Efficient production of biosurfactant surfactin by a newly isolated Bacillus subtilis (sp.) 50499 strain from oil-contaminated soil

Yulu Du,Yue Wang,Tianyou Cui,Lianpeng Ge,Fan Yu,Mengqian Zhao,Qihang Xu,Mingjie Jin,Bo Chen,Xuwei Long

Biosurfactant surfactin has a high market value and broad application prospects in the food industry and various industrial fields. However, its production efficiency limits its suitability for real applications. In this study, a new Bacillus subtilis (sp.) 50499 strain was isolated from crude oil-contaminated soil and used to efficiently produce surfactin. The initial surfactin titer was 2.25 g/L after three days of fermentation. Adding 2 g/L of L -Leu significantly improved the titer to over 5.5 g/L after optimizing the culture medium at 37 °C. However, adding D -Leu had the opposite effect on surfactin production , and the surfactin titer dropped to below 0.2 g/L. Furthermore, the product of this strain was analyzed to confirm the identity of surfactin. The surfactin obtained in the present study reduced the surface tension of water to 28.6 mN/m at a critical micelle concentration of 20 mg/L. The emulsification index against n -hexane, corn , and crude oil was as high as over 90%. Hence, the present study provides a newly over-producing strain for surfactin production.

Surfactin-methylene blue complex under LED illumination for antibacterial photodynamic therapy: Enhanced methylene blue transcellular accumulation assisted by surfactin

Juan Zhao,Lixian Xu,Hao Zhang,Yuhong Zhuo,Yanan Weng,Shuang Li,Dinghua Yu

Recently, increased attention has been focused on antibacterial photodynamic therapy (APDT) to treat multidrug-resistant bacterial infection due to the antibiotic abuse. Methylene blue has been used as a kind of efficient and cheap commercial photosensitizer in APDT. However, due to high hydrophilicity, methylene blue is not able to be transcellular intaken and accumulated efficiently. To promote accumulation and APDT efficiency of methylene blue, lipopeptide surfactin-methylene blue complex has been prepared through electrostatic interaction. The complex under LED irradiation was found to effectively reduce 5.0 Log 10 CFU and 7.6 Log 10 CFU for P. aeruginosa and S. aureus , respectively. The bacterial reduction efficiency is slightly higher than free methylene blue. The photosensitizers accumulation and APDT targeting protein have been characterized by fluorescence spectroscopy, fluorescence microscopy and protein electrophoresis techniques. These results demonstrated that more surfactin-methylene blue complex could be accumulated more into the cell, and inactivate bacteria through destroying intracellular protein under LED illumination. In comparison, free methylene blue under light could inactivate bacteria through destroying membrane protein and lipid structures. These results would provide valuable insight for developing advanced clinical medicine and designing photo-drug for photodynamic therapy.

Structural Diversity of the Microbial Surfactin Derivatives from Selective Esterification Approach

Chuanshi Shao,Lin Liu,Hongze Gang,Shizhong Yang,Bozhong Mu

Surfactin originated from genusBacillusis composed of a heptapeptide moiety bonded to the carboxyl and hydroxyl groups of a β-hydroxy fatty acid and it can be chemically modified to prepare the derivatives with different structures, owing to the existence of two free carboxyl groups in its peptide loop. This article presents the chemical modification of surfactin esterified with three different alcohols, and nine novel surfactin derivatives have been separated from products by the high performance liquid chromatography (HPLC). The novel derivatives, identified with Fourier transform infrared spectroscopy (FT-IR) and electrospray ionization mass spectrometry (ESI-MS), are the mono-hexyl-surfactin C14 ester, mono-hexyl-surfactin C15 ester, mono-2-methoxy-ethyl-surfactin C14 ester,di-hexyl-surfactin C14 ester,di-hexyl-surfactin ester C15,di-2-methoxy-ethyl-surfactin ester C14,di-2-methoxy-ethyl-surfactin ester C15,di-6-hydoxyl-hexyl-surfactin C14 ester and,di-6-hydoxyl-hexyl-surfactin C15 ester. The reaction conditions for esterification were optimized and the dependence of yields on different alcohols and catalysts were discussed. This study shows that esterification is one of the most efficient ways of chemical modification for surfactin and it can be used to prepare more derivatives to meet the needs of study in biological and interfacial activities. Keywords:Keywords: surfactin;esterification;derivatives;lipopeptide;carboxyl group

