Royal jelly as a functional food has great potential to promote human health. As a unique ingredient in royal jelly, 10-hydroxy-2-decenoic acid (10-HDA) is a vital criterion to evaluate the quality of royal jelly. Current approaches for 10-HDA detection mainly rely on high-performance liquid chromatographic (HPLC), but it requires large-scale equipment and skilled technicians that are costly and time-consuming. Herein, we report a colorimetric sensing strategy for 10-HDA detection based on Ag(I) and tetramethylbenzidine (TMB), a widely used chromogenic reagent. Ag(I) can oxidize TMB to generate oxidized TMB (oxTMB) showing a light-blue color. Due to the chelation with Ag (I), the introduction of 10-HDA can prevent the generation of oxTMB that results in a color fading and a decrease in absorbance. This colorimetric sensing strategy enables highly sensitive detection of 10-HDA with a linear range from 1 μM to 10 μM and a limit of detection of 0.13 μM. More importantly, this 10-HDA assay allows for a visual readout either by a smartphone or the naked eye with low cost, convenience, and rapid response that has been successfully applied for evaluating the quality of royal jelly in actual samples. The sensing strategy developed in this work provides a new colorimetric approach that holds great promise as a point-of-care platform for the food industry.

Royal jelly produced by nurse bees from tea tree (tea royal jelly) is a special bee product in China and it is popular with consumers owing to its multiple healthcare functions. However, the volatile compound profile of tea royal jelly has not been investigated. To obtain a comprehensive understanding of volatile compound profile of tea royal jelly, a method based on headspace solid-phase microextraction coupled with gas chromatography-mass spectrometry (HS-SPME/GC-MS) and chemometric analysis was applied. Moreover, two royal jelly samples produced from pagoda tree were also investigated to reveal the difference in volatile compounds between royal jelly produced from tea tree and other plants. A total of 66 volatile compounds were identified in tea royal jelly, including 16 alkanes, 14 aldehydes, 9 alcohols, 9 ketones, 8 esters and lactones, 5 phenols, 2 organic acids, and 3 other compounds, and most of them were discovered in royal jelly for the first time. Notably, α -ionone, an important aroma compound in tea, was first identified in tea royal jelly. Moreover, the result of chemometric analysis indicates a significant difference on volatile compound profile between royal jelly produced from tea and pagoda trees. To sum up, 35 differential compounds were identified and they could well distinguish royal jelly samples. Our data offers the first characterization of volatile compound profile of tea royal jelly and significantly expands the knowledge of volatile compounds in royal jelly. It is potentially useful as a new approach to distinguish royal jelly produced from different botanical origins.

Background: Safe and effective wound healing can be a major clinical challenge. Inflammation and vascular impairment are two main causes of inadequate wound healing. Methods: Here, we developed a versatile hydrogel wound dressing, comprising a straightforward physical mixture of royal jelly-derived extracellular vesicles (RJ-EVs) and methacrylic anhydride modified sericin (SerMA), to accelerate wound healing by inhibiting inflammation and promoting vascular reparation. Results: The RJ-EVs showed satisfactory anti-inflammatory and antioxidant effects, and significantly promoted L929 cell proliferation and migration in vitro . Meanwhile, the photocrosslinked SerMA hydrogel with its porous interior structure and high fluidity made it a good candidate for wound dressing. The RJ-EVs can be gradually released from the SerMA hydrogel at the wound site, ensuring the restorative effect of RJ-EVs. In a full-thickness skin defect model, the SerMA/RJ-EVs hydrogel dressing accelerated wound healing with a healing rate of 96.8% by improving cell proliferation and angiogenesis. The RNA sequencing results further revealed that the SerMA/RJ-EVs hydrogel dressing was involved in inflammatory damage repair-related pathways including recombinational repair, epidermis development, and Wnt signaling. Conclusion: This SerMA/RJ-EVs hydrogel dressing offers a simple, safe and robust strategy for modulating inflammation and vascular impairment for accelerated wound healing.

