Sesamol ameliorates hepatic lipid accumulation and oxidative stress in steatosis HepG2 cells via the PPAR signaling pathway

Longkai Shi,Emad Karrar,Xingguo Wang

Non-alcoholic fatty liver disease (NAFLD) is a chronic hepatopathy caused by disordered lipid metabolism in the liver. Sesamol, a phenolic compound derived from sesame oil, has been shown to inhibit obesity, hyperlipidemia, and atherosclerosis in previous investigations. However, the preventive effect of sesamol against hepatic steatosis and oxidative stress in NAFLD has not been well-studied. In this work, sesamol was observed to alleviate lipid accumulation and oxidative stress in high oleic acid (300 μM)/cholesterol (25 μM) induced HepG2 cells, thus indicating that sesamol was involved in regulating hepatic lipid metabolism and oxidative injury. Mechanism studies found that the activated peroxisome proliferator-activated receptors (PPAR) signaling pathway by sesamol intervention up-regulated gene and protein expressions related to fatty acid oxidation and cholesterol efflux and catabolism, thus accelerating lipid consumption and reducing intracellular lipid accumulation in the process of NAFLD. These data suggested that sesamol can effectively ameliorate hepatic steatosis and sesamol riched sesamol oil may be a potential agent for finding therapeutic strategies to treat the NAFLD. Practical applications Sesamol and sesamol-rich sesame oil have received much attention due to their performance on hepatic lipid regulation. The results of this study indicate that sesamol treatment could ameliorate hepatic steatosis by inhibiting lipid accumulation and oxidative stress, thus demonstrating that sesamol and sesame oil can be used for functional foods and nutraceutical applications in the future. In addition, the present work provides knowledge of the effects of sesamol on NAFLD and involved mechanisms, and further supplies nutritional guidelines for sesame oil consumption.

Electrospinning of gelatin nanofibers containing sesamol nanoparticles

Shiqing Li,Wenhui Shi,Xiaoyan Wang,Shujing Li,Xiaojing Pei,Yifan He

The aim of this study was to analyze the bioactivity of gelatin nanofiber membranes based on electrospinning different concentrations of sesamol nanoparticles. Sesamol (Ses) can be encapsulated with zein and chitosan to form nanoparticles (Ses-NPs). Then, Ses-NPs were mixed into gelatin nanofiber membrane (G-Ses-NPs-NF) by the electrospinning method. The results showed that Ses provided the nanofiber membranes with antioxidant and antibacterial activities. When the concentration of Ses was 1.125 mg/mL, 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2′-azinobis-(3-ethylbenzthiazoline-6-sulphonate) (ABTS) radical scavenging rates were 80.36 ± 0.62% and 91.00 ± 0.38%, respectively. In addition, G-Ses-NPs-NF can inhibit Escherichia coli (E. coli), Staphylococcus aureus (S. aureus) and Candida albicans (C. albicans). Additionally, when added with zein-chitosan and gelatine, the functional nanofiber membrane exhibited sustained-release capabilities. Therefore, G-Ses-NPs-NF can be considered as a material with good biologically activity, which has research value in the development of functional food and other fields.

Synthesis and isomeric effect of ethynyl-containing sesamol-based monobenzoxazine resins

