D (Actinomycin D)
(Camptothecin)
(Staurosporine)

This Apoptosis Inducers Kit (A1524530) contains three reagents that induce apoptosis through distinct molecular mechanisms: Actinomycin D, Camptothecin, and Staurosporine. These three inducers complement each other and can induce apoptosis in a wide range of common cell lines (e.g., HeLa, HEK293, CHO, COS, Jurkat, etc.), making them suitable for establishing positive controls in apoptosis research, drug screening, and mechanistic studies.
As apoptosis induction exhibits cell-type specificity and different cell lines may show varying sensitivities to the same inducer, this kit provides three inducers with distinct mechanisms of action to effectively enhance applicability across diverse cell types.
1. Actinomycin D
Mechanism: Antitumor antibiotic that intercalates into DNA and inhibits RNA synthesis (transcription inhibitor).
Typical Applications:
mRNA stability assay (transcriptional arrest): After adding Actinomycin D to block new RNA synthesis, measure the decay of a specific mRNA over time to calculate its half‑life.
Transcriptional regulation studies: Investigate whether gene expression depends on ongoing transcription.
Apoptosis model: Induce transcription‑dependent apoptosis.
Usage:
Dilute 1:1000 into culture medium (e.g., 1 μL stock into 1 mL medium). Initial working concentration = 10 μM.
Treatment time: typically 6-24 hours, but optimize according to cell type.
Reference range in literature: 0.25-4.0 mg/L for Vero cells, 12-60 hours.
2. Camptothecin
Mechanism: DNA topoisomerase I inhibitor. By inhibiting Topo I activity, it causes DNA damage, activates p53, and upregulates TRAIL‑R2 (DR5) and Bak expression.
Typical Applications:
Positive control for apoptosis assays: As a general method for inducing apoptosis, it provides a reliable positive control for various cell death detection assays (e.g., Annexin V staining, TUNEL staining, Caspase activity assays).
DNA damage and repair studies: As a specific Topo I inhibitor, it is used to study DNA damage response and repair mechanisms.
Cell cycle and apoptosis studies: Camptothecin primarily acts on S‑phase cells and is commonly used to investigate cell cycle checkpoint activation and apoptotic pathways.
Anticancer drug mechanism studies: Camptothecin and its derivatives (irinotecan, topotecan) are clinical anticancer agents; useful for studying anticancer mechanisms or drug combination effects.
Usage:
Dilute 1:1000 into culture medium. Initial working concentration = 2 μM.
Treatment time: typically 12-48 hours.
Reference range in literature: 50 nmol/L induces apoptosis in U937 cells at 24 h; 100 nmol/L-10 μmol/L effective in HeLa cells at 24 h.
3. Staurosporine
Mechanism: A natural product alkaloid isolated from Streptomyces staurosporeus and a broad-spectrum protein kinase inhibitor. It targets the ATP-binding sites of multiple kinases (including PKC, PKA, etc.), inhibiting their activity and inducing apoptosis. Staurosporine induces DNA fragmentation and apoptosis within 2-3 hours at 1 μM.
Typical Applications:
Positive control for apoptosis assays: Staurosporine is one of the most commonly used positive inducers of apoptosis, suitable for Annexin V staining, TUNEL staining, Caspase activity assays, and other apoptosis detection methods.
Protein kinase signaling pathway studies: As a broad-spectrum kinase inhibitor, it is used to study kinase-mediated signal transduction pathways.
Apoptosis mechanism studies: Its rapid induction of apoptosis makes it an ideal tool for studying apoptotic signaling pathways.
Usage:
Dilute 1:1000 into culture medium (e.g., 1 μL stock solution into 1 mL medium). Initial working concentration is 5 μM.
For sensitive cell lines (e.g., Jurkat, HL-60), a common starting concentration is 1 μM with 3-6 hours incubation to effectively induce apoptosis.
A time-course experiment is recommended to determine the optimal treatment time.
Reference concentration: Literature reports a common induction concentration of 500 nM; for Jurkat cells, 1 μM for 4 hours is recommended; 1 μM for 4-5 hours is very effective in HL-60 and Jurkat cells; 2 μM effectively induces apoptosis in HT-1080 cells. For sensitive cell lines (e.g., murine cortical neurons), concentrations as low as 30-100 nM can induce apoptosis.
Summary: How to Choose Which Inducer to Use?
The three inducers in this kit induce apoptosis through distinct mechanisms and are suitable for different research scenarios. Please select the appropriate inducer according to your experimental objectives – do not mix or substitute them arbitrarily.
Transcription or mRNA stability studies → Actinomycin D
DNA damage and repair studies, or as a positive control for apoptosis → Camptothecin
Important Note: These three inducers have completely different targets and mechanisms. Using an incorrect inducer without a clear purpose may fail to achieve the expected apoptotic effect or may lead to misleading results. If combination treatment with multiple inducers is required, please perform pilot experiments to optimize concentrations and treatment times, and include appropriate single-agent controls.
Components
|
1. Recommended Starting Concentration: A 1000-fold dilution of the provided stock solutions is recommended as a starting point for inducing apoptotic activity in many cell culture systems (e.g., add 1 μL to 1 mL of medium). Optimal dosages may vary depending on cell type and culture conditions.
2. Small Volume Addition Tip: For additions to volumes less than 1 mL, dilute the stock solution 1:10 in fresh DMSO, then further dilute 1:100 in medium or PBS to facilitate accurate addition.
3. Dose Optimization: The optimal inducing dose varies by cell type and culture conditions; optimization according to the experimental system is recommended.
4. Incubation: Incubate at 37°C in a 5% CO₂ incubator for 4, 8, 12, 16, or 24 hours, then observe apoptosis. Typically, obvious morphological changes can be observed under a light microscope after 16 or 24 hours, at which point significant apoptosis should be detectable.
5. In Vivo Applications: For in vivo apoptosis induction, please determine the optimal conditions empirically.