Low-Toxic and Nonirritant Biosurfactant Surfactin and its Performances in Detergent Formulations

Dan Fei,Guang-Wei Zhou,Zhou-Qiang Yu,Hong-Ze Gang,Jin-Feng Liu,Shi-Zhong Yang,Ru-Qiang Ye,Bo-Zhong Mu

Developing eco-friendly, nonirritant, low-toxic, and high-efficient surface active ingredients for detergents is an ongoing challenge in the detergent field. Surfactin is one of the representative lipopeptides produced by microorganisms. In this article, we report the surfactin isolated from cell-free broth of Bacillus subtilis HSO121 and purified by reversed-phase high-performance liquid chromatography for detergent formulations. The biodegradability, acute dermal irritation, acute oral toxicity (LD 50 and LC 50 ), surface activity, washing efficiency, and compatibility with hard water of the purified biosurfactant surfactin have been studied to explore the feasibility for applications of the surfactin in detergents. Acute oral toxicity tests (LD 50 > 5000 mg kg −1 , LC 50 > 1000 mg kg −1 ) and skin irritation tests (PII = 0) indicate that the surfactin is a low-toxic and nonirritant ingredient for detergent formulation. Moreover, the surfactin shows excellent surface and interfacial properties of emulsification and wettability, high compatibility, and stability in a wide range of temperatures, pH, and hard water and acceptable properties in biodegradability and foaming ability, which suggests that the biosurfactant surfactin is a promising ingredient for detergent formations in our daily life and for industrial applications.

Surfactin Variants from Bacillus subtilis natto CSUF5 and Their Antifungal Properities Against Aspergillus niger

Wang Qing-Yun,Lin Qin-Lu,Peng Kuan,Cao Jian-Zhong,Yang Chun,Xu Dong

An antifungal Bacillus strain isolated from the traditional fermented food natto was identified and named as B. subtilis natto CSUF5. Antifungal metabolites of the strain were extracted from the medium of the inhibition zone on the dual culture plate. 2 V7-surfactin isoforms (C14–V7, C13–Emet 1-V7) and 3 I/L7-surfactin isoforms (C13–I/L7, C13,14–Emet 1–I/L7 were identified from the RP-HPLC isolations by MS n (n = 1,2,3) analysis. Both V7-surfactins and I/L7 -surfactins showed slight antifungal activity against A. niger, and their MIC50 decreases in the order V7 > I/L 7. EDTA was found to be an effective strengthening agent which could cooperate with the surfactin variants and enhance their antifungal activity greatly. Microscopic observations revealed that the surfactin variants could delay the fungal spore germination and inhibit the hyphae growth, mainly displayed in hyphal shriveling and distortion. Surfactin variants and EDTA together could lead the hyphae to severe helical warping and fracture. These results will provide promising references for the development of new antifungal products.

Optimizing surfactin yield in Bacillus velezensis BN to enhance biocontrol efficacy and rhizosphere colonization

