As a low-density lipoprotein, tributyrin plays an essential role in food safety and human health. In this study, a novel lipase-conjugated carbon nanotube (CNT) surface plasmon resonance (SPR) fiber-optic sensor is used to specifically detect tributyrin for the first time. In this work, CNTs can be used as an amplifying material to significantly increase the sensitivity of SPR sensors due to their high refractive index and large surface area. CNTs can also be used as an enzyme carrier to provide abundant carboxyl groups for the specific binding of lipases. Covering the surface of the sensor with CNTs can not only enhance the performance of the sensor, but also provide sufficient detection sites for subsequent biomass detection, reduce the functionalization steps, and simplify the sensor preparation process. The experimental results demonstrate that the refractive index sensitivity of the traditional multimode fiber (MMF)–single mode fiber (SMF)–MMF transmissive optical fiber sensor is 1705 nm RIU−1. After covering the sensor with CNTs, the sensitivity is 2077 nm RIU−1, and the sensitivity has been improved very well. In addition, there are abundant functional groups on CNTs, which can provide abundant binding sites. Conjugating lipase on carbon nanotubes helps to achieve linear detection in the range of 0.5 mM to 4 mM tributyrin, with a sensitivity of 4.45 nm mM−1 and a detection limit of 0.34 mM, which is below the 2.26 mM detection standard and meets food safety monitoring requirements. Compared with other sensors, the optical fiber biosensor proposed in this study expands the concentration detection range of tributyrin. Furthermore, the sensor also has good stability, anti-interference performance and specificity. Therefore, the sensor proposed in this paper has good application prospects in the fields of food safety and biomedicine.

Tributyrin is widely used in medical treatment, drug development, and other fields. Lipase can decompose one tributyrin into one glycerol and three butyric acids, causing the effective refractive index (RI) and pH to change. Detecting the RI change or pH change during the experiment is usually used to determine the tributyrin in the sample. However, these methods encounter limitations at low or high concentrations. The RI difference detection method is difficult to implement in low-concentration detection, but the pH detection method is difficult to apply in high-concentration detection. In this article, we present an optical fiber sensor with dual channels based on surface plasmon resonance (SPR) to achieve the measurement of tributyrin. Polyacrylic acid (PAA)/chitosan (CS) pH-sensitive material is coated on half of the SPR sensor as channel 1 to monitor tributyrin-induced pH changes, and the lipase is bonded on the polydopamine (PDA)/maleic acid (MA) coated on the fiber as channel 2 to decompose tributyrin. Experimental results show that channel 1 realizes high pH sensitivity (34.52 nm/pH) and the tributyrin sensitivity reaches 12.8 nm/mM with a concentration between 0.1 and 1 mM. In the concentration range of 1–4 mM, channel 2 exhibits a tributyrin sensitivity of 4.85 nm/mM. In addition, the sensor proposed in this article has excellent stability and selectivity. Thus, the dual-channel optical fiber SPR sensor holds significant potential for drug development and biomedicine.

ABSTRACT The objective of this experiment was to investigate the influence of dietary tributyrin on reproduction performance and ovary function of broiler breeders with different egg laying rate. Two hundred fifty-six AA broiler breeders (48-wk-old) were allocated to 4 treatment in a 2 × 2 factorial arrangement with the main effects of tributyrin supplementation (0 and 1,000 mg/kg tributyrin [ TRI ]) and 2 egg laying rate levels (average [ AR , 81.01 ± 0.79%] and low [ LR , 70.98 ± 0.95%]). The results shown that the LR breeders presented higher egg weight, but lower egg laying rate, qualified egg rate and feed efficiency than the AR breeders ( P (laying) < 0.05). Also, the superoxidase dismutase ( SOD ) activity in magnum was lower while malondialdehyde ( MDA ) was higher in ovary and magnum of LR breeders than that in the AR breeders ( P (laying) < 0.05). Dietary supplementation with tributyrin significantly enhanced egg weight ( P (TRI) < 0.05), increased albumen height as well as Haugh unit ( HU ) in AR breeders ( P (interaction) < 0.05), and also had higher total antioxidant capacity ( T-AOC ) and lower MDA in ovary ( P (TRI) < 0.05). The cell apoptosis rate and proapoptosis related gene expression ( caspase 8, 9 and Bax ) in the ovary of LR breeders was higher, while anti-apoptosis related gene ( Bcl-2 ) expression were lower in LR breeders when compared with the AR breeders ( P (laying) < 0.05). Dietary supplementation with tributyrin decreased the cell apoptosis rate and downregulated caspase 9 expression in LR breeders ( P (Interaction) < 0.05), up-regulated the Bcl-2 expression in both 2 breeders ( P (TRI) < 0.05). These findings suggest that the breeders with lower egg laying rate also characterized by deteriorate ovary function indicated by lower antioxidant capacity and higher cell apoptosis rate. Dietary supplementation with tributyrin increased egg albumen quality, decreased ovarian proapoptosis related gene expression to improve reproductive tract function; and the positive effect on egg albumen quality is more pronounced in average reproductive breeders.

