Target-oriented rational design of composite bio-derived solvents for the green extraction of salvianolic acid B and tanshinones from Salvia miltiorrhiza root

Sheng Wang,Xin Liang,Tengfei Lu,Ying Ma,Ziteng Wang,Xiangyu Zheng,Kaiyu Ruan,Tong Wang

Finding sustainable and efficient ways to extract natural products is becoming increasingly important for both the pharmaceutical and food industries. In this study, we propose a target-oriented approach for the rational design of composite bio-derived solvents, specifically tailored for the simultaneous extraction of salvianolic acid B, tanshinone I, cryptotanshinone and tanshinone IIA from Salvia miltiorrhiza root. We systematically evaluated the extraction efficiency of various bio-derived solvents, including glycerol carbonate, ethylene glycol, levulinic acid, glycerolformal, ethyl lactate and γ-valerolactone, both individually and in combination. Using a mixture design integrated with statistical modeling and optimization algorithms, we identified that the composite solvent system of GF/LA (57.8/42.2, v/v) greatly improved the extraction of both hydrophilic and lipophilic compounds. The extraction yields were significantly higher compared to those obtained using individual bio-derived or traditional solvents, while the overall solvent consumption was lower. Additionally, we explored the use of macroporous adsorption resins to recover the extracted compounds from GF/LA (57.8/42.2, v/v) extracts solution, achieving impressive recovery yields of over 80%. Our findings provide valuable insights into the sustainable extraction of bioactive compounds and suggest practical applications for both analytical and industrial-scale processes, aligning with green chemistry principles.

Functionalized gelatin/poly(l-lactide-co-ε-caprolactone) fibrous membrane promotes scarless wound healing by modulating inflammation and reducing fibrosis

Jing Wen,Shue Jin,Xue Luo,Chunhong Chen,Huan Liu,Yubao Li,Jidong Li

In cases of deep skin defects, spontaneous tissue regeneration and excessive collagen deposition can result in the formation of proliferative scarring. Salvianolic acid B (SAB) demonstrates promising applications in the treatment of fibrotic diseases, including scarring. This study prepared biodegradable gelatin (Gel) and poly(lactic- co -ε-caprolactone) (PLCL) nanofibrous membranes using electrospinning technology and functionalized them with salvianolic acid B to create fibrous membrane dressings with anti-scarring properties. The resulting Gel/PLCL/SAB (GPS) fibrous membrane exhibited good mechanical properties and biodegradability. In vitro experiments demonstrated that the fibrous membranes of Gel/PLCL containing 0.5 wt% SAB (GPS 0.5 ) exhibited good cytocompatibility, regulated macrophage function and polarization, and inhibited fibrosis-related genes, including CD36, α-SMA, and collagen I. In a mouse model of full-thickness skin defects, the GPS 0.5 fibrous membrane effectively modulated the immune microenvironment and promoted vascular regeneration, thereby accelerating skin healing. Furthermore, the GPS 0.5 fibrous membrane reduced the expression of CD36 and TGF-β1, downregulated collagen I and III, and promoted the regeneration of hair follicles, sebaceous glands, and other appendages, ultimately reducing scar formation. Thus, SAB-loaded Gel/PLCL fibrous membranes hold potential as bifunctional skin dressings that promote wound healing and inhibit fibrosis.

Inhibition of Human UGT1A1-Mediated Bilirubin Glucuronidation by Polyphenolic Acids Impact Safety of Popular Salvianolic Acid A/B-Containing Drugs and Herbal Products

Guo Ma,Ying Zhang,Wenyan Chen,Zhifang Tang,Xiaoming Xin,Ping Yang,Xiaoqin Liu,Weimin Cai,Ming Hu

