use onR. roxburghiifruits, its residue rapid analysis as well as an investigation on its dissipation behaviors and terminal residues inR. roxburghiiand soil under field conditions were carried out. The QuEChERS method was simplified using LC–MS/MS detection and combined with liquid–liquid extraction purification to allow determination of pyraclostrobin levels inR. roxburghiifruits and the soil. The fortified recoveries at 0.1~5.0 mg/kg were 93.48~102.48%, with the relative standard deviation of 0.64~3.21%. The limit of detection of the analytical method was 0.16 and 0.15 µg/kg forR. roxburghiifruit and soil, respectively. The effects of different spray equipment and formulations on the persistence of pyraclostrobin inR.roxburghiiwere as follows: gaston gasoline piggyback agricultural sprayer (5.38 d) > manual agricultural backpack sprayer (3.37 d) > knapsack multi-function electric sprayer (2.91 d), suspension concentrate (SC) (6.78 d) > wettable powder (WP) (5.64 d) > water dispersible granule (WG) (4.69 d). The degradation of pyraclostrobin followed the first-order kinetics and its half-lives inR.roxburghiiand soil were 6.20~7.79 days and 3.86~5.95 days, respectively. The terminal residues of pyraclostrobin inR.roxburghiiand soil were 0.169~1.236 mg/kg and 0.105~3.153 mg/kg, respectively. This study provides data for the establishment of the maximum residue limit (MRL) as well as the safe and rational use of pyraclostrobin inR.roxburghiiproduction.Keywords:pyraclostrobin;Rosa roxburghii;soil;dissipation;residue

Acetalated dextran microparticles for the smart delivery of pyraclostrobin to control Sclerotinia diseases

Zhengang Xie,Wenlong Liang,Qiuyu Xiong,Yanyan Zhao,Jingli Cheng,Xianbin Li,Jinhao Zhao

Dextran has emerged as a promising biopolymer carrier for controlled release formulations of pesticides. In this study, pH-sensitive acetalated dextran microparticles (Pyr@Ac-Dex) are prepared to encapsulate and control the release of pyraclostrobin (Pyr). In vitro fungicidal activity experiments showed that the prepared Pyr@Ac-Dex particles show comparable fungicidal ability against S. sclerotiorum compared to that of Pyr technical. In a 10-day pot experiment, the control efficacy of the Pyr@Ac-Dex treatment against S. sclerotiorum (77.1%) is significantly higher than that of Pyr emulsifiable concentrate (Pyr EC) treatment (42.4%). Photodegradation experiments show that compared to Pyr technical, Pyr@Ac-Dex doubles the half-life of Pyr in water. Acute toxicity experiments show that Pyr@Ac-Dex significantly reduced the acute exposure toxicity of Pyr to zebrafish. This study provides an environmentally friendly, feasible, and sustainable strategy for plant disease management.

Pyraclostrobin induces developmental toxicity and cardiotoxicity through oxidative stress and inflammation in zebrafish embryos

Yuanzhao Wu,Yijing Wang,Zan Tong,Weihong Xie,Anli Wang,Chian Song,Weixuan Yao,Jiye Wang

Pyraclostrobin, a typical representative of strobilurin fungicides, is extensively used in agriculture to control fungi and is often detected in water bodies and food. However, the comprehensive toxicological molecular mechanism of pyraclostrobin requires further study. To assess the toxic effects and underlying mechanisms of pyraclostrobin on aquatic organisms , zebrafish embryos were exposed to pyraclostrobin (20, 40, and 60 μg/L) until 96 h post fertilization (hpf). These results indicated that exposure to pyraclostrobin induces morphological alterations, including spinal curvature, shortened body length, and smaller eyes . Furthermore, heart developmental malformations , such as pericardial edema and bradycardia, were observed. This indicated severe cardiotoxicity induced by pyraclostrobin in zebrafish embryos, which was confirmed by the dysregulation of genes related to heart development. Besides, our findings also demonstrated that pyraclostrobin enhanced the contents of reactive oxygen species (ROS) and malondialdehyde (MDA), up-regulated catalase (CAT) activity, but inhibited superoxide dismutase (SOD) activity. Subsequently, the NF-κb signaling pathway was further studied, and the results indicated that the up-regulation of tnf-α , tlr-4 , and myd88 activated the NF-κb signaling pathway and up-regulated the relative expression level of pro-inflammatory cytokines, such as cc - chemokine , ifn-γ, and cxcl - clc . Collectively, this study revealed that pyraclostrobin exposure induces developmental toxicity and cardiotoxicity, which may result from a combination of oxidative stress and inflammatory responses. These findings provide a basis for continued evaluation of the effects and ecological risks of pyraclostrobin on the early development of aquatic organisms .

