Gynecological specimens are collected with dedicated samplers following standard procedures. Place the sampler into a vial with preservative solution and thoroughly rinse it by moving up, down, left and right. Unidirectional stirring should be avoided to prevent cell-containing mucus from adhering to the sampler surface. Most gynecological specimens require no additional treatment and can be directly transferred into preservative vials. Approximately 15% of specimens contain excessive blood or mucus, which interferes with slide evaluation and requires homogenization pre-treatment.
Common specimen pre-treatment protocols include acetic acid digestion and acetic acid‑dithiothreitol (DTT) digestion. Acetic Acid Digestion Solution (Wash Solution) is composed of glacial acetic acid and purified water. It is used for homogenization pre-treatment of gynecological and non-gynecological specimens (aspirates, urine, sputum, pleural effusion, exudates, secretions from respiratory tract, gastrointestinal tract, breast, etc.), especially for specimens with high levels of blood or mucus.
For research use only. Not for clinical diagnosis or other applications.
Operating Procedures (For Reference Only):
(I) Mucus Specimens (e.g. Sputum)
1. Sampling and reagent addition: Transfer an appropriate amount of sputum specimen (soybean grain size) or other sample into a centrifuge tube containing 20 mL normal saline. Add approximately 10 mL Acetic Acid Digestion Solution (Wash Solution). For heavily blood-contaminated specimens, add 20–30 mL of this solution directly.
2. Vortex and centrifugation: Vortex and mix for 10 min, then centrifuge at 1500–2000 r/min for 5–10 min. Discard the supernatant. Repeat the procedure if blood or mucus is still visible in the cell pellet.
3. Transfer and slide preparation: Discard the supernatant and transfer the pellet into a specimen vial with cell preservative solution. Let stand for ≥15 min before slide preparation.
(II) Aspirate Specimens
1. Sampling and reagent addition: Place the specimen into a centrifuge tube and add 30 mL wash solution. For bloody specimens, add 20–30 mL of this product.
2. Vortex and centrifugation: Vortex at 1500–2000 r/min for 10 min, then centrifuge at 1500–2000 r/min for 5–10 min. Discard the supernatant. Repeat if blood or mucus remains visible.
3. Transfer and slide preparation: Transfer the pellet into the cell preservative vial. Let stand for ≥15 min before slide preparation.
(III) Bronchoscopic Brush Washings
1. Brushing: Immerse the bronchoscopic brush into 30 mL wash solution and rinse. Add an additional 20–30 mL of this product for blood-rich specimens.
Precautions:
1. This product contains glacial acetic acid, which is corrosive and emits an irritating odor. Perform operations in a well-ventilated area.
2. Specimens in preservative solution should appear slightly translucent. Re-dilute if turbidity occurs.
3. Adjust the volume of digestion solution according to the blood content of specimens. Excessive dosage may damage cell morphology; insufficient dosage results in poor blood removal. Use the standard volume (20–30 mL) for initial trials and fine-tune based on experience.
4. Do not attempt complete one-step cleanup for heavily contaminated specimens. Remove impurities gradually via repeated centrifugation and reagent addition to preserve intact target cellular components.
5. Discontinue use if the solution becomes turbid or forms precipitates.
6. Wear a lab coat and disposable gloves during operation for personal safety and health protection.
7. Use the reagent promptly after opening to avoid compromising experimental results.
