概述

本产品为胰蛋白酶的类似物，来源于微生物，识别位点为赖氨酸或者精氨酸，切割位点为精氨酸或者赖氨酸残基中的羧基端，不含任何动物源成分，无动物源性的病毒污染 (如猪流感病毒、猪细小病毒等), 天然缺乏糜蛋白酶活性，是猪或者牛胰蛋白酶的替代品。本品消化细胞作用温和，并且性质稳定易于存储。

储存液：DPBS 溶液 (含 1.1mM EDTA)。

贴壁细胞消化

使用方法 (仅供参考)

使用前将重组胰蛋白酶 LS 和完全培养基 37℃预热。

1.弃去培养液上清，用适量 DPBS 洗涤细胞，弃去 DPBS。

2.加入适量的重组胰蛋白酶 LS (0.5 ml / 10 cm²),37℃培养直至细胞脱落。

3.加入 5-10ml 的预热完全培养基，轻柔的混匀细胞，将细胞悬液转移至无菌 15ml 离心管中，100g 离心 5-10 分钟。

4.弃去上清，加入适量的完全培养基进行培养。

注意事项

1. 关于类器官的消化建议：囊性的类器官消化 15 分钟，实心的类器官消化时间翻倍；

2. 本品建议 2-8℃保存，特殊情况下，如需冻存，尽量避免反复冻融；

3. 使用时请穿实验服并佩戴一次性手套；

4. 本品不得直接用于临床诊断和治疗。

This product is a trypsin analog derived from microorganisms. It recognizes lysine or arginine residues and cleaves at the carboxyl terminal of lysine and arginine residues. It contains no animal-derived components and is free of animal-origin viral contaminants (e.g., swine influenza virus, porcine parvovirus). Naturally devoid of chymotrypsin activity, it serves as an alternative to porcine or bovine trypsin. This reagent enables gentle cell dissociation and features stable properties for convenient storage.

Storage buffer: DPBS containing 1.1 mM EDTA.

Product Applications

Dissociation of adherent cells

Protocol (For Reference Only)

Pre-warm Recombinant Trypsin LS and complete medium to 37 ℃ prior to use.

1. Discard culture supernatant, rinse cells with an appropriate volume of DPBS, then remove the DPBS.

2. Add an adequate amount of Recombinant Trypsin LS (0.5 mL per 10 cm²), incubate at 37 ℃ until cells detach.

3. Add 5–10 mL pre-warmed complete medium and gently resuspend cells. Transfer the cell suspension into a sterile 15 mL centrifuge tube, and centrifuge at 100 g for 5–10 minutes.

4. Remove the supernatant and resuspend the cell pellet with fresh complete medium for subsequent culture.

Precautions

1. Guidelines for organoid dissociation: incubate cystic organoids for 15 minutes; double the incubation time for solid organoids.

2. Store the product at 2–8 ℃. If cryopreservation is necessary under special circumstances, avoid repeated freeze-thaw cycles.

3. Wear a lab coat and disposable gloves during operation.

4. This product is not intended for direct clinical diagnosis or therapeutic use.