Min Zhong,Weichen Sheng,Jianfeng Chen,Han Zhang,Hongping Li,Kan Zhang

The objective of this paper is to conduct a comparative analysis of sesamol-based benzoxazine monomers derived from anilines with an ethynyl substituent in a different position and to obtain some laws to guide the design of thermosets. The synthesis of bio-benzoxazine monomers (S-a, S- o ea, S- m ea, and S- p ea) via a one-step method from sesamol, a biomass monophenol, in the presence of polyformaldehyde by combining four different amines (aniline, ortho -ethynyl aniline, meta -ethynyl aniline, and para -ethynyl aniline), respectively. The structures of four new benzoxazine monomers were characterized by 1 H NMR, 13 C NMR, 2D-HMQC, and FT IR analysis. The FT IR spectra calculated by Gaussian 16 exhibited a high degree of consistency with the experimental data. The DSC scanning results of the monomers demonstrated that the order of the maximum exothermic peak temperature of polymerization was as follows: S- o ea (198.3 °C) < S- p ea (202.1 °C) < S- m ea (216.8 °C) < S-a (240.2 °C), which is in general agreement with the energy level order of HOMO-LUMO, as determined by Gaussian analysis. In addition, the reaction mechanisms of the oxazine ring and the ethynyl moiety were proposed in conjunction with an investigation of the polymerization process by in-situ FT IR. The TGA and MCC test results showed that the polymers (poly(S- o ea), poly(S- m ea), and poly(S- p ea)) derived from the corresponding monomers with ethynyl substituent in various site exhibit both excellent thermal stability and flame retardancy. Notably, poly(S- p ea) exhibited the highest carbon yield (70.2 %) at 800 °C. The incorporation of the ethynyl group not only reduces the polymerization process of benzoxazine monomers but also enhances the performance of resulting polybenzoxazines. This work offers a valuable reference for the preparation of optimal benzoxazine structures in future research.

Selective recognition of sesamol using molecularly imprinted polymers containing magnetic wollastonite

Xue Wang,Jianming Pan,Wei Guan,Xiaohua Zou,Pengwei Huo,Yongsheng Yan,Wei Hu

The magnetic molecularly imprinted polymers (MMIPs) have been synthesized using piperonal molecules as dummy template and magnetic wollastonite composites as support. The resulting composites were applied to selective recognition of sesamol from aqueous solution. MMIPs were characterized by scanning electron microscopy (SEM), Fourier transform infrared (FT-IR) analysis, transmission electron microscopy (TEM), energy dispersive spectroscopy (EDS), thermogravimetric analysis (TGA), and vibrating sample magnetometer (VSM). MMIPs were demonstrated with an imprinted polymer film (90–100 nm) and exhibited magnetic property ( M s =8.60 emu/g) and thermal stability. The effective average diameter of the Fe 3 O 4 nanoparticles was about 10 nm and the thickness of magnetic film was about 160 nm. Batch mode adsorption studies were carried out to investigate the specific adsorption equilibrium, kinetics and selective recognition. The Langmuir isotherm model was fitted to the equilibrium data better than the Freundlich model, and the kinetic properties were well described by the pseudo-second-order equation. The value of E a in this study was 5.35 kJ/mol for MMIPs. In addition, the selective recognition experiments demonstrated that MMIPs have specific recognition ability toward sesamol.

Synthesis and insecticidal activity of sulfonate derivatives of sesamol against Mythimna separata in vivo

Zhi-Ping Che,Jin-Ming Yang,Xi-Jie Shan,Yue-E Tian,Sheng-Ming Liu,Xiao-Min Lin,Jia Jiang,Mei Hu,Gen-Qiang Chen

A series of sulfonate derivatives of sesamol were synthesized and evaluated for their insecticidal activity against a crop-threatening agricultural pest, the pre-third-instar larvae of Mythimna separata in vivo. Among all the target compounds, compounds 3b, 3g, 3h, and 3p exhibited more promising insecticidal activity than sesamol and toosendanin, and the final mortality rates (FMRs) of 3b, 3g, 3h, 3p, 1, and toosendanin were 60.7%/60.7%/67.9%/53.6%/32.1%/50.0%, respectively. Especially compound 3h exhibited the most potent insecticidal activity with FMRs of 67.9%. This suggested that a 4-fluorophenylsulfonyl group introduced at the hydroxyl position of sesamol was necessary for obtaining the most potent compound.

