Tebuconazole and Azoxystrobin Residue Behaviors and Distribution in Field and Cooked Peanut

Fan Hou,Peipei Teng,Fengmao Liu,Wenzhuo Wang

Residue behaviors of tebuconazole and azoxystrobin in field condition and the variation of their residue levels during the boiling process were evaluated. The terminal residues of peanut kernels were determined by using a modified QuEChERS method (quick, easy, cheap, effective, rugged, and safe) by means of the optimization of the novel purification procedure with multiwalled carbon nanotubes (MWCNTs) and Fe3O4-magnetic nanoparticle (Fe3O4-MNP) in the presence of an external magnetic field, and the terminal residues were all at trace level at harvest time. The residues in shells were detected as well to investigate the distribution in peanuts. Tebuconazole and azoxystrobin residue levels varied before/after boiling in kernels and shells to different degrees due to various factors, such as the modes of action and physicochemical properties of pesticides. The residues have been transferred from peanut into the infusion during boiling with the higher percentage of azoxystrobin as its lower logKow. The processing factors (PFs) for tebuconazole and azoxystrobin after processing were <1, indicating that home cooking in this study could reduce the residue levels in peanut. Risk assessment showed there was no health risk for consumers.

Rapid detection of tebuconazole based on hydrogel SERS chips

Mingming Chen,Xiaojing Lai,Bihang Su,Xiancai Jiang,Jinhua Xu,Fengfu Fu,Zhenyu Lin,Yongqing Dong

Tebuconazole is one of the most commonly used fungicides in agricultural production, that has the merits of highly effectiveness, broad spectrum and systemic function. Excessive tebuconazole may pose a great threat to human and animal health. Traditional detection techniques for tebuconazole usually have limitations such as expensive equipment, poor antibody stability, and time-consuming procedures. Herein, a sensitive sensor is developed for the rapid detection of tebuconazole based on hydrogel surface-enhanced Raman scattering (SERS) chips. Aggregated Ag nanoparticles (a-AgNPs) with tunable localized surface plasmon resonance (LSPR) wavelength are in-situ synthesized in polyvinyl alcohol (PVA) solution for preparing hydrogel SERS chips. Three hydrogel SERS chips are obtained to match the three commonly used laser wavelengths. On the basis, a match laser wavelength is selected according to the energy levels of tebuconazole and the Fermi level of a-AgNPs to gain a strong chemical enhancement. At the same time, the chip with a corresponding LSPR wavelength to the laser is applied to obtain a strong electromagnetic enhancement. Thus, highly sensitive SERS signal of tebuconazole is obtained. Furthermore, the obtained hydrogel SERS chips have good repeatability, outstanding reproducibility and strong anti-interference ability, and show outstanding reliability in practical applications. As a result, the SERS chips offer a reliable and convenient platform for the quick detection of tebuconazole in foods. The detection limit is as low as 1 ppb, and the recoveries is distributed in the range of 94.66–106.70 %. This work would promote greatly the application of SERS in small molecule detection.

An innovative hapten and monoclonal antibody-based immunoassay for determining tebuconazole residues in aqueous samples

Xiujin Chen,Zhaozhou Li,Fengxia Sun,Xiaotian Cao,Yao Wang,Li Cao,Hongli Gao,Dong Gao,Yawen Wang

Tebuconazole is widely applied for controlling pathogenic fungi of crops and proved hazardous to human being. To better measure tebuconazole residues, an enzyme-linked immunosorbent assay for screening tebuconazole residues in aqueous samples was established using an innovative hapten (hapten1) designed via molecular model. The work concentration of calibration standard curves based on aqueous matrices ranged from 0.51 to 10.71 ng mL–1. The cross-reactivities of other analogs were less than 6%. The detection limits of the assay were in the range of 0.197–0.297 ng mL–1 for tebuconazole in water samples. The average recoveries of tebuconazole in the spiked water samples varied from 76.76% to 107.52% with the variation coefficient scope of 4.54–13.26%. The obtained immunoassay was verified by gas chromatography coupled to mass spectrometry with high correlation coefficients (r > 0.9868). Herein, the proposed immunoassay offers a cost-effective, convenient, reliable tool for the determination of tebuconazole residues in aqueous samples.

