A sensitive and selective colorimetric biosensor for determination of gentamicin, amikacin and tobramycin was proposed with the unmodified gold nanoparticles (GNPs) as the sensing element. Gentamicin, amikacin and tobramycin can rapidly induce the aggregation of gold nanoparticles and is accompanied by a color change from red to blue. The concentration of gentamicin, amikacin and tobramycin can be determined by using UV-Vis spectrometer. The experimental parameters were optimized with regard to pH, incubation time and the concentration of the GNPs. Under optimal experimental conditions, the linear range of the colorimetric sensor for gentamicin/amikacin/tobramycin were 2.67–33.93 ng mL -1 , 13.33–66.67 ng mL -1 and 20–180 ng mL -1 , respectively. The corresponding limit of detection (3σ) was 0.354 ng mL -1 , 0.999 ng mL -1 and 0.579 ng mL -1 , respectively. This assay was simple and used to detect aminoglycoside antibiotics in milk and medicine products.

Chitosan (CS) is a biodegradable, biocompatible, and non-toxic natural amino-poly-saccharide with antibacterial ability, owing to its positively charged amino groups. However, the low charge density leads to poor antibacterial efficiency which cannot meet the biomedical application requirements. In this study, Tobramycin (TOB) was grafted onto the backbone of oxidized chitosan (OCS) to synthesize oxidized chitosan-tobramycin (OCS-TOB). FTIR,1H NMR and elemental analysis results demonstrated that OCS-TOB was successfully synthesized. OCS-TOB/PEO composite fibrous materials were produced by a self-made centrifugal spinning machine. In vitro experiments showed that cells proliferated on the submicro-fibrous OCS-TOB/PEO of appropriate concentration, and the antibacterial ability of OCS-TOB was much improved, compared with pristine CS. The results demonstrated that OCS-TOB/PEO nanofibrous materials could potentially be used for biomedical applications. Keywords:chitosan;tobramycin;centrifugal spinning;submicro-fibers;biomedical application

Sepsis is a highly heterogeneous syndrome normally characterized by bacterial infection and dysregulated systemic inflammatory response that leads to multiple organ failure and death. Single anti-inflammation or anti-infection treatment exhibits limited survival benefit for severe cases. Here a biodegradable tobramycin-loaded magnesium micromotor (Mg-Tob motor) is successfully developed as a potential hydrogen generator and active antibiotic deliverer for synergistic therapy of sepsis. The peritoneal fluid of septic mouse provides an applicable space for Mg-water reaction. Hydrogen generated sustainably and controllably from the motor interface propels the motion to achieve active drug delivery along with attenuating hyperinflammation. The developed Mg-Tob motor demonstrates efficient protection from anti-inflammatory and antibacterial activity both in vitro and in vivo. Importantly, it prevents multiple organ failure and significantly improves the survival rate up to 87.5% in a high-grade sepsis model with no survival, whereas only about half of mice survive with the individual therapies. This micromotor displays the superior therapeutic effect of synergistic hydrogen-chemical therapy against sepsis, thus holding great promise to be an innovative and translational drug delivery system to treat sepsis or other inflammation-related diseases in the near future.

The effective removal of hydrophilic pollutants is of vital significance for environmental protection mainly because of the low efficiency of traditional sewage treatment methods. In this work, the cyano-vinylene-linked covalent organic polymer was prepared using 2,2′,2′′-(benzene-1,3,5-triyl)triacetonitrile (BTNA) and 2,4,6-tris(4-formylphenyl)-1,3,5-triazine (TFPT) as the Knoevenagel reaction monomers. The cyano group was then hydrolyzed to obtain a carboxyl-functionalized covalent organic polymer (COP-COOH). With good surface area (73.9 m2 g–1) and large channel (∼3.82 nm), TFPT-BTAN-COP-COOH showed fast adsorption equilibrium and high adsorption capacity for tobramycin and ofloxacin via multiple-functional binding sites. Specifically, the electrostatic interaction between the carboxyl group and basic groups (e.g., amino, piperazine, and dihydropyridine) plays a critical role for the selective removal of tobramycin and ofloxacin. The binding energies of TFPT-BTAN-COP-COOH adsorbed tobramycin and ofloxacin calculated using the density functional theory are −59.83 and −35.63 kcal/mol, respectively. For other aminoglycoside and quinolone antibiotics, such as enrofloxacin, norfloxacin, neomycin, and streptomycin, the removal rate can reach over 95% within 30 min using TFPT-BTAN-COP-COOH as the adsorbent. This work explored a promising strategy for the elimination of hydrophilic organic pollutants during environmental remediation.

