Physicochemical properties, in vitro digestibility, and pH-dependent release behavior of starch–steviol glycoside composite hydrogels

Fan Wang,Rongrong Ma,Jingling Zhu,Jinling Zhan,Jun Li,Yaoqi Tian

Steviol glycosides possess Bola-form amphiphilic structure, which can solubilize hydrophobic phytochemicals and exert physical modification to the hydrophilic matrix. However, the effect of steviol glycosides on the starch hydrogel is still unclear. Herein, the physicochemical properties, in vitro digestibility, and release behavior of starch hydrogel in the presence of steviol glycosides were investigated. The results showed that the addition of steviol glycosides promoted the gelatinization and gelation of starch, and endowed the starch hydrogel with softer texture, larger volume, and higher water holding capacity. The hydrophobic curcumin was well integrated into hydrogel by steviol glycosides, providing the gel with improved colour brilliance. The introduction of steviol glycosides hardly affected the digestibility of starch gel, but it promoted the release rate of curcumin. Notably, this release behavior was pH dependent, which tended to target the alkaline intestine. This work provided some theoretical supports for the development of sugar-free starchy foods.

Theoretical design and preparation research of molecularly imprinted polymers for steviol glycosides

Zhu Yajie,Ding Yan,Tian Dongxu,Li Yan,Zhuang Linwu,Wang Yinpeng,Xiao Wei,Zhu Jingbo

In this paper, a novel molecularly imprinted polymer (MIP) for specific adsorption of steviol glycosides was designed, and the imprinting mechanism of self-assembly system between template and monomers was clearly explored. Firstly, steviol (STE) was chosen as dummy template, and the density functional theory (DFT) at B3LYP/6–31 + G (d, p) level was used to select monomers, imprinting molar ratios, solvents, and cross-linking agents. The selectivity to five steviol glycosides was also calculated. Importantly, reduced density gradient (RDG) theory combined with atom in molecules (AIM) and infrared spectrum (IR) was applied to investigate the bonding situation and the nature of noncovalent interaction in self-assembly system. The theoretical designed results showed that the template which interacts with acrylic acid (AA) has the minimum binding energy, and the complex with the molar ratio of 1 : 4 has the most stable structure. Toluene (TL) and ethylene glycol dimethacrylate (EGDMA) were chosen as the optimal solvent and cross-linking agent, respectively. Five hydrogen bonds formed in the self-assembly system are the key forces at the adsorption sites of MIPs through the RDG and AIM analyses. The MIPs were synthesized by theoretical predictions, and the results showed that the maximum adsorption capacity towards dulcoside A is 26.17 mg/g. This work provided a theoretical direction and experimental validation for deeper researches of the MIPs for steviol glycosides. In addition, the method of RDG theory coupled with AIM and IR also could be used to analyze other imprinting formation mechanisms systematically.

Pore engineering of phenylboronic covalent organic framework membranes for Steviol glycosides separation

Shiyao Qiu,Yuqing Zhang,Haoyun Hu,Shanshan Lu,Yonggang Zhao,Sitong Lin,Xijie Lin,Hao Guo,Liping Jiang,Jianhua Qiu