The incidence of hyperlipidemia, a lipid metabolic disorder, continues to increase globally, but current pharmacological therapies are often limited by adverse effects. Royal jelly acid, a unique monounsaturated fatty acid (MUFA) derived from royal jelly, shows therapeutic promise against hyperlipidemia; however, its direct targets and regulatory mechanisms remain unclear. In a diet-induced rat model of hyperlipidemia, royal jelly acid exhibited favorable safety and significantly improved body weight, serum lipid indices, hepatic enzyme activity, and antioxidant capacity, while alleviating hepatic lipid droplet accumulation. Royal jelly acid significantly increased the abundance of short-chain fatty acid (SCFA)-producing intestinal microorganisms, promoted SCFA production, and restored hyperlipidemia-associated intestinal microbial imbalance. Liver transcriptomic and plasma metabolomic analyses demonstrated that royal jelly acid regulated genes involved in glycolipid metabolism, oxidative stress, and inflammatory responses, as well as multiple key metabolites. Integrated multi-omics analysis and molecular docking confirmed that the glycerophospholipid metabolic pathway represents the core metabolic axis mediating these protective effects. Royal jelly acid mainly alleviates hyperlipidemia by enhancing antioxidant and anti-inflammatory defense through the gut–liver axis–mediated SCFAs–glycerophospholipid pathway. These findings support the use of royal jelly acid as a promising MUFA-based therapeutic strategy for hyperlipidemia and provide a scientific basis for its application.

Nano-embedding has appeared as a feasible technology to improve the high-quality utilization of royal jelly (RJ). Therefore, the ionic gelation method was proposed to prepared chitosan nanoparticles loaded with royal jelly (RJNPs) and the characterization and biological activity of RJNPs were evaluated in this study. Fourier-transform infrared spectroscopy, differential scanning calorimetry and x-ray diffraction results showed that the methyl and methylene groups of royal jelly combine with the amino groups of chitosan (CS) to become an amorphous polymer. In addition, the 48.68 % encapsulation efficiency and 31.90 % loading capacity were obtained under the optimal ratio of 1:1 RJ to CS, and the average particle size was <80 nm. The antioxidant activity of RJNPs gradually increased with the increase of the RJ proportion. Interestingly, the antibacterial activity on gram-positive bacteria was better than gram-negative bacteria. Most important, RJNPs exhibited better stability and digestibility rather than single RJ. Overall, these findings indicated that RJ can be embedded in chitosan, and RJNPs exhibited good thermal stability, antioxidant activity, antibacterial activities and bioavailability, which was important for the development and application of the high-quality utilization of RJ.

In recent years, population aging together with the increased prevalence of diabetes and obesity has fuelled a surge in the instances of cutaneous non-healing wounds. Royal jelly (RJ) is a traditional remedy for wound repair; however, the subjacent mechanisms and ingredient profiles are still largely unknown. Our previous study found that Castanea mollissima Bl. RJ (CmRJ-Zj) possessed superior wound healing-promoting effects on both the in vivo and in vitro models than Brassica napus L. RJ (BnRJ-Zj). This study conducted an in-depth investigation on the wound-repairing mechanisms of CmRJ-Zj and BnRJ-Zj to explain the previously observed phenomenon and also comprehensively characterized their constituents. It was found that chestnut RJ could enhance cutaneous wound healing by boosting the growth and mobility of keratinocytes, modulating the expression of aquaporin 3 (AQP3), regulating MAPK and calcium pathways, and mediating inflammatory responses. By employing LC-MS/MS-based proteomic and metabolomic techniques, the comprehensive molecules present in CmRJ-Zj and BnRJ-Zj were elucidated, resulting in a clear discrimination from each other. A total of 15 and 631 differential proteins and compounds were identified, and 217 proteins were newly found in RJ proteome. With bioinformatic functional analysis, we speculated that some differential components were responsible for the wound-healing properties of CmRJ-Zj. Therefore, this study provides an insight into the wound-healing mechanisms of RJ and is the first to explore the compositions of RJ from different nectar plants. It will facilitate the development of therapeutic agents from RJ to treat difficult-to-heal wounds and the distinction of different RJ categories.