Tributyrin (TB), a stable triglyceride prodrug of butyrate, holds immense potential for modulating gut health and managing ulcerative colitis (UC) via dietary intervention. However, its application in functional foods is restricted by its hydrophobicity, gastric instability, and rapid absorption in the upper gastrointestinal tract (GIT). To address these challenges, this study engineered a food-grade, colon-specific delivery system based on hierarchical emulsion-filled hydrogel microbeads. TB was initially stabilized by octenyl succinic anhydride (OSA)-modified Tartary buckwheat starch to form robust Pickering emulsions (OSP), which were subsequently embedded within a pH-responsive sodium alginate (SA)/carboxymethyl chitosan (CMCS) composite matrix. The intermolecular polyelectrolyte complexation between SA and CMCS, reinforced by Ca 2+ -mediated crosslinking, significantly enhanced the structural integrity and encapsulation efficiency (∼64%) of the microbeads (TB-OSP@C/S). In vitro digestion kinetics demonstrated that this multi-compartment architecture acted as an effective diffusion barrier, suppressing gastric release (< 20%) while facilitating sustained release in the simulated colonic environment driven by pH-dependent swelling and the enzymatic degradation of the protective starch shell. In a DSS-induced murine colitis model, oral administration of TB-OSP@C/S microbeads markedly improved the disease activity index, alleviated colonic tissue damage, and exhibited superior anti-inflammatory efficacy compared to free TB. This work presents a facile, surfactant-free strategy for the precise delivery of hydrophobic nutraceuticals, offering a promising paradigm for developing precision nutrition products targeting intestinal inflammation.

Oral administration of tributyrin, a prodrug of butyrate, to the colon is challenging due to its unpleasant odor, rapid metabolism, and fast absorption within the upper gastrointestinal tract. In this study, the colon-targeted delivery of tributyrin was investigated using Pickering emulsion-filled alginate hydrogel beads. Initially, a tributyrin-loaded oil-in-water Pickering emulsion was formed, containing oil droplets coated with chitin nanofibers and zein colloid nanoparticles. These droplets were then encapsulated within the biopolymer network of calcium alginate beads. The potential of these structured delivery systems to control the release of tributyrin was assessed by using a simulated gastrointestinal tract, anaerobic fecal fermentation, and intestinal accumulation studies. The ability of the encapsulated tributyrin to ameliorate dextran sulfate sodium-induced colitis was evaluated. The Pickering emulsion-filled hydrogel beads retained their integrity under-simulated upper gastrointestinal tract conditions, leading to superior resistance against tributyrin hydrolysis by lipase compared to Pickering emulsions. During fecal fermentation, hydrogel beads were gradually eroded as a result of intestinal microbiota action, resulting in a sustained release of tributyrin over 48 h and a significant enhancement of butyrate levels (2.35 times at 4 h) in the cecum in a rat study. Tributyrin-loaded hydrogel beads appreciably reduced the secretion of pro-inflammatory cytokines, thereby relieving symptoms associated with colitis. In the future, it would be useful to test these tributyrin-loaded delivery systems in clinical trials to assess their potential for the treatment of colitis.

As a precursor to butyric acid and also a short-chain triglyceride, tributyrin plays an important role in repairing small intestinal villi, promoting the absorption and utilization of nutrients, and monitoring human health. The traditional carboxylation functionalization method of optical fiber sensors takes a long time and is complicated to operate. Therefore, we need a sensor with a functional period, high sensitivity, and the ability to quickly detect samples to achieve specific detection of tributyrin. In this paper, based on the chemical cross-linking method of polydopamine-maleic acid (PDA-MA), a novel fiber-optic microfluidic surface plasmon resonance (SPR) sensor was proposed to realize the detection of tributyrin. Compared with the traditional functionalization method, the fixation method of PDA-MA chemical cross-linking method not only shortens the functionalization time, but also increases the sensitivity of the sensor. Experimental results show that the sensitivity of the microfluidic sensor coated with PDA reaches 3392.7 nm/RIU, which is 1.75 times that of the uncoated sensor. The sensor has a sensitivity of 8.22 nm/mM in the range of 0.5–5 mM and a response time of only 8 min at the concentration of 1 mM. In addition, the sample detection error of the sensor is within 12 % in the range of 0.5–5 mM. The proposed sensor offers the advantages of low sample consumption (100 μL), high sensitivity, good selectivity, good repeatability, anti-interference ability, and label-free measurement. The sensor holds promising applications in food safety and health monitoring due to its superior performance.