The adsorption-diffusion sites in the channels of the COF membrane markedly enhance SGs diffusion. • Its phenylboronic acid groups selectively form dynamic boronate ester bonds with the vicinal diol moieties of SGs. • This specific interaction enables a hopping-diffusion mechanism of SGs in the TAPB-BuPa COF membrane. • The TAPB-BuPa COF membrane exhibited the SGs transmission of 98.1% and the permeance of 175 L m −2  h −1  bar −1 . Membrane separation offers high efficiency, environmental compatibility, and precise molecular-level discrimination, making it highly promising for the purification of steviol glycosides (SGs). However, the directional construction of high-permeance membranes that allow efficient SGs transmission remains challenging. To address these issues, we constructed adsorption-diffusion functional sites within the well-defined nanochannels of the covalent organic framework (COF) membrane. The hydrophobic TAPB-TFB COF membrane from 1,3,5-tris(4-aminophenyl)benzene (TAPB) and 1,3,5-triformylbenzene (TFB), and the hydrophilic boronic-acid-functionalized TAPB-BuPa COF membrane from TAPB and 3,5-diformylphenylboronic acid (BuPa) were fabricated via liquid-liquid interfacial polymerization. The TAPB-TFB COF membrane exhibited SGs transmission of 45.4% and the permeance of 48 L m −2  h −1  bar −1 . The TAPB-BuPa COF membrane created a hydrophilic affinity microenvironment within its ordered nanochannels. Its pendant phenylboronic acid groups selectively form dynamic, reversible boronate ester bonds with the vicinal diol moieties of SGs. This specific interaction enabled a hopping diffusion mechanism. As a result, the permeance increased 3.6-fold to 175 L m −2  h −1  bar −1 , and achieved a high SGs transmission of 98.1%. Elucidating this microenvironment-controlled switch from adsorption to diffusion provides a design strategy for high-permeance membranes tailored for natural product concentration. In the TAPB-BuPa COF membrane, PBA specifically forms bonds with the adjacent dihydroxy groups on the SGs molecules, causing the bound SGs to undergo hopping-diffusion along the phenylboronic acid sites lining the pore walls. This established pore microenvironment determines the adsorption-to-diffusion switching paradigm, opening a new pathway for engineering COF-based membranes that use dynamic covalent bonds as the recognition mechanism, and providing a theoretical basis and material design strategy for efficient separation of natural products. Tunable and Recyclable Natural Deep Eutectic Solvents for Efficient Extraction of Steviol Glycosides from Stevia rebaudiana

Jinling Bai,Yike Han,Mengting Fu,Mengting Cui,Zhiyu Liu,Zhixin Ouyang,Liyun Kong,Xinyan Dong

Steviol glycosides (SGs), particularly rebaudioside A (RA) and stevioside (STV), are the primary natural sweeteners in Stevia rebaudiana and are increasingly utilized as zero-calorie sucrose substitutes. Although water is environmentally friendly, its poor selectivity limits efficient extraction of SGs, while conventional organic solvents raise environmental concerns. Auxiliary techniques such as ultrasound-assisted extraction often require high energy input or specialized equipment. Natural deep eutectic solvents (NADES) offer a sustainable, tunable alternative for SGs extraction. In this study, the Conductor-like Screen Model for Real Solvent (COSMO-RS) was applied to screen 403 DES combinations, revealing that NADES with strong hydrogen-bonding abilities significantly enhance SGs extraction. Experimental validation identified a betaine- propionic acid NADES (1:2 molar ratio) with 45 wt % water as optimal, achieving yields of 98.15 ± 2.62 mg/g RA, 59.83 ± 1.60 mg/g STV, and 7.16 ± 0.45 mg/g rebaudioside C (RC). To address challenges in SGs recovery from NADES, static adsorption/desorption experiments were conducted using AB-8 resin. Hydrophilic NADES were effectively recovered with deionized water washing and maintained over 85% extraction efficiency after two reuse cycles. Target SGs were efficiently desorbed with 70% ethanol, while impurities were removed with 30% ethanol. The resulting yields of RA, STV, and RC were 90.56%, 86.68%, and 65.89%, with corresponding purities of 53.23%, 29.06%, and 4.42%, respectively. These results demonstrate that the combination of the betaine-propionic acid NADES and AB-8 resin offers an effective and sustainable strategy for the extraction and purification of SGs from S. rebaudiana.

