裸鼠成瘤细胞稀释液是裸鼠成瘤实验的专用配套试剂，核心用于肿瘤细胞接种前的稀释与状态维持，确保成瘤效果稳定。专为裸鼠（免疫缺陷小鼠）成瘤实验设计，用于将培养好的肿瘤细胞稀释至适宜接种浓度，同时维持细胞活性，为后续皮下 / 原位接种、肿瘤形成提供稳定的细胞悬液环境。
肿瘤细胞或肿瘤组织块造模方法：一般是皮下注射成瘤（腋下或背部皮下）。
本稀释液：接种细胞密度 1x10⁶cells-5x10⁷cells/ml，接种0.1ml，就是将培养的细胞收集起来调整到适宜浓度重悬于本裸鼠成瘤细胞稀释液中，放于冰盒中携至动物房，直接注射即可。
本稀释液的优点：
1、用稀释液的细胞注射后，不会出现漏出来现象。
2、使用本试剂，提高成瘤率，我们验证的细胞成瘤率100%。
3、使用本试剂，缩短成瘤时间。
4、完全取代基质胶。
使用步骤：
1、准备细胞，调节好细胞状态，最好连续3天做到每天传代细胞，可以1:2传代。
2、收集好细胞，用1ml pbs重悬细胞，做好细胞计数。
3、离心弃掉pbs，保留沉淀，计算好要加的稀释液的量。
4、把细胞中加入需要的稀释液，放入冰盒中备用。
5、把注射器放入冰盒中预冷备用。
6、腋下或背部皮下接种细胞密度 1x10⁶cells-5x10⁷cells/ml，接种100μl，短暂停留数秒后退针。
注意事项：
1、本产品在室温长时间置放，会凝固，放入4度后会自动溶解。
2、本产品4度保存1个月，-20度保存1年。
3、本产品为无菌产品，注意在无菌环境打开。
4、本产品仅限于专业人员的科学研究用，不得用于临床诊断或治疗，不得用于食品或药品，不得存放于普通住宅内。

The Dilution of nude mouse tumor cells is a dedicated supporting reagent for nude mouse tumor formation experiments. Its core function is to dilute tumor cells and maintain their state before inoculation, ensuring stable tumor formation results. Specifically designed for nude mouse (immunodeficient mouse) tumor formation experiments, it is used to dilute cultured tumor cells to an appropriate inoculation concentration while maintaining cell viability, providing a stable cell suspension environment for subsequent subcutaneous/orthotopic inoculation and tumor formation.
Tumor Cell or Tumor Tissue Block Modeling Method:
Typically, tumor formation is achieved via subcutaneous injection (in the axillary region or subcutaneous area of the back).
Usage of This Diluent:
Inoculation cell density: 1×10⁶ cells–5×10⁷ cells/ml
Inoculation volume: 0.1 ml
The specific operation is as follows: Collect the cultured cells, adjust them to the appropriate concentration, resuspend them in this Tumor Cell Diluent for Nude Mice, place the suspension in an ice box, transport it to the animal facility, and then perform direct injection.
Advantages of This Diluent:
1. After injecting cells resuspended in this diluent, there is no leakage.
2. Using this reagent improves the tumor formation rate; the tumor formation rate of cells verified by us reaches 100%.
3. Using this reagent shortens the tumor formation time.
4. It can completely replace Matrigel.
Usage Steps:
1. Prepare the cells and adjust them to a good state. It is recommended to passage the cells daily for 3 consecutive days, with a passage ratio of 1:2.
2. Collect the cells, resuspend them in 1 mL of PBS, and perform cell counting accurately.
3. Centrifuge the cell suspension to discard the PBS, retain the cell pellet, and calculate the required volume of the diluent.
4. Add the required amount of diluent to the cell pellet, then place the cell suspension in an ice box for later use.
5. Place the syringe in the ice box for pre-cooling, and set it aside for later use.
6. For subcutaneous inoculation (in the axillary region or dorsal subcutaneous area): Use a cell concentration of 1×10⁶ cells–5×10⁷ cells/mL, inject 100μL of the cell suspension, and leave the needle in place for a few seconds before withdrawing it.
Precautions:
1. This product will solidify if left at room temperature for an extended period, but it will dissolve automatically when placed at 4°C.
2. This product can be stored at 4°C for 1 month and at -20°C for 1 year.
3. This product is sterile; please open it in a sterile environment.
4. This product is intended solely for scientific research by professional personnel. It shall not be used for clinical diagnosis or treatment, nor for food or pharmaceutical purposes, and shall not be stored in ordinary residential premises.