Royal jelly (RJ), a natural secretion of nurse bees, is a functional food with health-promoting properties. To investigate RJ quality changes with enhanced output and harvesting time, we compared metabolic profiles of RJ harvested at 24 h, 48 h, and 72 h after larval grafting into queen cells from Italian bees (ITBs) and high RJ-producing bees (RJBs) selected from ITBs. A total of 77 high-abundance compounds were identified with reverse-phase liquid chromatography– and hydrophilic interaction liquid chromatography–high-resolution mass spectrometry. Metabolite abundance, water content, and antibacterial activity were found largely unchanged between both bee strains, indicating that a similar RJ quality was achieved. Notably, the observed 10-hydroxy-2-decenoic acid content in RJBs (1.98%) and ITBs (2.41%) meet the standard for premium RJ in China. RJ harvested at 48 h and 72 h had similar metabolic profiles, indicating little influence of the two harvesting times on RJ quality. The harvesting at 24 h could lead to a higher RJ quality with higher abundance levels of many bioactive compounds and lower water content. Our data based mainly on RJ metabolic composition constitute evidence for mass production of high-quality RJ.

This study aimed to identify novel angiotensin-converting enzyme (ACE)-inhibitory peptides from royal jelly proteins (RJPs) by integratingin silicodigestion, virtual screening, andin vitroevaluation. Three major royal jelly proteins (MRJP1-3) were subjected toin silicodigestion using 16 enzymatic systems, yielding 1411 unique peptides. Virtual screening based on predicted bioactivity, toxicity, water solubility, and ADMET profiles resulted in the selection of 27 candidate peptides. Molecular docking revealed strong binding affinities for these peptides compared with the positive control captopril, among which PYPDWSFAK and RPYPDWSF exhibited potent ACE-inhibitory activity, with IC50values of 110 ± 1.02 μmol/L and 204 ± 0.61 μmol/L, respectively. Kinetic analysis indicated that PYPDWSFAK acts as a mixed-type ACE inhibitor. Docking visualization demonstrated that PYPDWSFAK forms multiple hydrogen bonds with key residues in the ACE active pocket and directly coordinates with the catalytic Zn2+ion. Cellular assays showed that PYPDWSFAK was non-cytotoxic, suppressed Ang II–induced endothelial cell migration, restored NO and ET-1 balance, and enhanced SOD and GSH-Px activities. Overall, this study enriches the repertoire of ACE-inhibitory peptides derived from royal jelly proteins. Furthermore, PYPDWSFAK is identified as a promising ACE-inhibitory peptide with potential for incorporation into natural antihypertensive ingredients or functional foods.

Royal jelly acid is a unique unsaturated fatty acid isolated from royal jelly. Recently, royal jelly acid was proposed to have potential therapeutic effects on hyperlipidemia. However, its effect on hyperlipidemia and the underlying molecular mechanism remains unclear. Therefore, in this study, we analyzed the mechanism of anti-hyperlipidemia of royal jelly acid through animal experiments and plasma metabolomics in conjunction with human network pharmacology and molecular docking analyses. We found that royal jelly acid could significantly decrease the serum lipid levels, ameliorate hepatic pathological injury, and reduce the level of oxidative stress in the experimental rats. A total of 41 key metabolites and 10 hub targets played key roles in the exertion of anti-hyperlipidemic effects, including tumor necrosis factor (TNF), insulin (INS) and epidermal growth factor receptor (EGFR). A total of 24 pathways, including tryptophan, citrate cycle, and arachidonic acid metabolisms, were identified as the key pathways involved in royal jelly acid-alleviated hyperlipidemia. The present findings provide new insights into the pathogenesis, diagnosis, and treatment targets of hyperlipidemia as well as contribute to the development and utilization of royal jelly acid related products.