Tongshu Liu,Yanli Zheng,Litao Wang,Xu Wang,Haiyan Wang,Yongqiang Tian

Introduction: Surfactins, a class of lipopeptide biosurfactants secreted by plant growth-promoting rhizobacteria (PGPR), have garnered significant attention due to their dual functionality in promoting plant growth and controlling plant diseases. Their potential as biopesticides is underscored by their unique physicochemical properties and biological activities. However, the practical application of surfactin is currently limited by its low yield in natural strains. Methods: This study aimed to optimize the culture conditions for Bacillus velezensis BN, a strain with exceptional biocontrol properties, to enhance its surfactin yield. Critical factors, including nitrogen sources and amino acid supplementation, were systematically investigated to determine their impact on surfactin production. Results: The study revealed that nitrogen sources and amino acid supplementation were pivotal factors influencing surfactin yield. Compared to the baseline, these factors resulted in a remarkable 5.94-fold increase in surfactin production. Furthermore, a positive correlation was established between surfactin yield and biocontrol efficacy. Enhanced surfactin yield was associated with improved antifungal activity, biofilm formation, and rhizosphere colonization capacity of B. velezensis BN on potato plantlets.Discussion: These findings provide novel insights into the practical application of surfactin and establish a scientific foundation for the development of innovative and eco-friendly antifungal agents suitable for agricultural use. The results demonstrate that optimizing culture conditions can significantly enhance surfactin yield and biocontrol efficacy, thereby highlighting the potential for sustainable agricultural practices.

Lignin/Surfactin Coacervate as an Eco-Friendly Pesticide Carrier and Antifungal Agent against Phytopathogen

Jie Wang,Zhichen Xiong,Yaxun Fan,Hongliang Wang,Changcheng An,Bo Wang,Ming Yang,Xue Li,Yan Wang,Yilin Wang

Excessive usage of biologically toxic fungicides and their matrix materials poses a serious threat to public health. Leveraging fungicide carriers with inherent pathogen inhibition properties is highly promising for enhancing fungicide efficacy and reducing required dosage. Herein, a series of coacervates have been crafted with lignin and surfactin, both of which are naturally derived and demonstrate substantial antifungal properties. This hierarchically assembled carrier not only effectively loads fungicides with a maximum encapsulation efficiency of 95% but also stably deposits on hydrophobic leaves for high-speed impacting droplets. Intriguingly, these coacervates exhibit broad spectrum fungicidal activity against eight ubiquitous phytopathogens and even act as a standalone biofungicide to replace fungicides. This performance can significantly reduce the fungicide usage and be further strengthened by an encapsulated fungicide. The inhibition rate reaches 87.0% when 0.30 mM pyraclostrobin (Pyr) is encapsulated within this coacervate, comparable to the effectiveness of 0.80 mM Pyr alone. Additionally, the preventive effects against tomato gray mold reached 53%, significantly surpassing those of commercial adjuvants. Thus, it demonstrates that utilizing biosurfactants and biomass with intrinsic antifungal activity to fabricate fully biobased coacervates can synergistically combine the functions of a fungicide carrier and antifungal agent against phytopathogens and guarantee environmental friendliness. This pioneering approach provides deeper insights into synergistically enhancing the effectiveness of agrochemicals from multiple aspects, including fungicide encapsulation, cooperative antifungal action, and droplet deposition.

Ameliorated effects of a lipopeptide surfactin on insulin resistance in vitro and in vivo

Xiaoyu Chen,Hongyuan Zhao,Fanqiang Meng,Libang Zhou,Xinyi Pang,Zhaoxin Lu,Yingjian Lu

Surfactin, produced by Bacillus amyloliquefaciens fmb50, was used to treat insulin-resistant (IR) hepatocyte. It was found that surfactin increased glucose consumption in insulin-resistant HepG2 (IR-HepG2) cells and ameliorated IR by increasing glucose transporter 4 (GLUT4) protein expression and AMP-activated protein kinase (AMPK) mRNA expression, promoting GLUT4 translocation and activating phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt) in IR-HepG2 cells. Meanwhile, surfactin downregulated protein expression of phosphoenolpyruvate carboxy kinase (PEPCK) and glucose-6-phosphatase (G6Pase), further inhibiting hepatic gluconeogenesis. In addition, surfactin played important roles in eliminating reactive oxygen species (ROS), improving mitochondrial dysfunction, and inhibiting proinflammatory mediators. We observed that surfactin promoted glucose consumption, meanwhile increased translocation and protein expression of GLUT4 in Caco-2 cells. These results confirmed the conclusion in hepatic cells. Furthermore, surfactin supplement decreased body weight, food intake, and fasting blood glucose of type 2 diabetes mellitus (T2DM) mice induced by streptozotocin (STZ)/high-fat diet (HFD). Our data indicated that surfactin ameliorated insulin resistance and lowered blood glucose in intro and in vivo.

