概述
外泌体 TSG101 蛋白检测试剂盒是针对外泌体标志蛋白 TSG101 检测而研发的一款方便高效的试剂盒产品。产品中的外泌体蛋白专用裂解液可以高效地裂解外泌体并获得外泌体蛋白；TSG101 阳性对照品及一抗、二抗可以监测实验体系并准确检测到外泌体中的 TSG101 蛋白。

产品使用：
1、 实验准备：实验前先将外泌体蛋白专用裂解液2~8℃融化混匀后置于冰上；
2、 将外泌体样本与裂解液按体积比1:1混匀后置于冰上裂解10 min；
3、 于4℃以12,000 ×g（~12,400 rpm*）离心5 min取上清，用BCA法测定蛋白浓度及进行Western Blot等后续实验；
*为约 7 cm 有效离心半径的小离心机换算（≤2 mL 离心管）。
4、 进行Western Blot实验时，取TSG101阳性对照品5~20 μL与实验样品平行上样。（注：初次使用TSG101阳性对照品时，可根据自身实验条件, 通过预实验确定合适的上样量以获得更佳的实验结果）；
5、 二抗 辣根过氧化物酶标记山羊抗兔 IgG (H+L) 抗体 (HRP)稀释比例建议为1:2000~1:5000；
6、 建议使用ECL化学发光法进行显色反应，其他如DAB法灵敏度欠佳。
注：TSG101阳性对照品上样前需70℃煮样5 min，BCA及Western Blot实验相关试剂需自备。

实验条件：
SDS-PAGE 胶：10% 分离胶
样品上样量：阳性对照品（PC）10 μL，293T 细胞来源外泌体总蛋白 10 μg
一抗：即 TSG101 抗体，稀释比例 1:1000；4℃条件下孵育过夜
二抗：即 辣根过氧化物酶标记山羊抗兔 IgG (H+L) 抗体(HRP) ，稀释比例 1:2000，室温孵育 50 分钟
The Exosome TSG101 Protein Assay Kit is a user-friendly and high-efficiency kit developed for the detection of exosome marker protein TSG101. The special exosome protein lysis buffer included in the kit enables efficient lysis of exosomes to extract exosomal proteins. The TSG101 positive control, primary antibody and secondary antibody are provided to monitor the experimental system and achieve accurate detection of TSG101 protein in exosomes.

Product Protocol
1.Experimental preparation: Thaw the lysis buffer at 2–8 °C, mix thoroughly and place on ice before use.
2.Mix exosome sample with lysis buffer at a volume ratio of 1:1, then lyse on ice for 10 min.
3.Centrifuge at 12,000 ×g (≈12,400 rpm*) for 5 min at 4 °C, collect the supernatant. Determine protein concentration via BCA assay and proceed to subsequent experiments such as Western Blot.
*Conversion value for small centrifuges with an effective radius of approximately 7 cm (for centrifuge tubes ≤2 mL).
4.For Western Blot analysis, load 5–20 μL of TSG101 positive control alongside test samples.
(Note: For first-time use of the TSG101 positive control, perform preliminary tests to optimize loading volume according to your experimental conditions for better results.)
5.The recommended dilution ratio for Goat anti-Rabbit IgG antibody (HRP) is 1:2000–1:5000.
6.ECL chemiluminescence is recommended for signal development; alternative methods such as DAB staining exhibit inferior sensitivity.
Note: Boil the TSG101 positive control at 70 °C for 5 min before loading. Reagents required for BCA assay and Western Blot are not included and should be prepared separately.

SDS-PAGE gel: 10% resolving gel
Sample loading volume: 10 μL of positive control (PC); 10 μg total protein of exosomes derived from 293T cells
Primary antibody: TSG101 specific antibody, diluted at 1:1000; incubate overnight at 4 °C
Secondary antibody: HRP-conjugated Goat Anti-Rabbit IgG, diluted at 1:2000; incubate for 50 minutes at room temperature
