The influence of the chemical composition of essential oils of Clausena lansium seeds on the growth of Candida strains

Ma Yinzheng,Wang Yuanxiao,Zhou Xia,Yang Heng,Zhang Huixin,Chen Wenhan,Zhang Haiying,Zhang Yunxia,He Xiaowen

Clausena lansium (Lour.) Skeels seeds have been shown to have diverse beneficial medical value due to their unique active components. This study analysed the composition of essential oils (EOs) of C. lansium seeds and investigated their potential antifungal effects against Candida strains . A total of forty-six components were identified in all samples by gas chromatography-mass spectrometry (GC–MS). The main components were sabinene, β -phellandrene and 4-terpineol. Thirteen EOs of C. lansium seeds were classified into three clusters based on their components. Cluster analysis showed that the difference between the tropics and subtropics was the greatest. These EOs and the three main chemicals showed different antifungal activities against five Candida species ( C. albicans , C. tropicalis , C. glabrata , C. krusei and C. parapsilosis ). The antifungal activity against C. glabrata and C. krusei was higher than that against other Candida strains. EOs of C. lansium seeds displayed noteworthy antifungal activity against both sensitive and fluconazole-resistant strains, with inhibition zone diameters in the range of 9.4–23.4 mm. Comprehensive analysis illustrated the importance of sabinene, β -phellandrene and 4-terpineol to antifungal activity, and there may be some synergistic effects with other components. These results represent the first report about the correlation between the chemical composition of EOs of C. lansium seeds and antifungal activity. Taken together, the results obtained provide scientific evidence for the traditional use of C. lansium seeds waste.

De Novo Biosynthesis of Sabinene from Methanol by Multiple Engineered Pichia pastoris

Chenchen Nan,Xiaolong Guo,Jiale Cheng,Xin Li,Jun Feng,Jufang Wang,Hongxin Fu

Sabinene, a high-value monoterpene with broad industrial applications, is facing growing demand. To enable sustainable and cost-efficient production, we engineered the yeast Pichia pastoris for sabinene production from methanol. Through systematic metabolic engineering, we implemented a multipronged strategy to optimize sabinene biosynthesis: (1) enhancing precursor supply and synthase activity via fusion proteins (ERG20WW-tNPPS1 and ERG20WW-t37SabS1 (H561F)), (2) redirecting carbon flux through the phosphoketolase-phosphotransacetylase pathway, and (3) upregulating the mevalonate pathway via overexpression of a truncated HMGR. The engineered strain achieved record-high sabinene titers of 3.24 g/L in shake flasks and 6.38 g/L in a 3-L bioreactor. Notably, this yield surpasses those achieved with conventional sugar-based feedstocks in other microbial hosts, underscoring the potential of methanol as a superior carbon source for terpenoid biosynthesis. This work not only establishes P. pastoris as a promising platform for sabinene production, but also paves the way for methanol-based biomanufacturing of diverse natural products.

The effect of linalool, limonene and sabinene on the thermal stability and structure of rabbit meat myofibrillar protein under malondialdehyde-induced oxidative stress

Zefu Wang,Zhifei He,Dong Zhang,Xiaosi Chen,Hongjun Li

This study aimed to assess the protective effects of the main components of Zanthoxylum bungeanum Maxim. essential oil (ZBMEO) on the characteristics of rabbit meat myofibrillar protein (MP) under MDA (malondialdehyde) induced oxidation. The MP suspensions were mixed with various concentrations (5 mM, 25 mM and 50 mM) of linalool, limonene, sabinene under MDA-induced oxidation. The retained MDA contents were determined, and the thermal stability and structure of rabbit meat myofibrillar protein were analyzed. Linalool, limonene and sabinene can prevent MDA from binding to MP effectively. The addition of linalool, limonene and sabinene led to the increase in the sulfhydryl and free amine contents, and the decrease in the carbonyl content and surface hydrophobicity compared to control MP. Also, the addition had protective effects on maintaining the secondary structure and morphology of MP under MDA-induced oxidation. The Differential scanning calorimetry (DSC) and SDS-PAGE analysis indicated that linalool, limonene, sabinene had an obvious protective effect on MHC and actin. The above results suggest that linalool, limonene and sabinene could reduce the oxidative damage of MDA to MP, which might have the potential in protecting the structure and physicochemical properties of the protein in foods .
