概述
致畸试验固定液(骨骼检查)，主要由乙醇、纯化水配制的即用型溶液，液体无色澄清。作用原理：高浓度乙醇快速固定胎仔组织蛋白，防止组织自溶、腐烂，完整保留骨骼、软组织原有形态结构，适配完整胎仔法骨骼染色前长期标本固定处理。适用范围：用于食品、化妆品、保健食品、工业化学品毒理学安全性评价，专门用于大鼠、小鼠、家兔致畸试验完整胎仔标本前期固定。该试剂仅用于科研领域，不适用于临床诊断或其他用途。
操作步骤 (仅供参考)：
1、样本取材分组 (大鼠、小鼠适用)：
①取每窝 1/2 剖宫活胎仔，选用完整胎仔染色方案，无需剥皮去内脏；
②将全部胎仔完全浸没于本品中密封浸泡 2–3 周；
注：家兔致畸试验不可分半取样，全部胎仔均需同步固定处理。
2、固定后清洗工序:
①固定期满后取出胎仔，流动流水充分冲洗，去除标本表面残留乙醇；
②冲洗完成后转入 KOH 透明液进行软组织通透处理，进入骨骼染色流程。
3、后续完整试验流程衔接:
固定→冲洗→KOH 透明液通透→茜素红应用液I骨骼染色→透明液 A、B 梯度脱色→甘油保存液封存镜检。
4、镜检统计要求:
① 标本全部处理完成后置于甘油保存液中长期留存；
② 采用透射光源，体视显微镜下依次检查颅骨、脊柱、肋骨、胸骨、四肢骨，记录骨化缺失、骨骼融合、肋骨分叉、骨缺损、骨化发育不全等各类骨骼畸形；
③ 毒理试验统计要求：所有骨骼畸形、发育异常指标同时以胎仔和窝为单位统计，计算胎仔骨骼畸形率、单项骨骼畸形率及窝畸形发生率。
注意事项：
1、密封保存，乙醇极易挥发，长期敞口会降低固定效果，导致标本自溶损坏。
2、仅适用于完整胎仔骨骼染色前固定，剥皮法胎仔无需乙醇长期固定，不可用于内脏组织长期固定。
3、配套试验提示：每窝另一半胎仔可使用致畸试验固定液(内脏检查) (阿拉丁货号：T1529295) 固定，用于内脏畸形同步检测。

Fixative for Teratogenicity Test (Skeletal Examination) This ready-to-use solution is mainly composed of ethanol and purified water, and appears colorless and clear. Principle: High-concentration ethanol rapidly denatures tissue proteins of fetuses to prevent autolysis and putrefaction of tissues. It can well preserve the original morphology of bones and soft tissues, and is suitable for long-term specimen fixation prior to whole-mount fetal skeletal staining.
Scope of application: For toxicological safety evaluation of foods, cosmetics, health foods and industrial chemicals. Specially used for pre-fixation of intact fetal specimens in teratogenicity tests with rats, mice and rabbits. This reagent is for research use only and is not intended for clinical diagnosis or other purposes.
Operating Procedures (For reference only):
1. Sample collection and grouping (Applicable to rats and mice):
①Take 1/2 of the live fetuses obtained by cesarean section from each litter for the whole-mount fetal staining protocol, without peeling skin or removing internal organs.
②Fully immerse all fetuses in this product and seal for soaking for 2–3 weeks.
Note: For rabbit teratogenicity tests, litter splitting sampling is not allowed; all fetuses shall be fixed synchronously.
2. Washing procedure after fixation:
①After fixation, take out the fetuses and rinse thoroughly with running water to remove residual ethanol on the specimen surface.
②After rinsing, transfer the specimens to KOH clearing solution for soft tissue clearing, and proceed to the skeletal staining procedure.
3. Subsequent full test workflow:
Fixation → rinsing → clearing with KOH clearing solution → skeletal staining with Alizarin Red Solution I → gradient decolorization with Clearing Solution A and B → storage in glycerol preservation solution for microscopic examination.
4. Requirements for microscopic examination and statistics:
①All processed specimens are stored long-term in glycerol preservation solution.
②Use transmitted light source, and examine the skull, spine, ribs, sternum and limb bones sequentially under a stereomicroscope. Record various skeletal malformations including ossification deficiency, bone fusion, rib bifurcation, bone defect, hypo-ossification, etc.
③Toxicological statistical requirements: All indicators of skeletal malformations and developmental abnormalities shall be counted at both fetus and litter levels. Calculate the fetal skeletal malformation rate, individual skeletal malformation rate and litter incidence of malformations.
Precautions：
1. Store in a tightly sealed container. Ethanol is highly volatile. Long-term exposure to open air will reduce fixation performance and cause specimen damage due to autolysis.
2. Only for fixation of intact fetuses before skeletal staining. Long-term ethanol fixation is not required for fetuses treated by skin-peeling method, and this product cannot be used for long-term fixation of internal tissues.
3. Supplementary test note: The other half of fetuses from each litter can be fixed with Fixative for Teratogenicity Test (Visceral Examination) (Aladdin Cat. No.: T1529295) for parallel detection of visceral malformations.
4. For your safety and health, please wear a lab coat and disposable gloves during operation.