Preparation of P(EGDMA-co-VPBA) Adsorbent and Its Application in the Separation of Steviol Glycosides

Qiaoge Fu,Nana Chen,Guanyu Wang,Ruili Guo

The poly (ethylene glycol dimethacrylate- co -vinylphenylboronic acid) P(EGDMA- co -VPBA) adsorbents are prepared via reversible addition-fragmentation chain transfer (RAFT) precipitation polymerization with 4-vinylphenylboronic acid (VPBA) as the functional monomer, and used for selective adsorption of steviol glycosides (SGs). The boron content and pore structure of the P(EGDMA- co -VPBA) adsorbent are adjusted by changing the polymerization conditions. The adsorption results show that the pore structure of the P(EGDMA- co -VPBA) affects the adsorption capacity to SGs, the presence of boric acid functional groups shows favorable adsorption selectivity for stevioside (STV), and the adsorption selectivity is proportional to the content of boric acid in P(EGDMA- co -VPBA) adsorbents. The adsorption performance of P(EGDMA- co -VPBA) toward SGs is the best when the ratio of monomer to cross-linker is 1:50 and the solvent is acetonitrile. In the SG solution (6.0 mg mL −1 , C STV / C RA = 1:1), the adsorption capacities and adsorption selectivity ( α STV/RA ) of P(EGDMA- co -VPBA) to SGs are 113.20 mg g –1 and 4.99, respectively. The adsorption kinetics and isotherm data obey pseudo-second-order kinetic model and Langmuir isotherm model, respectively. The boric acid copolymers with VPBA as the monomer prepared by the one-step method can become a promising material for efficient separation and purification of SGs.

A well-defined hierarchically porous metal–organic framework and its application in separation and purification of steviol glycosides

Nana Chen,Mingli Huang,Qiaoge Fu,Chaoyi Shi,Ruili Guo

The separation and removal of stevioside from natural product steviol glycosides to obtain high-purity rebaudioside A is of great significance for the application of steviol glycosides in food, medicine, and other fields. Here, in order to explore the adsorbent pore structure suitable for the separation of stevioside and rebaudioside A, a hierarchically porous amino-functionalized metal–organic framework (HP-NH 2 -MIL-53) with an appropriate and narrow pore size distribution was prepared using a modulator-induced defect-formation strategy. The results showed that the hierarchically porous structure with micropores and mesopores increased the specific surface area and exposed amino groups compared with original metal organic framework (NH 2 -MIL-53), and the maximum adsorption capacity of HP-NH 2 -MIL-53 for stevioside and rebaudioside A was 233.89 mg/g. The narrow pore size distribution close to 3.80 nm promoted the screening effect, resulting in a maximum adsorption selectivity of 4.13. This work proves that when the pore size of the adsorbent is between 1.41 and 3.80 nm, it has a certain pore size screening effect on stevioside and rebaudioside A, and the hierarchically porous metal-organic frameworks provide a pre-design idea of adsorbent structure for the separation of natural products with molecular weight of 800–1000 Da.

Influence of substrates on the in vitro kinetics of steviol glucuronidation and interaction between steviol glycosides metabolites and UGT2B7

Jun-Ming Chen,Yong-Mei Xia,Yan-Dong Zhang,Tong-Tong Zhang,Qing-Rui Peng,Yun Fang

Steviol glycosides, a natural sweetener, may perform bioactivities via steviol, their main metabolite in human digestion. The metabolising kinetics, i.e. glucuronidation kinetics and interaction between steviol glycosides or their metabolites and metabolising enzyme, are important for understanding the bioactivity and cytotoxicity. The present study investigated kinetics of steviol glucuronidation in human liver microsome and a recombinant human UDP-glucuronosyltransferases isomer, UGT2B7, along with molecular docking to analyse interaction between UGT2B7 and steviol or glucose. The active pocket of UGT2B7 is consisted of Arg352, Leu347, Lys343, Phe339, Tyr354, Lys355 and Leu353. The influence of stevioside, rebaudioside A, glucose and some chemotherapy reagents on the glucuronidation was also studied. The predicted hepatic clearence suggested that steviol could be classified as high-clearence drug. The steviol glycosides did not affect the glucuronidation of steviol notably.