Tobramycin aptamer was adsorbed on the surface of gold nanoparticles (Au NPs) by van der Waals forces , hydrogen bonds , and other factors, limiting nanozyme peroxidase activity. Because tobramycin has a higher affinity for the tobramycin aptamer, including tobramycin in Au@Apts nanozymes was able to dislodge the aptamer from the surface of Au NPs and restore the catalytic activity of nanozymes. Raman spectroscopy of the enzyme-catalyzed product TMB ox can detect this shift in enzyme activity . The restored value of nanozyme activity increased with tobramycin concentration and had a good linear correlation in the concentration range of 10 −10 ∼10 −1 M with a detection limit of 2.04 × 10 −11 M. Aptamer biosensors are built using the SERS platform, which has the advantages of high sensitivity, high precision, and a wide detection range. It is prospectively employed in food safety and the detection of antibiotic residues.

In addition to its application in genome editing, the CRISPR/Cas system provides a new molecular tool for the construction of biosensor platforms due to its high target sequence specificity and programmability . To exploit the potential of the CRISPR/Cas system for detecting non-nucleic-acid targets, two ultrasensitive biosensors (sensor-ss and sensor-ds) consisting of allosteric aptamer probes integrated with the CRISPR/Cas12a system were designed and constructed for tobramycin detection. Both sensors utilized hairpin DNA containing a tobramycin aptamer sequence as the target recognition probe, which can respond to target binding and further produce signal transduction sequences. However, there are two differences between the sensors. First, in sensor-ss, single-stranded DNA was set to be the activator to trigger cutting activity ( trans -cleavage) of CRISPR/Cas12a, leading to cleavage of the FQ-reporter (a short single-stranded DNA labeled with a fluorophore and quencher at each end) and increasing the fluorescent signal. However, double-stranded DNA containing a protospacer adjacent motif (PAM) site was produced in sensor-ds as the signal transduction sequence, which was recognized by CRISPR/Cas12a, resulting in the cleavage of the FQ-reporter. Second, sensor-ss involved a strand displacement amplification (SDA) reaction as the signal enhancement step, while no DNA amplification procedure was designed in sensor-ds. Through DNA sequence and reaction condition optimization, sensor-ds showed a linear relationship between the fluorescence response and tobramycin concentrations ranging from 10 to 300 pM with a limit of detection (LOD) as low as 3.719 pM. Although sensor-ss exhibited higher sensitivity (LOD = 1.542 pM) than sensor-ds, rapidly reaching the fluorescent signal saturation platform led to a narrow linear response range (5–30 pM). Moreover, our CRISPR/Cas12a-based assay can realize on-site detection by the naked eye readout under UV light. The constructed sensor-ds was also used to detect tobramycin in milk and lake water samples, and the results indicated that the detection system was reliable. The design strategy reported in this study could shed new light on CRISPR/Cas12a-based biosensor construction for small molecule detection.