Bilirubin-related adverse reactions (ADR, e.g., jaundice and hyperbilirubinemia) induced by herbs rich in certain polyphenolic acids are widely reported. However, the causes and the mechanisms underlying these ADR are not well understood. The purpose of this article is to determine the mechanism by which certain polyphenolic acids inhibit UGT1A1-mediated bilirubin glucuronidation, leading to jaundice or hyperbilirubinemia. We investigated in vitro inhibitory effects on bilirubin glucuronidation of salvianolic acid A (SAA), salvianolic acid B (SAB), danshensu (DSS), protocatechuic aldehyde (PA), and rosmarinic acid (RA), as well as two Salvia miltiorrhiza injections (DSI and CDI) rich in polyphenolic acids. The results showed that average formation rates of three bilirubin glucuronides displayed a significant difference (p < 0.05) and the formation of monoglucuronide was favored regardless if an inhibitor was present or not. SAA, SAB, DSI, and CDI, but not DSS, PA, and RA, significantly inhibited human UGT1A1-mediated bilirubin glucuronidation via a mixed-type inhibitory mechanism. Average IC50 values of SAA, SAB, DSI, and CDI-mediated inhibition of bilirubin glucuronidation were bilirubin concentration-dependent, and their values (against total bilirubin glucuronidation) were in the range 0.44 ± 0.02 to 0.86 ± 0.04 μg/mL (for SAA), 4.22 ± 0.30 to 12.50 ± 0.93 μg/mL (for SAB), 9.29 ± 0.76 to 18.82 ± 0.63 μg/mL (for DSI), and 9.18 ± 2.00 to 22.36 ± 1.39 μg/mL (for CDI), respectively. In conclusion, SAA and its analog SAB are the main ingredients responsible for inhibition of bilirubin glucuronidation by DSI and CDI, whose use is associated with many high bilirubin-related ADR.

Salvianolic acid B blocks hepatic stellate cell activation via FGF19/FGFR4 signaling

Shuxia Tian,Min Chen,Bing Wang,Yonglong Han,Haonan Shang,Junming Chen

Introduction and objectives The activation of hepatic stellate cells (HSCs) is the main cause of liver fibrosis. The beneficial effects of fibroblast growth factor (FGF) 19 on liver fibrosis were recently reported. The S. miltiorrhiza as well as S. miltiorrhiza derived bioactive chemical components has shown prominent antifibrotic effects in liver fibrosis but the mechanism is still not fully understood. We aimed to investigate the bioactive compounds derived from S. miltiorrhiza which exerts antifibrotic effects in HSCs via regulating FGF19. Materials and methods FGF19 level in culture media was determined by enzyme-linked immunosorbent assay. Cell proliferation was measured by Cell Counting Kit-8 assay. Further, mRNA and protein expressions were assessed by quantitative polymerase chain reaction and western blotting, respectively. Knocking down of FGF receptor 4 (FGFR4) by transfection with siRNA was used to confirm the role of FGF19/FGFR4 signaling. Results Using the human HSC cell line LX-2, we screened several natural products and found that bioactive compounds isolated from Salvia miltiorrhiza , particularly salvianolic acid B, strongly upregulated FGF19 secretion by LX-2 cells. We further showed that salvianolic acid B inhibited lipopolysaccharide (LPS)-induced HSC proliferation and activation. LPS treatment may also reduce the mRNA and protein levels of FGF19 and its receptor FGFR4. Salvianolic acid B treatment restored the impaired expressions of FGF19 and FGFR4. Finally, FGFR4 knockdown abolished the antifibrotic effects of salvianolic acid B in the LPS-induced HSC activation model. Conclusions Salvianolic acid B prevented LPS-induced HSC proliferation and activation by enhancing antifibrotic FGF19/FGFR4 signaling.

Salvianolic acid B attenuates abdominal aortic aneurysm via inhibition of ferroptosis and inflammation

