It produces 7-ACA (7-aminocephalosporanic acid) from CPC (cephalosporin C) by one-step.
Standard
Activity
≥130 U/g wet weight
Size of immobilization support
150~300μm (> 95%)
Water content
<60%
Enzyme assay
One unit (U) represents the amount of enzyme required for adding 1μmol NaOH per min to adjust pH with degradation of CPC to 7-ACA in 20mM K-phosphate buffer containing 40mM CPC, pH 8.3, at 25℃.
1. Remove the water after washing immobilized enzyme with water more than three times. 2. Dissolve 75mM CPC-Na and 0.3% NaHCO₃ in 130ml. 3. Leave the substrate in gradient cylinder for 10min at 20℃. 4. Pour the catabolic enzyme (Immobilized Cephalosporin C Acylase, 6.5 U/ml) in conversion reactor and measure time with pouring substrates. 5. Adjust pH to 8.3 with 1.0N NaOH. 6. Extract 0.1ml reaction solution (supernatant) and dilute with water in 100ml flask. 7. Measure the yield of 7-ACA through peak area in HPLC detector.