Preparation and characterization of tebuconazole metal-organic framework-based microcapsules with dual-microbicidal activity

Jingyue Tang,Guanglong Ding,Junfan Niu,Wenbing Zhang,Gang Tang,You Liang,Chen Fan,Hongqiang Dong,Jiale Yang,Jianqiang Li,Yongsong Cao

Based on excellent optical properties and microbicide activity, porphyrinic metal-organic frameworks (MOFs) are widely used as drug release carriers for their photodynamic therapy (PDT). In this work, porous porphyrinic MOFs (PCN-224) nanoparticles were synthesized for loading and controlling release of tebuconazole by using meso-tetra(4-carboxyphenyl)porphine (H 2 TCPP) as organic linker, metal ions clusters Zr (IV) as nodes, and subsequently assembled layer by layer of pectin and chitosan to obtain tebuconazole microcapsules (Tebuc@PCN@P@C). The results showed that the prepared MOFs had loading efficiency of tebuconazole about 30% (w/w), and the obtained stimuli-responsive microcapsules showed adjustable controlled-release characteristics with pectinase in acidity environment. Tebuc@PCN@P@C microcapsules displayed excellent double activities of PDT and microbicide against Xanthomonas campestris pv. campestris , Pseudomonas syringae pv. Tomato and Alternaria alternate . Safety evaluation on Chinese cabbage indicated they almost had no any damage to plants. Thus, stimuli-responsive dual-microbicidal MOFs microcapsules were demonstrated a great potential as an efficient delivery system in pesticide formulations.

Magnetic Fe3O4@Au@PDA core-shell nanoparticle-enhanced SPR detection of tebuconazole

Bing zhang,Yu-lian Li,Li Wang,Haoyuan Cai

Conventional surface plasmon resonance (SPR) sensor often fails to detect low-concentration small molecules due to insufficient refractive index changes. To address this challenge, we have developed an enhanced SPR sensor utilizing Fe 3 O 4 @Au@polydopamine (PDA) core–shell magnetic nanoparticles as signal amplification tags for the detection of tebuconazole (TBZ). These nanoparticles improve sensitivity through their high refractive indices and enable TBZ enrichment from complex matrices via their magnetic properties. The sensor surface is further modified with PDA, which acts as a reducing agent for Au nanoparticle deposition, enhancing monoclonal antibody (Ab) immobilization and boosting the SPR signal. Our optimized sensor reaches a limit of detection (LOD) at 0.61 ng/mL for TBZ, with a linear response from 1-200 ng/mL, and demonstrates high selectivity and reproducibility, ideal for pesticide monitoring in food samples.

Preparation and application of tebuconazole molecularly imprinted polymer for detection of pesticide residues in tobacco leaves

Jing Lian Peng,Yang Fa Rong,Gu Li Li,Li Guo Dong,Li Zeng Liang,Tang Xu Yu,Shi Jun Li

In this paper, computer simulation technology was used to design the pre-polymerization system of Tebuconazole Molecularly Imprinted Polymer (TBZ-MIPs). The simulation results illustrated that the best functional monomer was MAA, the polymerization solvent was acetonitrile, and the ratio of TBZ to MAA was 1:3. Thermodynamic analysis showed that the self-assembly process of TBZ-MAA was a non-spontaneous endothermic reaction, and the appropriate pre-polymerization temperature was 30 ℃. Based on the computer simulation results, the TBZ-MIPs nanospheres were prepared by precipitation polymerization with TBZ as template molecule, MAA as functional monomer, EGDMA as crosslinking agent, AIBN as initiator, and acetonitrile as solvent. The nanospheres had a uniform particle size, and the average diameter was 188.83 nm. Adsorption experiments indicated that the TBZ-MIPs had good specific adsorption capacity for TBZ and its structural analogs, the maximum dynamic adsorption capacity could reach to 1.8 mg/g in 60 min. Further, the TBZ-MIPs nanospheres were used as the filler of the solid phase extraction tube to pretreat the tobacco samples, and the method of TBZ-MISPE-UHPLC-MS/MS was established to detect trace tebuconazole, myclobutanil and triadimenol in tobacco leaves simultaneously. Their average recovery rates and RSD were 70.5% ~ 110.17%, and 0.78% ~ 12.71% (n = 3) respectively, which realized trace detection of triazole fungicides in tobacco leaves.