Si-Jia Jin,En-Guang Dou,Lei Wang,Lian-Wen Qi,Feng Wei,Lei Zhang

Background Abdominal aortic aneurysm (AAA) is a life-threatening vascular disorder characterized by progressive aortic wall degeneration, inflammation, and vascular smooth muscle cells (VSMCs) loss. Despite extensive research, no effective pharmacological treatment is currently available to prevent or halt AAA progression. Purpose This study aimed to discover effective therapeutic agents for AAA and identify potential natural compounds with pharmacological efficacy against the disease by targeting matrix metalloproteinase-2 (MMP2), a key enzyme involved in extracellular matrix degradation and aneurysm progression. Methods An integrated screening strategy combining cytotoxicity evaluation, high-content immunofluorescence imaging, and molecular docking was applied to an angiotensin II (Ang II)-stimulated rat aortic smooth muscle cells (RASMCs) model. A total of 138 natural products were systematically assessed. The anti-aneurysmal efficacy of the identified compound was further validated in both CaCl₂-induced and Ang II-infused ApoE -/- mouse models of AAA. Network pharmacology and metabolomic analyses were performed to elucidate the underlying molecular mechanisms. Results Salvianolic acid B (Sal B), a major polyphenol from Salvia miltiorrhiza, was identified as a potent anti‑AAA candidate. It suppressed aneurysm formation, reduced elastin degradation, and attenuated inflammatory infiltration in vivo, while preserving the contractile phenotype, lowering ROS, and inhibiting MMP activity in Ang II‑stimulated RASMCs in vitro. Mechanistically, Ang II suppresses NRF2, leading to downregulation of the System Xc⁻-GPX4 axis and promoting lipid peroxidation and VSMC ferroptosis. This ferroptosis then activates the AGE-RAGE pathway, amplifying inflammation and MMP‑driven matrix degradation. Sal B counteracts this cascade by restoring NRF2 activity, improving lipid metabolism, and inhibiting MMPs, thereby blocking ferroptosis‑initiated inflammation and preserving aortic integrity. Conclusion Salvianolic acid B exerts protective effects against AAA by attenuating oxidative stress, ferroptosis, and inflammation. These findings highlight Sal B as a promising natural therapeutic candidate for the prevention and treatment of abdominal aortic aneurysm.

Salvianolic acid B and Senkyunolide I synergistically alleviate cardiac hypertrophy and regulate MAP3K1 signaling

Liu Changtong,Guo Rui,Zhou Yue,Zhu Miao,Shao Li,Wang Yingchao,Zhao Lu

Background Cardiac hypertrophy, characterized by the thickening of the heart muscle, arises from factors such as hypertension and genetic mutations, often leading to adverse outcomes like heart failure and arrhythmias. Guanxinning tablets (GXNT), a botanical drug composed of the blood-activating herbs Salvia miltiorrhiza Bunge. and Ligusticum striatum DC., are widely used in the treatment of cardiovascular diseases. However, the active ingredients and their molecular mechanisms are yet to be fully understood. Methods We evaluated the anti-hypertrophic effects of GXNT and screened its active substances via cardiac function live-imaging on an aristolochic acid A-stimulated zebrafish cardiac hypertrophy model, and through F-actin immunostaining on a phenylephrine-induced hypertrophic NRCMs model. Additionally, the protective effects of GXNT’s active substances were analyzed in a mouse model of cardiac hypertrophy using echocardiography, histopathology analysis, and western blotting. Results The anti-hypertrophic effects of GXNT were assessed using an aristolochic acid A-stimulated zebrafish model and phenylephrine-induced hypertrophic NRCMs. GXNT demonstrated significant anti-hypertrophic effects in both models. Phenotypic screening identified Senkyunolide I (Sen I) from Ligusticum striatum as the active component in the zebrafish model, while Salvianolic acid B (Sal B) and Rosmarinic acid from Salvia miltiorrhiza emerged as the key anti-hypertrophic compound in NRCMs. In a mouse model of isoproterenol-induced cardiac hypertrophy, Sal B and Sen I showed synergistic effects, improving cardiac function, reducing oxidative stress, and suppressing inflammation. Mechanistically, transcriptomic sequencing highlighted cooperative modulation of MAP3K1 signaling by the two compounds. Notably, siRNA-mediated knockdown of MAP3K1 in cardiomyocytes attenuated the hypertrophic phenotype, supporting its essential role in the pathological process. Molecular docking and dynamic simulations further supported their binding potential to MAP3K1. Conclusion These findings underscore GXNT’s potent anti-hypertrophic effects, possibly driven by the synergistic actions of Sal B and Sen I, and offer insights into its therapeutic potential through MAP3K1 signaling regulation.

Salvianolic acid B alters the gut microbiota and mitigates colitis severity and associated inflammation

Xinyue Wu,Lijuan Wang,Lei Tang,Li Wang,Shuyuan Cao,Qian Wu,Zhan Zhang,Lei Li

Consumption of Salvia miltiorrhiza could improve dextran sulfate sodium (DSS) induced colitis. Whether salvianolic acid B (Sal B) can ameliorate colitis is still unclear. Here, colitis induced by 2.2% DSS was treated with 100 mg/kg Sal B by oral gavage. Sal B improved weight loss, disease activity index, mucosal damage and inflammatory cell infiltration in colitis mice. Sal B could increase the production of acetic acid, propionic acid and valeric acid in colitis mice. The reduction of diversity and richness of gut microbiota in colitis mice was restored after Sal B treatment. Sal B reversed the decrease of S24-7 and the increase of Bacteroides in DSS-treated mice. There were 3 KEGG pathways related to enzyme families, cancer and lipid metabolism enriched in mice colitis after Sal B treatment. In conclusion, the beneficial aspects of dietary Sal B upon colitis are associated with microbial habitat.