A novel cascade dual-DNAzyme amplification platform was engineered. • The aptasensor utilized a non-competitive, strand displacement-free transduction mechanism. • The aptasensor exhibits excellent analytical performance with the LOD of 21.0 nM. • Practical applicability was validated in real food samples Tobramycin (TOB), a widely employed broad-spectrum antibiotic in animal husbandry, leaves residual traces that exert inevitable adverse effects on human health, including kidney impairment, neuromuscular blockage, respiratory paralysis, and cross-allergic reactions. Herein, we constructed a novel ligand-induced assembled E6-split DNAzyme cascaded 8–17 DNAzyme aptasensor and applied it to the highly sensitive detection of TOB. In this sensing design, we incorporated the aptamer sequence with a short stem into the input strand. In the absence of the target TOB, the structurally flexible aptamer-programmed input strand fails to form a stable complex with the E6-split DNAzyme. Upon binding of TOB to the aptamer, the input strand undergoes a transition to a compact and rigid conformation, thereby forming a stable three-strand complex consisting of the input strand, E6-split DNAzyme 1, and E6-split DNAzyme 2; this complex assembles a catalytically active domain and restores the substrate cleavage activity. Upon activation, the DNAzyme cleaves a rationally designed hairpin DNA containing an RNA cleavage site to release an extra 8–17 DNAzyme sequence. The liberated DNAzyme subsequently drives the continuous cleavage of fluorophore-quencher labeled reporter probes, resulting in the progressive accumulation of fluorescence signals. Under the optimized reaction conditions, the detection range displays a well-defined linear relationship from 100 nM to 200 μM, with a limit of detection (LOD) of 21.0 nM. By circumventing equilibrium constraints and obviating protein enzymes or nanomaterial modifications, this cascade DNAzyme architecture offers a versatile, sequence-independent, and robust strategy for small-molecule antibiotic monitoring, with broad potential for adaptation to diverse non-nucleic acid targets in food safety. As temporary skin substitutes, wound dressings should have multiple functions, such as anti-inflammatory, sterilization, and tissue regeneration-promoting effects. In this study, a multilevel construction strategy was used to construct a bilayer tobramycin (TOB)/acid fibroblast growth factor-coloaded composite functional dressing by electrospinning, emulsification cross-linking, and freeze-drying for sustained drug release and wound repair. The dressing simulated the double-layer structure of the skin, with an upper layer composed of a TOB-loaded poly(lactic-co-glycolic acid)/multiwalled carbon nanotube electrospun membrane, which can prevent body fluid loss, resist external bacterial invasion, and act as a protective barrier, and a lower layer composed of an FGF1-loaded calcium alginate microsphere/chitosan composite sponge with hemostatic, moisturizing, cell proliferation- and tissue regeneration-promoting, and wound healing acceleration functions. The morphology, water contact angle, cell proliferation and migration abilities, antibacterial properties, and angiogenic and coagulation effects of the dressing were determined in vitro, and its skin wound repair properties were evaluated in vivo. The upper layer of the dressing was hydrophobic, whereas the lower layer was hydrophilic. Moreover, the dressing significantly promoted L929 cell proliferation and migration; had strong antibacterial effects on Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa; and promoted angiogenesis and the healing of full-thickness skin wounds in Sprague-Dawley (SD) rats, along with possessing good hemostatic properties.

A highly sensitive photoelectrochemical aptasensor based on phosphorus-doped hollow tubular g-C 3 N 4 /Bi/BiVO 4 (PT-C 3 N 4 /Bi/BiVO 4 ) for tobramycin (TOB) detecting was developed. This aptasensor is a self-powered sensing system which could generate the electrical output under visible light irradiation with no external voltage supply. Based on the surface plasmon resonance (SPR) effect and unique hollow tubular structure of PT-C 3 N 4 /Bi/BiVO 4 , the PEC aptasensor exhibited an enhanced photocurrent and favorably specific response to TOB. Under the optimized conditions, the sensitive aptasensor presented a wider linearity to TOB in the range of 0.01–50 ng mL −1 with a low detection limit of 4.27 pg mL −1 . This sensor also displayed a satisfying photoelectrochemical performance with optimistic selectivity and stability. In addition, the proposed aptasensor was successfully applied to the detection of TOB in river water and milk samples.