As a low-density lipoprotein, tributyrin plays a vital role in food safety and human health. In this work, we first proposed a novel high-performance tributyrin sensor made by supporting zeolitic imidazolate framework-8 (ZIF-8) and lipase onto the optic fiber sensor based on the surface plasmon resonance (SPR) principle. After adsorption of lipase within the ZIF-8 framework, through a rapid aqueous synthesis method, a thin and dense ZIF-8 film can be uniformly formed and stably and tightly bonded to the surface of the optical fiber sensor. ZIF-8 is one of the metal–organic frameworks (MOFs), which not only has the high porosity and specific surface area of MOFs, but also shows good affinity with lipase. The experimental results show that the proposed sensor can achieve the specific detection of tributyrin in the linear range of 0.1–3 mM, the sensitivity is 14.75 nm/mM, with a detection limit of 0.096 mM. The sensor proposed in this article for the first time using ZIF-8/lipase composite material to detect tributyrin has an accuracy of more than 83% in sample detection at the range of 0.1–3 mM. In addition, the stability test of the proposed sensor is as good as 0.47 nm, and the selectivity (in the presence of glycine, acetylcholine, glucose, and urea) of the sensor was analyzed. The sensor proposed in this article has the advantages of a low sample consumption of $100~\mu \text{L}$ , high sensitivity, simple structure, and label-free measurement. This method provides a new route for the rapid and reliable detection of tributyrin, which has potential applications in medical research and clinical diagnosis.

Butyric acid is the main nutrient of intestinal epithelium, but it has an unpleasant odor. In order to increase the content of butyric acid in the intestine, soy hull polysaccharide (SHP) nanomicelle carrier was designed to deliver the precursor of butyric acid, tributyrin (TB). The SHP nanomicelles were characterized by laser particle size analyzer, XRD and AFM. And the effects of nanomicelles on the interfacial properties of the digestive system were investigated by simulating the gastrointestinal tract in vitro . The results show that SHP–TB nanomicelles are spherical. The peak value of C=O tensile vibration at 1650 cm −1 increased. The inclusion rate reached 30.44%, and the loading rate was 80.51%. The cumulative TB release rate was <20% after 2 h of release in the stomach and was <25% after 4 h of release in the small intestine, and the cumulative release rate in the colon reached >95%. With prolongation of fermentation, the OD 600 of the fermentation solution significantly increased, the pH and total sugar content significantly decreased, and total sugar utilisation rate >66.04%. The butyric acid content in the fermentation solution significantly increased and the proportion was above 0.4 on average, the average contents of total SCFAs increased from 2.04 mmol/L to 45.79 mmol/L, the interfacial tension decreased and tended to be stable at the late stage of interfacial adsorption. At a 1:0.5 mass ratio, the SHP–TB nanomicelles exhibited the best cell survival rate and colysis process, with no toxicity. Therefore, SHP nanomicelles can target TB delivery as an ideal carrier in the colon, and the intestinal environment is conducive to the degradation of TB into acid, promoting absorption in the body.

The objective of the current research was to evaluate the influence of dietary tributyrin (TB) inclusion on the growth performance and physiological well-being ofHexagrammos otakii. Juvenile fish (initial weight 24.01 ± 1.35 g) were fed one of six isonitrogenous and isolipidic diets containing graded levels of TB (0–1.5 g/kg) for 90 days. Supplementation significantly enhanced the weight gain and specific growth rates. The optimal level of 0.9 g/kg TB improved muscle nutritional quality, muscle texture, and intestinal morphology. This dosage also significantly increased hepatic and intestinal antioxidant capacity, elevating superoxide dismutase (SOD), catalase (CAT), and total antioxidant capacity (T-AOC), while reducing malondialdehyde (MDA) content. Furthermore, dietary TB at 0.9 g/kg reduced plasma and hepatic triglycerides and total cholesterol, and elevated key plasma immune parameters, including complement components (C3, C4) and phosphatase activities (ACP, AKP). Intestinal health was further enhanced, as evidenced by increased digestive enzyme activities (amylase, lipase, chymotrypsin) and improved microbial diversity, marked by a proliferation of beneficial bacteria. In conclusion, dietary TB supplementation at 0.9 g/kg optimally promotes growth, improves muscle and intestinal health, and enhances antioxidant and immune functions inH. otakii.