Impact of surfactin on the physicochemical properties of dough and quality of corresponding steamed bread

Huifang Wang,Xinyue Wei,Dengdeng Li,Jiai Yan,Yina Wu,Zhongkai Zhou

BACKGROUND The extensive use of additives in ultra-processed foods presents considerable health concerns. In light of the growing consumer demand for clean labels, a prominent trend is the development of multifunctional food additives that are both natural and beneficial to health. Surfactin, a compound produced by Bacillus subtilis , features both hydrophilic and hydrophobic groups and is noted for its safety, emulsifying and antimicrobial properties. This compound holds significant potential as a multifunctional additive in flour-based products. The present study aimed to evaluate the effects of surfactin on the physicochemical properties of dough and the quality of steamed bread, as well as to investigate the underlying mechanisms. RESULTS The results showed that the addition of surfactin significantly improved its rheological properties, increased elasticity and viscosity, improved extension resistance and increased disulfide bonding content in dough ( P < 0.05), subsequently stabilizing the gluten network structure. With a 0.3% surfactin addition, the digestibility of steamed bread significantly reduced. After storing for 7 days, surfactin inhibited water migration, reduced the transfer from bound water to free water, delayed starch recrystallization, improved resistance to starch retrogradation and markedly extended the shelf life in steamed bread. CONCLUSION The addition of surfactin improved the quality of steamed bread through stabilizing the gluten network structure and improving storage properties, presenting it as a promising natural, multifunctional food additive for flour product innovation. © 2024 Society of Chemical Industry.

Surfactin alleviates insulin resistance by modulating the AMPK/PI3K/Akt signalling pathway and inactivating inflammatory pathways

Xiaoyu Chen,Hongyuan Zhao,Fanqiang Meng,Juan Shen,Chao Tang,Zhaoxin Lu,Yingjian Lu

Type 2 diabetes mellitus (T2DM) is accompanied by insulin resistance in liver and peripheral tissue. Bacillus amyloliquefaciens fmb50 produces lipopeptide surfactin, which has a wide range of biological activities. However, the effects of surfactin on insulin resistance in any mouse model have not been reported. Our previous research demonstrated that dietary supplementation with surfactin can alleviate high fat diet (HFD)/ streptozotocin (STZ)-induced T2DM in mice by regulating intestinal microbiota. This study aims to explore the detailed mechanism of the effect of surfactin on liver and peripheral tissue. The results indicated that surfactin inhibited hepatic gluconeogenesis and promoted hepatic glycogen synthesis by activating the adenosine monophosphate-activated protein kinase (AMPK) and phosphatidylinositol-3 kinase (PI3K)/protein kinase B (PKB/Akt) signalling pathways. Similarly, surfactin also significantly increased muscle glycogen synthesis by activating the PI3K/Akt signalling pathway. The glucose transporter 4 (GLUT4) levels of skeletal muscle and adipose tissue, which are insulin-mediated vital organs, was noticeably increased after surfactin treatment. In addition, our results indicate that decreased hepatic fat accumulation and suppressed inflammation and oxidative stress in liver, skeletal muscle and adipose tissue also play critical roles in ameliorating insulin resistance. These findings demonstrate that surfactin not only ameliorates liver dysfunction but also enhances insulin sensitivity in HFD/STZ-induced T2DM mice. Finally, using fasting blood glucose, oral glucose tolerance test (OGTT) and insulin tolerance test (ITT), we show that early surfactin intervention can mitigate T2DM in mice. The study results warrant future functional food studies which will provide insight into diet-host interactions.