A novel photoluminescent Hcy-AuNCs has been developed through one-pot reduction method, to establish a tobramycin sensing by second-order scattering (SOS). Hcy-AuNCs could spontaneously assemble to small-scaled aggregation, resulting in remarkable intensity enhancement of scattered luminescence signals. The luminescence of Hcy-AuNCs could be clearly observed under ultraviolet lamp, when excited at 365 nm, a significant luminescent intensity at 741 nm was monitored in SOS spectra. The introduction of AuNPs would cause large-scaled aggregation of Hcy-AuNCs that was rapidly settled in the solution, resulting in the decrease of SOS intensity. Besides, the non-radiative energy transfer between AuNPs and Hcy-AuNCs would also reduce the luminescent intensity. However, the addition of tobramycin would cause the aggregation of AuNPs due to the electrostatic and covalent bonding between AuNPs and tobramycin, thus eliminating the interference of AuNPs. The luminescence of Hcy-AuNCs reappeared, exhibiting an optical response toward tobramycin. The good linearity was obtained in a wide range from 4 nM to 300 nM with a low detection limit of 0.27 nM. The selectivity was acceptable toward different types of antibiotics. Finally, the proposed method was successfully applied to the widely used tobramycin eye drops.

Tobramycin (TOB) is a broad-spectrum aminoglycoside extensively used as veterinary medicine to treat bacterial infections. However, TOB residue in food and environment may cause great threat to human health. Here, we report, for the first time, the upconversion nanoparticles (UCNPs) for TOB detection based on Förster resonance energy transfer (FRET). The upconversion (UC) luminescence from core-shell structured UCNPs was effectively quenched after addition of MnO 2 nanosheets due to the efficient energy transfer from the UCNPs to MnO 2 nanosheets. When interacting with TOB, the UC luminescence was restored and increased as a result of coupling between TOB and aptamer, with MnO 2 nanosheets dropping off the latter one. The UCNPs@SiO 2 -MnO 2 aptasensors for TOB detection have a good selectivity and sensitivity in a linear range of 10–180 ng mL −1 with a limit of detection (LOD) of 3.04 ng mL −1 . The sensors were tested for TOB detection in real samples of liquid milk and fishes and the results are very good and satisfactory. These type of aptasensors are promising for practical applications as they have a high sensitivity, selectivity, low LOD, and are autofluorescence-free.

An inhalable chitosan-coated liposomal system was developed for Tob/NO co-delivery. • LC@Tob/NO enhanced mucus penetration and enabled mildly acidic pH-responsive NO release. • Nebulized LC@Tob/NO disrupted MRSA biofilms and alleviated infected lung inflammation. Methicillin-resistant Staphylococcus aureus (MRSA) pneumonia remains difficult to treat because mucus-associated transport barriers, biofilm formation, and excessive inflammation jointly limit therapeutic efficacy. Here, we developed a nebulizable chitosan-coated liposomal co-delivery system, LC@Tob/NO, for pulmonary treatment of MRSA pneumonia. The formulation integrates liposome-mediated pulmonary delivery, chitosan-associated surface reconstruction, tobramycin-mediated antibacterial activity, and mildly acidic inflammation-associated NO release from DETA/NO, thereby enabling localized Tob/NO co-loading for MRSA pneumonia therapy. LC@Tob/NO exhibited favorable physicochemical characteristics, enhanced transport through an artificial mucus matrix, and effective pulmonary deposition after nebulization. In vitro, the formulation showed potent antibacterial and antibiofilm activities against MRSA, while also attenuating oxidative stress and pro-inflammatory activation in macrophages. In a murine MRSA pneumonia model, nebulized LC@Tob/NO markedly reduced bacterial burden, alleviated lung edema and tissue injury, and suppressed inflammatory responses, with acceptable systemic biosafety. Supportive lung transcriptomic analysis suggested treatment-associated changes in immune and inflammatory pathways that were consistent with the observed bacterial clearance, cytokine reduction, and histological improvement. Collectively, LC@Tob/NO represents a promising inhalable biomaterial-based therapeutic strategy for MRSA pneumonia by integrating antibacterial activity with microenvironment regulation.
