布雷菲德菌素A（BFA）是一种细胞内蛋白质转运抑制剂。在细胞体外活化过程中添加BFA，可通过阻断蛋白质向高尔基体复合体的转运，导致其在内质网中蓄积，从而显著增强细胞内细胞因子的染色检测信号。该方法对大多数小鼠和人细胞的细胞因子检测均有效。其最佳使用条件（如处理时长）因细胞类型和具体实验体系而异；通常在收获细胞前的最后几小时至4-24小时内加入。建议研究者在特定实验体系中评估BFA以及其他抑制剂（如莫能菌素）的效果。BFA溶液常以1000X浓缩液形式提供，使用时需按1:1000比例用细胞培养液稀释至工作浓度（1X) 。

Brefeldin A (BFA) is an intracellular protein transport inhibitor. Adding BFA during in vitro cell activation blocks protein transport to the Golgi Complex, causing accumulation in the Endoplasmic Reticulum, thereby enhancing the detection signal for intracellular cytokine staining. This method is particularly useful and effective for staining most cytokines in mouse and human cells. Optimal conditions, including the duration of treatment, are cell type and assay-dependent. Typically, BFA is added for the last few hours up to 4-24 hours prior to cell harvest. Researchers are advised to evaluate the performance of BFA, along with other transport inhibitors like monensin, in their specific assay system. Brefeldin A Solution is commonly supplied as a 1,000X concentrate and should be diluted to a 1X working concentration in cell culture medium.
Usage Protocol
Dilute the 1000X solution to 1X in the cell culture medium.
Precautions
It is recommended that cells are cultured with BFA for ≤ 24 hours, as this can become toxic for cell viability.