Jianguo Feng,Zhiyang Chen,Wang Chen,Li Sun,Jinghan Yang,Kangli He,Sa Dong,Shuzhong Yuan

BACKGROUND A controlled-release formulation based on mesoporous silica nanoparticles (MSNs) provides an effective way for reducing pesticide use and protecting the ecological environment. In this study, MSNs loaded with pyraclostrobin (PYR@MSNs) were prepared using a one-pot method. RESULTS The characteristics of PYR@MSNs were systematically investigated, including morphology, loading content, ultraviolet (UV) resistance, release behavior, control effects against pathogens, and safety to nontarget organisms. The results show that the prepared PYR@MSNs presented characteristics of regular spherical shapes, uniform particle size (200 nm), high drug loading (38.9%), and enhanced UV resistance. Compared with traditional formulation, PYR@MSNs exhibited improved control effects against Fusarium graminearum , an extended control period, and lower toxicity to zebrafish, earthworms and BEAS-2B cells. CONCLUSIONS This research will facilitate the development of efficient and safe pesticide delivery systems. The PYR@MSNs has showed its potential as a new controlled-release formulation with increased efficacy and is expected to benefit the sustainable development of agriculture. © 2022 Society of Chemical Industry.

Enhanced fungicidal activity and mechanism of pyraclostrobin nanoparticle with reactive oxygen species responsiveness against Rhizoctonia solani

Ning Zhang,Wei Hu,Ruiquan Hou,Pengrui Du,Xiaoran Miao,Ruifei Wang,Hao Wu,Shengnan Li,Youzhi Li,Zhixiang Zhang,Hanhong Xu

The frequent application of pesticides has caused adverse effects on humans and the environment. Stimuli-responsive nanoparticles can enhance the control efficacy of pesticides to pest insects or fungus and reduce impacts on non-target organisms. In the present work, 4-(hydroxymethyl) phenylboronic acid pinacol ester (PBAP) groups were conjugated onto β-cyclodextrin (β-CD) to form a H 2 O 2 -eliminating material (Ox-bCD), and then the nanoparticiles (PYR@OxbCD) loaded pyraclostrobin (PYR) were successfully fabricated by a nanoprecipitation. The prepared PYR@OxbCD had excellent reactive oxygen species (ROS) release dependency, and significantly enhanced fungicidal activity against Rhizoctonia solani in vitro. PYR@OxbCD could accurately release PYR at infected rice plants, providing a better control efficacy than PYR suspension concentrate (SC). The contact angle of PYR@OxbCD on leaves was reduced, and the spreading and antiphotolysis properties were increased, which may lead to an increase of deposition and a lower dissipation rate . Furthermore, the released PYR from PYR@OxbCD enhanced the activity of peroxidase (POD) and catalase (CAT), and up-regulated the expression of defense genes including PAL , PR10, CHI, CAT, GluPOD and AsPOD. PYR@OxbCD exhibited a much weaker effect on microbial abundance in rice cropping systems. The study provides an idea for smart delivery of fungicides to prevent plant pathogens and offers a more comprehensive view about the resistance of R. solani -infected rice triggered by PYR@OxbCD in levels of phenotype, enzymatic activity , gene expression, and microbiome.