We investigated the effects of dietary supplementation with sodium propionate (SP) and tributyrin (TB) on hepatic lipid deposition and antioxidant capacity of spotted seabass (Lateolabrax maculatus) via an 8-week feeding experiment and a hypoxia stress experiment. The fish were fed five experimental diets: a control diet (CON), a diet supplemented with 2 g/kg SP (SP-0.2%), 4 g/kg SP (SP-0.4%), 2 g/kg TB (TB-0.2%), or 4 g/kg TB (TB-0.4%). No significant difference in growth performance was presented among the groups (P > 0.05). The SP-0.4% and TB-0.2% groups presented significantly lower hepatosomatic and viscerasomatic indexes compared with the CON group. Then, the SP-0.4% and TB-0.2% groups presented stronger resistance to hypoxic stress than the other groups and were analyzed further. The hepatic histology and triglyceride levels revealed that SP-0.4% and TB-0.2% reduced hepatic lipid deposition. Similarly, the downregulation of malondialdehyde and the upregulation of total antioxidant capacity, superoxide dismutase, and catalase activities and the related gene expression levels revealed that SP-0.4% and TB-0.2% improved the antioxidant capacity. Additionally, the RNA sequencing demonstrated that SP-0.4% and TB-0.2% regulated gene expression to a similar extent. Among the 117 differentially expressed genes, 67 genes were enriched in the same pattern, and involved the FoxO signaling, PI3K-Akt signaling, and insulin-related pathways. In conclusion, supplementing SP-0.4% and TB-0.2% as feed additives effectively improved hepatic lipid metabolism, antioxidant capacity, and hypoxic stress resistance of spotted seabass.

This study aimed to investigate the impact of dietary tributyrin (TB) in the low fish meal (high- Clostridium autoethanogenum protein, CAP) diet on the growth, antioxidation, lipid metabolism and intestinal microbiota of largemouth bass ( Micropterus salmoides ). For eight weeks, largemouth bass (15.05 ± 0.58 g) were fed on eight isonitrogenous and isolipid diets with graded levels of dietary TB (0 g/kg, 0.2 g/kg, 0.4 g/kg, 0.8 g/kg, 1.6 g/kg, 3.2 g/kg, 4.0 g/kg and 8.0 g/kg). The findings showed that the weight gain rate (WGR) and specific growth rate (SGR) were increased by 1.6 g/kg of dietary TB. The protein efficiency and deposition ratios were linearly affected by dietary TB. The broken-line model based on the WGR revealed that 1.617 g/kg of dietary TB was sufficient for optimal growth. The digestive enzyme activities and villus length of the intestine were increased by dietary TB (1.6 g/kg). Total antioxidant capacity (T-AOC), superoxide dismutase (SOD) and catalase (CAT) activities were significantly improved, and the malondialdehyde (MDA) and protein carbonyl (PC) contents were significantly decreased after dietary TB. The expressions of nuclear factor erythroid 2-related factor 2 ( nrf2 ) and kelch like ech associated protein 1 ( keap1 ) in the liver and intestine were significantly increased and decreased by 1.6 g/kg of dietary TB, respectively. The mRNA levels of copper-zinc superoxide dismutase ( cuznsod ), cat , heme oxygenase-1 ( ho-1 ) and glutathione peroxidase ( gpx ) were significantly upregulated. Dietary TB could significantly affect the total protein (TP) and albumin (ALB) content in the serum. The aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities were the lowest at 1.6 g/kg of TB group. Besides, the hepatocyte vacuolation and nuclear translocation were alleviated after supplementation of 0.2‰− 3.0‰ of dietary TB. The total cholesterol (T-CHO), high density lipoprotein cholesterol (HDLC) and low density lipoprotein cholesterol (LDLC) were significantly increased by dietary TB. The triglycerides (TG) content was decreased by dietary TB. The mRNA levels of sterol regulatory element-binding protein 1-c ( srebp1-c ), acetyl-coa carboxylase alpha ( accα ), fatty acid synthase ( fasn ) and accβ in the liver were significantly downregulated, and peroxisome proliferator-activated receptor alpha ( pparα ) and carnitine palmitoyltransferase 1 ( cpt-1 ) were upregulated by 1.6 g/kg of dietary TB. Dietary TB had effects on the intestine microbes of largemouth bass. The Cetobacterium was present in the highest percentage and increased from 41% (0 g/kg TB) to 59% (1.6 g/kg TB). The Mycoplasmataceae was the second most predominant genus and decreased from 90% in the 0 g/kg of TB group to 10% in the 1.6 g/kg of TB group. In summary, the dietary TB could be supplemented in the CAP-based diet for better growth and antioxidation, lipid metabolism and intestinal microbiota of largemouth bass.