Exploring the potential of sesamol/asarinin-enriched sesame oil in skin lightning and wound repair

Yuan-Qing Luo,Zhi-Qi Li,Qi Wang,Jing-Wen Qin,Tong Yu,Hua-Min Liu,Jin-Chu Yang

Sesame oil enriched with sesamol (SRO)/asarinin (ARO) is obtained by a catalytic process. • SRO and ARO show distinct lignan profiles and skin-protective activities from CSO. • SRO showed strongest skin lightning efficacy via tyrosinase inhibition. • ARO exhibited the best overall skin wound repair efficacy in zebrafish assays. • Lignan-enriched processing technology enhances the skin care efficacy of sesame oil. Driven by the demand for natural skincare actives, this study explored high-value applications of functional sesame oil by utilizing processing-induced endogenous lignan conversion. A controlled solid-acid catalytic process was employed to convert cold-pressed sesame oil (CSO) into sesame oil enriched with either sesamol (SRO) or asarinin (ARO), without significantly altering the fatty acid composition. Meanwhile, the content of γ-tocopherol and the major phytosterols decreased after catalytic treatment, with no significant difference observed between SRO and ARO. In vitro and in vivo comparative evaluations revealed the unique bioactivities of the three oils. SRO showed remarkable skin-lightning potential, exceeding CSO by 88.9 percentage points in mushroom tyrosinase inhibition at 1000 μg/mL. Zebrafish assays confirmed that SRO suppresses melanin expression via tyrosinase inhibition. Molecular docking suggests that sesamol in SRO may inhibit catalytic activity by interacting with key histidine residues in the catalytic center of tyrosinase. Meanwhile, ARO displayed superior wound repair efficacy, improving tail fin regeneration efficacy by 44% and 56% relative to SRO and CSO, respectively. In contrast, SRO exhibited stronger anti-inflammatory activity at low concentrations (5 mg/mL), with a 65% higher neutrophil clearance efficacy than CSO. CSO consistently showed the weakest effects. The distinct bioactivities between SRO/ARO and CSO are plausibly linked to their differential lignan profiles, particularly the enrichment of sesamol and asarinin. These findings suggest that processing-induced endogenous lignan conversion is an effective strategy for selectively modulating the biological functions of sesame oil, thereby enhancing its application value. Sesamol Ameliorates Cholestatic Liver Injury by Modulating the Liver-Gut Axis and EGFR-FXR Signaling Pathway

Shasha Chen,Wenwen Li,Xinran Zhu,Yanlu Han,Mengyao Wang,Ke Gong,Jie Xiao,Chenzhong Liao,Yajun Duan,Xiaohua Jiang,Shuang Zhang

Estrogen-induced cholestasis (EIC) is a condition associated with intrauterine fetal death, preterm delivery, and meconium in amniotic fluid. The role of sesamol, a major bioactive compound in sesame oil, in alleviating cholestasis remains unclear. In this study, we investigated the mechanisms underlying the protective effects of sesamol against cholestatic liver injury using C57BL/6 male mice with 17α-ethynylestradiol (EE)-induced cholestasis, combined with network pharmacology, 16S rRNA sequencing, and the in vitro assays. Our results demonstrate that sesamol alleviates cholestatic liver injury by inhibiting inflammation, oxidative stress, and apoptosis. Mechanistically, sesamol activates EGFR/FXR/CYP7A1 signaling pathway, thereby improving bile acid metabolism. Additionally, sesamol modulates the gut microbiota by increasing the relative abundance of Firmicutes and Bacteroidota ( P < 0.05), which are crucial for bile acid metabolism. Furthermore, we observed a positive correlation ( P < 0.05) between serum liver injury and cholestasis relative markers and the abundance of certain harmful bacteria ( Anaerovorax, Erysipelatoclostridium, Fusobacterium ), suggesting that sesamol may protect against EIC through modulation of the liver-gut axis, contributing to the restoration of bile acid homeostasis. This study provides new insights into the potential of sesamol as a functional food ingredient for the prevention and treatment of cholestasis.