Highly Specific Monoclonal Antibody and Sensitive Quantum Dot Beads-Based Fluorescence Immunochromatographic Test Strip for Tebuconazole Assay in Agricultural Products

Yulong Wang,Junli Xu,Yulou Qiu,Pan Li,Beibei Liu,Lifei Yang,Bogdan Barnych,Bruce D. Hammock,Cunzheng Zhang

A monoclonal antibody (mAb) was raised against tebuconazole (TEB) using a hapten where the p-chloro substituent of the TEB molecule was replaced with a long-chain carboxylic acid. The resulting mAb showed high sensitivity and specificity against TEB characterized by ELISA with a half-maximal inhibitory concentration (IC50) of 0.19 ng mL–1 and with cross-reactivity (CR) values below 0.01% to several analogues of triazole fungicides. On the basis of the mAb produced, a quantum dot beads-based fluorescence immunochromatographic test strip assay (QBs–FITSA) was developed for rapid and sensitive detection of TEB in agricultural product samples. The QBs–FITSA exhibited a linear detection range from 0.02 to 1.25 ng mL–1 with a limit of detection (LOD) of 0.02 ng mL–1. Furthermore, using produced mAb, multiple high-throughput rapid immunoassay formats could be achieved as a convenient monitoring tool for evaluation of human and environmental exposure to TEB.

Metabolic pathway of tebuconazole by soil fungus Cunninghamella elegans ATCC36112

Meng Min,Zhai Zhaochi,Zhang Zhenxing,Kim Jeonghan,Zhu Yongzhe

Tebuconazole is the most widely used fungicide in agriculture. Due to its long half-life, tebuconazole residues can be found in the environment media such as in soil and water bodies. Here, the metabolic pathway of tebuconazole was studied in Cunninghamella elegans ( C. elegans ). Approximately 98% of tebuconazole was degraded within 7 days, accompanied by the accumulation of five metabolites. The structures of the metabolites were completely or tentatively identified by gas chromatography-mass spectrometry (GC–MS) and ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). To identify representative oxidative enzymes that may be involved in the metabolic process, treatment with piperonyl butoxide (PB) and methimazole (MZ) was performed. PB had a strong inhibitory effect on the metabolic reactions, while MZ had a weak inhibitory effect. The results suggest that cytochrome P450 (CYP) and flavin-dependent monooxygenase are involved in the metabolism of tebuconazole. Based on the results, we propose a metabolic pathway for the fungal metabolism of tebuconazole. Data are of interest to gain insight into the toxicological effects of tebuconazole and for tebuconazole bioremediation.

Tebuconazole mediates cognitive impairment via the microbe-gut-brain axis (MGBA) in mice

Tingting Ku,Yutong Liu,Yuanyuan Xie,Jindong Hu,Yanwen Hou,Xin Tan,Xia Ning,Guangke Li,Nan Sang

Tebuconazole, one of the most widely used triazole fungicides, is reported to potentially pose a risk of inducing neurological disorders in human beings. Considering the increasing exposure, whether it could influence cognitive function remains to be elucidated. Herein, we used a mouse model to evaluate the potential cognitive risks and possible mechanisms from the continuous edible application of tebuconazole at low concentrations. Our study revealed that tebuconazole deteriorated spatial learning and memory and downregulated the expression of glutamate receptor subunits. Importantly, metagenomic analysis indicated that tebuconazole not only led to significant shifts in the composition and diversity of the gut microbiota but also changed intestinal homeostasis. Specifically, after exposure, tebuconazole circulated in the bloodstream and largely entered the gut–brain axis for disruption, including disturbing the Firmicutes/Bacteroidetes ratio, interrelated neurotransmitters and systemic immune factors. Moreover, pretreatment with probiotics improved immune factor expression and restored the deterioration of synaptic function and spatial learning and memory. The current study provides novel insights concerning perturbations of the gut microbiome and its functions as a potential new mechanism by which tebuconazole exposes cognitive function-related human health.