The stability of active ingredients in royal jelly (RJ) was significantly enhanced. • RJ-SANPs exhibits broad-spectrum antibacterial activity and skin-cell biocompatibility. • RJ-SANPs promote type I collagen synthesis in HFF-1 cells. • Ca 2+ -crosslinked alginate preserves RJ bioactivity via structural stabilization. To improve the stability of the natural bioactive substance royal jelly (RJ), this study prepared RJ-loaded sodium alginate nanoparticles (RJ-SANPs) via Ca 2+ -mediated ion-gelation method. SEM and TEM showed predominantly spherical nanoparticles. FTIR spectroscopy confirmed the formation of hydrogen bonds between RJ and SA, while XRD analysis revealed the coexistence of crystalline RJ domains with the amorphous SA matrix, together confirming its successful encapsulation. Moreover, the RJ-SANPs showed hydrodynamic diameter of 532.63 ± 64.22 nm to 662.73 ± 63.65 nm and its encapsulation efficiency (EE) ranges from 42.33% to 58.62%. The in vitro release of 10-HDA from RJ-SANPs followed a Fickian diffusion mechanism, as described by the Korsmeyer–Peppas model. DSC results indicated that SA's “egg-box” structure enhanced the thermal stability of RJ by altering its thermal behavior. The 1:1 RJ-SANPs exhibited good biocompatibility toward HaCaT and HFF-1 cells. It increased type I collagen content in HFF-1 cells in a concentration-dependent manner, reaching 593.64 ng/mL at 0.025% ( v / v ) ( p < 0.05), and also showed antioxidant activity with a maximum ABTS radical scavenging rate of 62.10%, as well as antibacterial activity against E. coli , P. fluorescens , B. cereus , and S. aureus. However, the low concentrations tested did not significantly upregulate AQP3 expression, which might be attributed to the dose-dependent bioactivity of RJ-SANPs. They retained stability at 4 °C, pH 3.0 – 7.0, and 0 – 300 mmol/L NaCl. The work confirmed that SA encapsulation enhances RJ stability while preserving key bioactivities, suggesting the potential of RJ-SANPs as antioxidant and collagen-promoting functional ingredients for skin-care applications. In the bacterial infection of humans, sepsis is of great concern based on its high fatality rate. Correction of immune dysfunction by targeting dendritic cells (DCs) is a promising strategy for sepsis therapy. Royal jelly, a natural health product, is produced by worker bees for feeding queens. In the study, we evaluated whether royal jelly could attenuate LPS-induced immune dysfunction of DCs. We found that royal jelly reduced the expression levels of mature phenotype (MHCII, CD40, CD80, and CD86), activation marker (CD69), and inflammatory cytokines (TNF-α, IL-6, and IL-10) in LPS-induced DCs. In addition, royal jelly enhanced the endocytosis levels and abrogated allogeneic CD4 + T cell proliferation in LPS-induced DCs. Finally, we found that the Nrf2/HO-1 axis played a vital role in the inhibition of LPS-induced immune dysfunction of DCs by royal jelly. These results indicated that royal jelly had a potential application in sepsis therapy by regulating DC function.

This study aimed to fabricate a polyvinyl alcohol/chitosan (PVA/CS) nanofiber loaded with royal jelly (RJ) using blending electrospinning for potential wound dressings. The different PVA/CS ratios in electrospun nanofibers resulted in continuous nanofibers with an average diameter ranging from 219 to 299 nm. The FTIR spectra indicated that RJ was successfully incorporated into the nanofibers through hydrogen bonding with PVA/CS, which was further confirmed by the subsequent TGA experiments. Meanwhile, the RJ/PVA/CS nanofibers exhibited excellent water vapor permeability and hydrophilic properties. The encapsulation efficiency of RJ reached the maximum value of 89.00 %, while the cumulative release rate was up to 84.87 %. Furthermore, the RJ/PVA/CS nanofibers could inhibit the growth of Gram-positive ( Staphylococcus aureus ) and Gram-negative bacteria ( Escherichia coli ). The optimal PVA/CS ratio was determined to be 7:3, achieving inhibition rates of 97.83 % for S. aureus and 72.08 % for E. coli , demonstrating an excellent antibacterial performance. Therefore, this study successfully fabricated a wound dressing nanofiber with potential antibacterial efficacy.
