概述
结晶紫染色液(Crystal Violet Staining Solution)是一种组织或细胞染色时常用的可以把细胞核染成深紫色的染色液。结晶紫是一种碱性染料，可以和细胞核中的DNA结合，从而产生细胞核染色。

操作步骤：
样品处理：
a)石蜡切片：
1. 二甲苯中脱蜡5-10分钟。
2. 换用新鲜的二甲苯，再脱蜡5-10分钟。
3. 无水乙醇5分钟。
4. 90%乙醇2分钟。
5. 70%乙醇2分钟。
6. PBS洗，2分钟，3次。
7. 结晶紫染色液10分钟。
8. 用C液封片。
注意：如果颜色不深，可以增加染色时间；如果颜色过深，可以用B液洗，每次30秒到一分钟，后用水洗。
b)冰冻切片：
1. 用4%多聚甲醛固定30分钟。
2. PBS洗3次每次5分钟。
3. 结晶紫染色液10分钟。
4. 用C液封片。
注意：如果颜色不深，可以增加染色时间；如果颜色过深，可以用B液洗，每次30秒到一分钟，后用水洗。
c)培养细胞：
1. 用4%多聚甲醛固定10分钟。
2. PBS洗3次每次5分钟。
3. 结晶紫染色液10分钟。
4. 用C液封片
注意：如果颜色不深，可以增加染色时间；如果颜色过深，可以用B液洗，每次30秒到一分钟，后用水洗。
注意事项：
1.第一次使用本试剂时建议先取1-2个样品做预实验，确定一个好染色时间。

Crystal Violet Staining Solution is a commonly used staining solution in tissue or cell staining that can stain cell nuclei dark purple. As a basic dye, crystal violet can bind to DNA in the cell nucleus, thereby achieving nuclear staining.

Operating Procedures:
Sample Processing:
a) Paraffin Sections:
1. Dewax in xylene for 5–10 minutes.
2. Replace with fresh xylene and dewax again for 5–10 minutes.
3. Immerse in absolute ethanol for 5 minutes.
4. Immerse in 90% ethanol for 2 minutes.
5. Immerse in 70% ethanol for 2 minutes.
6. Rinse with PBS for 2 minutes, repeated 3 times.
7. Stain with Crystal Violet Staining Solution for 10 minutes.
8. Mount the section with Solution C.
Note: If the staining color is too light, extend the staining time. If the staining color is too dark, rinse with Solution B (30 seconds to 1 minute each time), followed by rinsing with water.
b) Frozen Sections:
1. Fix with 4% paraformaldehyde for 30 minutes.
2. Rinse with PBS 3 times, 5 minutes each time.
3. Stain with Crystal Violet Staining Solution for 10 minutes.
4. Mount the section with Solution C.
Note: If the staining color is too light, extend the staining time. If the staining color is too dark, rinse with Solution B (30 seconds to 1 minute each time), followed by rinsing with water.
c) Cultured Cells:
1. Fix with 4% paraformaldehyde for 10 minutes.
2. Rinse with PBS 3 times, 5 minutes each time.
3. Stain with Crystal Violet Staining Solution for 10 minutes.
4. Mount with Solution C.
Note: If the staining color is too light, extend the staining time. If the staining color is too dark, rinse with Solution B (30 seconds to 1 minute each time), followed by rinsing with water.
Precautions:
1. When using this reagent for the first time, it is recommended to take 1-2 samples for a preliminary experiment to determine an optimal staining time.
2. For your safety and health, please wear a lab coat and disposable gloves during operation.