Kidney-targeted antioxidant salvianolic acid B nanoparticles restoring lysosome homeostasis for acute kidney injury therapy

Jian-Li Chen,Xiao-Hui Jia,Xuan Wu,Ming-Heng Yuan,Xinyue Xia,Dan Yin,Xu Chen,Ze-Yun Gu,Jia-Zheng Liu,Li-Ping Bai,Kathy Qian Luo,Jianfang Wang,Xiao-Ming Zhu

Acute kidney injury (AKI) is highly correlated with oxidative stress, and the application of antioxidants is one of the promising treatments. Salvianolic acid B (SAB) is a natural polyphenol with powerful antioxidant effects. Due to its inherent characteristic of inferior bioavailability, its clinical application is impeded. In this study, small SAB-incorporated nanoparticles (SFA) were synthesized via the self-assembly of SAB, ferric ions, and bovine serum albumin (BSA). Apart from antioxidant activity, the SFA nanoparticles were found to efficiently restore lysosome dysfunction induced by reactive oxygen species (ROS) in human kidney proximal tubular epithelial HK-2 cells. The small size and BSA incorporation make the SFA nanoparticles to be promising AKI-targeting nanoparticles. They are rapidly accumulated and long-term retained in injured kidneys of glycerol-induced AKI mice, especially in the proximal tubules which are the key lesion location during AKI. In vivo studies indicate that SFA nanoparticles show a superior protective effect compared with the free drug by suppressing oxidative stress and maintaining lysosome homeostasis indicated by enhanced heme oxygenase-1 (HO-1) expression and restored cathepsin B (CTSB) activity, respectively. In summary, this study develops a delivery system for the antioxidant SAB enhancing local delivery and offering an additional lysosome restoring mechanism for AKI therapy.

The mitophagy targeted nano-assemblies with high Salvianolic acid B -loading sufficiently enhances osteoarthritis therapy

Yulin Jiang,Mingzhu Wang,Hanshuang Liang,Xuejie Hu,Liuwei Chu,Ranran Yang,Kang Wang,Ruhong Fang,Cailiang Shen,Xinming Wang,Wei Wei,Qingtong Wang

Osteoarthritis (OA) is characterized by chondrocyte degeneration and associated intracellular organelle dysfunction. Restoring impaired mitophagy represents a critical strategy for ameliorating OA-induced chondrocyte dysfunction and promoting cartilage repair, yet a significant therapeutic gap persists due to the lack of highly potent agents that can effectively restore mitophagy in OA chondrocyte. In this work, we identified Salvianolic acid B (SaB), a novel small-molecule agent, which binds to PINK1/Parkin proteins to stabilize them, promote their expression, and activate mitophagy. SaB nano-assemblies (SaB NAs) were fabricated via hydrogen bonding and π–π stacking between SaB and triphenylphosphine, achieving exceptional drug loading (74.4%) and mitochondrial-targeted synergistic therapeutic effects. In vitro and in vivo assessments confirmed that SaB NAs significantly outperformed free SaB in restoring mitochondrial function, facilitating chondrocyte recovery, prolonging therapeutic residence time and alleviating OA progression. The intra-articular delivery of SaB NAs represents a promising approach for treating OA and other joint inflammatory diseases.

Mechanistic Chemistry of Extraordinary Capacity of Salvianolic Acid B on Oxidatively-damaged Mesenchymal Stem Cells

