酸溶短链脂酰基肉碱经处理后水解为游离肉碱，肉碱与乙酰辅酶A（Acetyl-CoA）在肉碱乙酰转移酶（CrAT）催化下生成游离态辅酶A（CoASH），CoASH与DTNB反应生成TNB，TNB在412 nm处具有特征吸收峰，通过吸光值变化即可定量检测游离肉碱与总肉碱的含量。
Acid‑soluble short‑chain acylcarnitines are hydrolyzed to free carnitine upon treatment. In the presence of carnitine acetyltransferase (CrAT), carnitine reacts with acetyl‑CoA to form free coenzyme A (CoASH). CoASH then reacts with DTNB to generate TNB, which exhibits a characteristic absorption peak at 412 nm. The content of free carnitine and total carnitine can be quantitatively determined by measuring the change in absorbance.