Detection of myclobutanil and tebuconazole in apple using magnetic molecularly imprinted polymer surface-enhanced Raman spectroscopy

Jingjing Li,Jia Cheng,Juan Du,Ming Xiao,Miao Wang,Jing Wang,Yongxin She,A.M. Abd El-Aty,Xiaolin Cao

The main challenge in Raman detection of pesticide residues in agricultural products is matrix interference, which hinders accurate identification of trace compounds in complicated agricultural samples. In this study, a novel surface-enhanced Raman spectroscopy approach based on magnetic molecularly imprinted polymers (MMIP-SERS) was introduced for the rapid analysis of myclobutanil and tebuconazole in apples. The synthesized magnetic molecularly imprinted polymer selectively adsorbed triazole pesticides, effectively eliminating matrix interference in real samples. The saturation adsorption time for MMIPs was only 10 min. In addition, the synthesized colloidal gold served as an excellent substrate for enhancing the specific signals for myclobutanil and tebuconazole . We successfully identified the characteristic peak of tebuconazole at 1089 cm −1 , while myclobutanil showed its characteristic peak at 632 cm −1 . The series of experiments and recovery tests confirmed the high sensitivity of the developed MMIP-SERS method using colloidal gold as an enhanced substrate for fungicide analysis in apple samples. The linear detection ranges for myclobutanil and tebuconazole in apples were determined to be 0.2–10 mg/kg and 0.5–10 mg/kg, respectively. The limit of detection was 0.09 mg/kg for tebuconazole and 0.05 mg/kg for myclobutanil. This study provides a rapid and simple method for detecting myclobutanil and tebuconazole based on the synergistic effect of the imprinted polymer and SERS with good separation and detection performance.

Porphyrin-Based Covalent Organic Framework Reinforced Hollow Fiber for Solid-Phase Microextraction of Tebuconazole and Propiconazole

Marzieh Kavian,Milad Ghani,Jahan Bakhsh Raoof

Herein, an amino-functionalized covalent organic framework was synthesized and accommodated in the pores of porous hollow fiber. In this context, tetra (4-aminophenyl) porphyrin was synthesized for preparing the desired covalent organic framework as the extracting sorbent and employed for hollow fiber solid-phase microextraction of tebuconazole and propiconazole. With respect to the amino groups of the as-synthesized porphyrin-based covalent organic framework, the extracting device has the ability of establishing a hydrogen bond with the selected model analytes. Under the optimum condition, the linear range of the method for both analytes were estimated in the range of 0.5–200 µg L −1 (the coefficient of determination of 0.9962 for tebuconazole and 0.9990 for propiconazole). The limits of detection of the method for tebuconazole and propiconazole were calculated to 0.02 µg g −1 and 0.03 µg g −1 , respectively. The limits of quantification of the method were also estimated for two analytes equal to 0.07 and 0.08 µg g −1 , respectively. The intra- and inter-day relative standard deviations, which fell between 1.8% and 4.7%, were computed to assess the accuracy of the suggested approach. The proposed method was used for the extraction and determination of tebuconazole and propiconazole in tomato, cucumber, apple, cabbage, and carrot, which the obtained results showed the success of the method in extracting and determining of these analytes from the target samples.

Histology and multi-omic profiling reveal the mixture toxicity of tebuconazole and difenoconazole in adult zebrafish

Jinhua Jiang,Liezhong Chen,Xingang Liu,Luyan Wang,Shengan Wu,Xueping Zhao

The combination effects of triazole fungicides on aquatic organisms remain largely unknown. In current study, an integrated histological, transcriptome, metabonomics and microbiology was applied to investigate the mixture effects and risk of tebuconazole (TEB) and difenoconazole (DIF) co-exposure on zebrafish liver and gonad at aquatic life benchmark. TEB and DIF mixture showed additive effect on the acute toxicity to adult zebrafish, the combined toxicity on liver was less than the additive effect of individual TEB and DIF, and TEB and DIF mixture also reduced the toxic effects on gonad and intestinal microflora. Transcriptomics and metabolomics further showed TEB and DIF mixture could induce more differentially expressed genes (DEGs) to regulate the metabolic pathways involved in energy metabolism, steroid hormone biosynthesis, retinol metabolism and microbial metabolism, to balance the energy metabolism and supplies, and maintain the steroid hormone and RA level, further reduced the toxic effect on liver and gonad caused by TEB and DIF. Our results showed the different responses and patterns on transcriptional and metabolic profiles mediated in the diverse toxicity and combination effects of TEB and DIF. The present results provided a deep mechanistic understanding of the combined effects and mode of action of DIF and TEB mixture on aquatic organisms, suggesting the concept of additive effects might sufficiently protective when evaluated the combination effects and ecological risk of TEB and DIF at aquatic life benchmarks.
