抗双链 DNA 抗体；天然 DNA 抗体(dsDNA-Ab）是一组抗核抗体（ANA）， 其目标抗原是双链 DNA。它们对系统性红斑狼疮（SLE）有很强的诊断性， 并与狼疮肾炎的发病机制有关。其生成源于免疫系统对自身DNA的错误识别，导致抗体与dsDNA结合，形成免疫复合物，进而引发炎症反应和组织损伤。dsDNA-Ab是SLE的特异性血清学标志物，其水平与疾病活动度密切相关，尤其在狼疮性肾炎等严重并发症中具有重要诊断价值。通过动态检测dsDNA-Ab水平，可评估治疗效果、预测疾病复发风险，并指导个体化治疗。 本试剂盒采用间接法酶联免疫吸附试验（ELISA）。往预先包被有人抗双链DNA抗体;天然DNA抗体(dsDNA-Ab)抗原的微孔中，依次加入样本、阳性对照、阴性对照、HRP标记的二抗，中间经过温育和洗涤，用底物TMB显色，TMB在过氧化物酶(HRP)的催化下转化成蓝色，并在酸的作用下转化成最终的黄色。颜色的深浅和样本中的人抗双链DNA抗体;天然DNA抗体(dsDNA-Ab)呈正相关。用酶标仪在450nm波长下测定吸光度（OD值），判断样本阴阳性。
This kit employs the indirect enzyme-linked immunosorbent assay (ELISA) principle. Microplate wells are pre-coated with Human dsDNA-Ab antigen. The sample or standard is first added to the wells, followed by an HRP (horseradish peroxidase)-labeled secondary antibody. The procedure involves incubation and wash steps between each addition. TMB substrate is then added for color development. Under the catalysis of the peroxidase (HRP), TMB is converted into a blue product, which subsequently turns into a final stable yellow color upon the addition of an acid stop solution. The intensity of the yellow color is directly proportional to the concentration of Human dsDNA-Ab in the sample. The optical density (OD) is measured at a wavelength of 450 nm using a microplate reader, and the concentration of the target analyte in the sample is calculated based on the standard curve.
