The activity level of cytoplasmic glycerol-3-phosphate dehydrogenase (ctGPD, EC 1.1.1.8) directly determines the flux of metabolites directed toward glycerol synthesis during glucose catabolism, and consequently dictates the level of glycerol production.
ctGPD is NAD-dependent and catalyzes the conversion of dihydroxyacetone phosphate to glycerol-3-phosphate. The enzyme activity is determined by measuring the decrease in NADH at 340 nm.
