Animal model of neonatal hypoxic-ischemic encephalopathy

Summary

Neonatal hypoxic-ischemic eneplalopathy (HIE) is a common clinical disease in the neonatal period, in which children develop a series of encephalopathic manifestations after birth due to hypoxic-ischemic brain damage caused by perinatal hypoxia and asphyxia.

Principle

Formation of hypoxic-ischemic brain injury was induced by first ligating the unilateral common carotid artery of the model animals to reduce cerebral blood flow, followed by inhalation of low oxygen concentrations.

Appliance

The model can be used to explore the pathogenesis, molecular mechanism, clinical symptoms, mechanism function rehabilitation and drug therapy of hypoxic-ischemic encephalopathy.

Operation method

Establishment of an animal model of neonatal hypoxic-ischemic encephalopathy

Principle

Formation of hypoxic-ischemic brain injury was induced by first ligating the unilateral common carotid artery of the model animals to reduce cerebral blood flow, followed by inhalation of low oxygen concentrations.

Materials and Instruments

Animal model:
7 days old 10-15 g neonatal mouse.
Equipment:
Syringe, gelatin sponge, ophthalmic scissors.
Reagents:
① 1% chloral hydrate;

Move

The basic process of establishing an animal model of neonatal hypoxic-ischemic encephalopathy can be divided into the following steps:

A, 1% chloral hydrate (0.15-0.2 mL) was injected intraperitoneally via neonatal rats, anesthetized, and fixed on a surgical plate.

B. Ophthalmic scissors were used to cut the skin of the neck near the midline, ophthalmic forceps were used to bluntly separate the subcutaneous fat, and the left common carotid artery was separated from the medial part of the sternocleidomastoid muscle, and the left common carotid artery was ligated with a 5-0 silk thread to ensure that there was a passage of blood and a sense of pulsation.

C. Gelatin sponge was used to stop bleeding during the operation, and the operation time was 6-10 minutes per patient.

D. After 3-4 hours of postoperative recovery, the mice were placed in an anoxic chamber at 37 ℃, 8% oxygen and 92% nitrogen in a sealed environment for 2 hours, and then returned to their mothers for feeding.

E. The temperature of the feeding environment is (22±2)℃, the humidity is 50%±20%, the day/night cycle is 12/12 hours, and the mice are free to take food and water.