Enhancement of lipid accumulation and docosahexaenoic acid synthesis in Schizochytrium sp. H016 by exogenous supplementation of sesamol

Zhendong Bao,Yuanmin Zhu,Yumei Feng,Kai Zhang,Meng Zhang,Zhikuan Wang,Longjiang Yu

Schizochytrium sp. is one of the most promising marine oleaginous microorganisms for industrial production of docosahexaenoic acid (DHA). In this study, the exogenous supplementation of 1 mM sesamol to the fermentation medium effectively prevented the peroxidation of polyunsaturated fatty acids in the fermentation process, which thereby significantly increasing the lipid and DHA yield by 53.52% and 78.30%, respectively. The addition of sesamol also increased the total antioxidant capacity of cells and induce the gene expression of polyketide synthase and antioxidant enzyme system. Moreover, the supply of nicotinamide adenine dinucleotide phosphate was regulated by sesamol by inhibiting the malic enzyme activity and promoting the glucose-6-phosphate dehydrogenase activity. Finally, fed-batch fermentation showed that the addition of sesamol significantly enhanced the DHA yield by 90.76%. This study provides an important reference for enhancing the DHA productivity of Schizochytrium sp. in industrial fermentation.

The inhibitory effects of sesamol and sesamolin on the glycidyl esters formation during deodorization of vegetables oils

Lipeng Han,Jiahui Li,Shujie Wang,Weiwei Cheng,Lukai Ma,Guoqin Liu,Dongxue Han,Li Niu

BACKGROUND Glycidyl esters (GEs) have attracted worldwide attention for their potential harm to human health. The GEs in edible oils mainly form during the deodorization of the oil refining processes. We used sesamol and sesamolin to inhibit the formation of GEs in model corn oil (MCO), model palm oil (MPO) and model rice bran oil (MRO) during a deodorization process. RESULTS The results showed that, in the three model oils, the total GE content was in the following order from highest to lowest: MRO (1437.98 μg kg −1 ) > MPO (388.64 μg kg −1 ) > MCO (314.81 μg kg −1 ). The inhibitory effect of the three antioxidants on the formation of GEs in the MCO was in the following order from strongest to weakest: tert-butylhydroquinone (TBHQ) > sesamol > sesamolin. CONCLUSION When the mass percentage of sesamol was 0.05%, its inhibition percentage on GEs was close to the inhibition percentage of 0.02% added TBHQ. The present study provides a foundation for understanding how to inhibit the formation of GEs in oils by adding sesamol during the deodorization process.

Natural products as sources of new antioxidants: Synthesis and antioxidant evaluation of Mannich bases of novel sesamol derivatives

Yong Guo,Jiangping Fan,Lailiang Qu,Chongnan Bao,Qian Zhang,Hong Dai,Ruige Yang

Sesamol, a natural phenolic compound with good antioxidant activity, is a nonoil component of sesame seed oil. Two series of novel Mannich bases, 4-(aminoalkyl)-6-allyl-sesamols were synthesized from sesamol, substituted benzaldehydes and piperidine/morpholine via the one-pot reaction. An efficient way using toluene as solvent, catalyst-free and one-pot reaction for the synthesis of these sesamol derivatives was developed. The structures of all 4-(aminoalkyl)-6-allyl-sesamols were determined by spectral analysis. Compound 6f was unambiguously confirmed by X-ray diffraction further. The antioxidant assays have demonstrated that the sesamol derivative 7i displayed excellent antioxidant activity. The structure–activity relationships (SARs) of these sesamol derivatives were also observed. Further, the representative compounds 7a , 7g and 7i exhibited relative low toxicity to normal cells. Based on these characteristics, these sesamol derivatives will have great potential application in food and drugs.