Cinnamaldehyde-surfactin nano-capsulation mitigates high-fat diet/ streptozotocin-induced diabetes and modulate gut microbial dysbiosis

Xiaoyu Chen,Hongyuan Zhao,Fanqiang Meng,Chao Tang,Yingjian Lu,Zhaoxin Lu

Type 2 diabetes mellitus (T2DM) is associated with liver dysfunction and intestinal dysbiosis. Cinnamaldehyde have been reported to ameliorate T2DM, on the other hand, its mechanism is unclear. In the present study, surfactin-loaded cinnamaldehyde microemulsion (Cin-NEs) supplementation significantly promoted glucose uptake in insulin resistant HepG2 cells. According to further research, Cin-NEs treatment dramatically upregulated phosphorylated-phosphoinositide-3 kinase (p-PI3K) and phosphorylated-protein kinase B (PKB/Akt), downregulated phosphoenolpyruvate carboxykinase (PEPCK) and glucose-6-phosphatase (G6pase), these further enhance hepatic glycogen synthesis and suppress gluconeogenesis to promote glucose uptake. Cin-NEs supplementation lowered fasting blood glucose (FBG) by 34.29-41.43% and insulin resistance (IR) by 36.0-40.0%, increased glucose tolerance by 49.33-53.33% compared with those in the T2DM group, reduced liver’s lipid accumulation and inflammation in mice with T2DM induced by a high-fat diet (HFD) and streptozotocin (STZ). A Cin-NEs administration reduced Prevotella and Shigella , while increased Akkermansia genus and colonic tight junction protein Claudin-1, ZO-1 and Occludin. Furthermore, Cin-NEs also improved hepatic glucose metabolism by activating the adenosine monophosphate-activated protein kinase (AMPK) and PI3K/Akt signaling pathway, downregulating PEPCK and G6Pase protein, and upregulating GLUT4 and GS protein levels. All of these findings revealed that Cin-NEs not only ameliorated HFD/STZ-induced glucose metabolism disorder but also reversed gut dysbiosis and preserved intestinal barrier integrity in T2DM mice. Cinnamaldehyde exhibits a similar effect to Cin-NEs, and the effect of Cin-NEs with 10 mg/kg·BW is equal to that of cinnamaldehyde with 20 mg/kg·BW. All data reveals that cinnamaldehyde-surfactin nano-capsulation possesses high stability in vivo and bioavailability to mitigates T2DM in mice.

Surfactin alleviated hyperglycaemia in mice with type 2 diabetes induced by a high-fat diet and streptozotocin

Xiaoyu Chen,Hongyuan Zhao,Fanqiang Meng,Libang Zhou,Zhaoxin Lu,Yingjian Lu

Type 2 diabetes mellitus (T2DM) is associated with liver dysfunction and intestinal dysbiosis. Bioactive peptides (BAPs) have been reported to ameliorate T2DM by preventing oxidative damage to the liver. Bacillus amyloliquefaciens fmb50 produces the lipopeptide surfactin with a wide range of biological activities. The effects of surfactin on T2DM, on the other hand, have not been studied. In the present study, 80 mg/kg body weight surfactin supplementation lowered fasting blood glucose (FBG) levels by 21.05 % and insulin resistance (IR) by 18.18 % compared with those in the T2DM group, reduced inflammation, and increased antioxidant activity in mice with T2DM induced by a high-fat diet (HFD) and streptozotocin (STZ). According to further research, surfactin administration reduced Firmicutes-to-Bacteroidetes ratios while increasing Bifidobacterium abundance by 20 times and the level of the tight junction protein Occludin by 18.38 % and ZO-1 by 66.60 %. Furthermore, surfactin also improved hepatic glucose metabolism by activating the adenosine monophosphate-activated protein kinase (AMPK) signalling pathway, increasing glycogen synthesis and glucose transporter 2 (GLUT2) protein expression while reducing glucose-6-phosphatase (G6Pase) protein expression. In addition, the increased Bifidobacterium abundance indirectly reduced the liver burden of the metabolic products indole, cresol and amine produced by saprophytic bacteria. All of these findings revealed that surfactin not only ameliorated HFD/STZ-induced gut dysbiosis and preserved intestinal barrier integrity but also enhanced hepatic glucose metabolism and detoxification function in T2DM mice. The gut microbiota appeared to be important in controlling glucose metabolism, IR, fat accumulation, inflammation and antioxidation, according to Spearman’s correlation coefficients. All data indicated that surfactin alleviated hyperglycaemia in mice with T2DM induced by HFD/STZ.