Tingting Wang,Xican Li,Yunrong Li,Jianrun Wu,Yuyin Huang,Gang Wei,Dongfeng Chen

As the main bioactive component of Chinese herbal medicine Danshen, salvianolic acid B (Sal B, or lithospermic acid B) was observed to maintain the viability of mesenchymal stem cells (MSCs) treated by Fenton's reagent in the study. Interestingly, at a higher concentration, Sal B could even increase the viability rate to 175.1%. In mechanistic analysis experiments, Sal B was found to resist DNA destruction by •OH radical, scavenge various radicals in vitro , reduce Cu 2+ → Cu + , chelate Fe 2+ to yield an absorption maximum at 710 nm. Based on these results, we concluded that, (1) Sal B can not only protect MSCs against •OH-induced damages, but also promote their proliferation. This extraordinary capacity makes Sal B an ideal candidate in MSCs transplantation especially when MSCs are polluted by iron-overload or other oxidative stress factors and, can partly be responsible for the versatile properties of Sal B in pharmacology. The possible mechanisms of its protective effect are hypothesized to include Fe 2+ chelating, and direct radical scavenging which is involved in electron transfer (ET) or hydrogen atom transfer (HAT) from the catechol moieties. Its proliferation-promoting effect is presumed to be from its ester group, carboxylic group, or benzofuran ring.

Functionalized biomimetic nanoparticles loaded with salvianolic acid B for synergistic targeted triple-negative breast cancer treatment

Nuo Cheng,Qianqian Zhou,Zongfang Jia,Yang Mu,Sheng Zhang,Lei Wang,Yunna Chen

The therapeutic effect of immune checkpoint inhibitors (ICIs) in triple-negative breast cancer (TNBC) is unsatisfactory. The immune "cold" microenvironment caused by tumor-associated fibroblasts (TAFs) has an adverse effect on the antitumor response. Therefore, in this study, mixed cell membrane-coated porous magnetic nanoparticles (PMNPs) were constructed to deliver salvianolic acid B (SAB) to induce an antitumor immune response, facilitating the transition from a "cold" to a "hot" tumor and ultimately enhancing the therapeutic efficacy of immune checkpoint inhibitors. PMNP-SAB, which is based on a mixed coating of red blood cell membrane and TAF membrane (named PMNP-SAB@RTM), can simultaneously achieve the dual effects of "immune escape" and "homologous targeting". Under the influence of an external magnetic field (MF), SAB can be targeted and concentrated at the tumor site. The SAB released in tumors can effectively inhibit the production of extracellular matrix (ECM) by TAFs, promote T-cell infiltration, and induce antitumor immune responses. Ultimately, the combination of PMNP-SAB@RTM and BMS-1 (PD-1/PD-L1 inhibitor 1) effectively inhibited tumor growth. Finally, this study presents a precise and effective new strategy for TNBC immunotherapy on the basis of the differentiation of "cold" and "hot" microenvironments.

Protective effects and potential mechanism of salvianolic acid B on sodium laurate-induced thromboangiitis obliterans in rats

Ziru Zhang,Jianbo Ji,Dawei Zhang,Maoqiang Ma,Longru Sun

Background The root of Salvia miltiorrhiza f. alba (RSMA) (Lamiaceae) is used for the treatment of patients with thromboangiitis obliterans (TAO) in traditional Chinese medicine . Previously, a mixture of phenolic acids extracted from RSMA has shown significant protective effects on TAO rats. Purpose This study investigates the inhibitory effects of salvianolic acid B on TAO induced by sodium laurate injection in rats to explore the effective constituents of RSMA in TAO treatment. Methods TAO rats were developed using injected sodium laurate. After treatment with ligustrazine hydrochloride (15 mg/kg) and various doses of salvianolic acid B (10, 20, 40 mg/kg) by tail intravenous injection , levels of thromboxane B 2 (TXB 2 ), 6-keto-prostaglandin F 1α (6-keto-PGF 1α ) and endothelin-1 (ET-1) in plasma were determined using enzyme-linked immunosorbent assay. The right femoral arteries were studied by hematoxylin and eosin staining and immunohistochemical analysis to determine pathological changes and overexpression of tumor necrosis factor (TNF)–α and inducible nitric oxide synthase (iNOS) in the femoral artery walls of TAO rats. Results Salvianolic acid B significantly decreased the expressions of TXB 2 and ET-1 and increased the expression of 6-keto-PGF 1α in plasma, and significantly inhibited the overexpression of TNF-α and iNOS in the femoral artery walls of TAO rats at medium and high doses. Conclusion Salvianolic acid B has a protective effect on TAO rats. The mechanism may involve inhibition of thrombosis and TAO-associated inflammatory responses, which may explain the success of RSMA treatment of TAO in humans in traditional Chinese medical practice. Hence, it may be a potential drug for TAO treatment in conventional medicine.