Sesamol serves as a p53 stabilizer to relieve rheumatoid arthritis progression and inhibits the growth of synovial organoids

Xian Lin,Tengyu Lin,Xiaocheng Wang,Jiaxin He,Xu Gao,Shuyan Lyu,Qingwen Wang,Jian Chen

Background Rheumatoid arthritis (RA) is an autoimmune disease known as a leading cause of disability with considerable mortality. Developing alternative drugs and targets for RA treatment is an urgent issue. Sesamol is a phenolic compound isolated from natural food sesame ( Sesamum indicum L.) with various biological activities. Purpose The current research intended to illuminate the bioactivity and mechanisms of sesamol in RA fibroblast-like synoviocytes (FLS), and aimed to estimate the potential clinical application value of sesamol in RA treatment. Methods CCK-8, EdU, and flow cytometry assays, as well as transwell tests were applied to observe the effects of sesamol on the abnormal functions of RA-FLS. Moreover, synovial organoids and a collagen-induced arthritis (CIA) mouse model were constructed to further explore the therapeutic capacity of sesamol on RA. Furthermore, RNA sequencing combined with quantitative real-time PCR assay, Western blot as well as co-immunoprecipitation were employed to clarify the mechanism of sesamol in regulating RA progression. Results Sesamol suppressed the proliferation through inhibiting DNA replication, triggering cell cycle arrest and apoptosis of RA-FLS. Besides, sesamol impaired RA-FLS migration and invasion. Interestingly, sesamol inhibited the growth of constructed synovial organoids and alleviated RA symptoms in CIA mice. Moreover, RNA sequencing further implicated p53 signaling as a downstream pathway of sesamol. Furthermore, sesamol was shown to decrease p53 ubiquitination and degradation, thereby activating p53 signaling. Finally, bioinformatics analyses also highlighted the importance of sesamol-regulated networks in the progression of RA. Conclusions Our investigation demonstrated that sesamol served as a novel p53 stabilizer to attenuate the abnormal functions of RA-FLS via facilitating the activation of p53 signaling. Moreover, our study highlighted that sesamol might be an effective lead compound or candidate drug and p53 could be a promising target for the therapy of RA.

Sesamol Attenuates Amyloid Peptide Accumulation and Cognitive Deficits in APP/PS1 Mice: The Mediating Role of the Gut–Brain Axis

Qing Liu,Tianzhi Xie,Yujia Xi,Ling Li,Fengfeng Mo,Xuebo Liu,Zhigang Liu,Jin-Ming Gao,Tian Yuan

Alzheimer’s disease (AD), a neurodegenerative disease, is the leading cause of dementia. Sesamol is a lignan extracted from sesame oil and has been found to exert neuroprotective effects. The present study aimed to investigate the neuroprotective effects of sesamol on APPswe/PS1dE9 transgenic AD mice. The AD mice were fed with a diet supplemented with sesamol (0.075 w/w %). Sesamol treatment improved spatial memory and learning ability in AD mice, improved neuronal damage, and decreased Aβ accumulation. Sesamol protected the synaptic ultrastructure and inhibited neuroinflammatory responses in the brain of AD mice. Sesamol also significantly inhibited the overactivated microglia and reduced the overexpression of TNF-α and IL-1β in the brain of AD mice. Notably, sesamol reshaped gut microbiota by significantly decreasing the relative abundance of Helicobacter hepaticus, Clostridium, and Bacillaceae, enhancing the relative abundance of Rikenellaceae and Bifidobacterium in AD mice. It has been found that sesamol protected the gut barrier integrity and prevented the LPS leakage into the serum. Importantly, sesamol remarkably enhanced the content of SCFAs, including acetate, propionate, isobutyrate, butyrate, and valerate, in AD mice. Correlation analysis indicated that there was a strong correlation between the levels of SCFAs and cognitive functions. These results demonstrated that sesamol attenuated AD-related cognitive dysfunction and neuroinflammatory responses, which could be partly explained by its role in mediating the gut microbe–SCFA–brain axis. Thus, sesamol is a promising nutritional intervention strategy to prevent AD via the microbiota–gut–brain axis.
