Animal model of neonatal meconium aspiration syndrome

Summary

Meconium aspiration syndrome (MAS) refers to a group of syndromes in which the fetus inhales amniotic fluid contaminated by meconium during intrauterine or parturition, resulting in airway obstruction, inflammation of the lungs and a series of systemic symptoms, with respiratory distress as the main manifestation, and it is most common in term infants and term infants. For the preparation of animal models of newaborn meconium aspiration syndrome, newborn pigs, sheep and dogs are usually used.

Principle

The simulation of fetal inhalation of amniotic fluid contaminated with fetal inflammation in utero or during labor, airway obstruction, intrapulmonary inflammation, and a series of systemic symptoms, with respiratory distress as the main manifestation, causes an imbalance in the ratio of ventilation to perfusion and results in hypoxia through four main effects, including inhomogeneous airway obstruction, pulmonary surface-active substance (PS) dysfunction, chemical pneumonitis, and pulmonary arterial hypertension.

Operation method

Establishment of an animal model of neonatal meconium aspiration syndrome

Materials and Instruments

Animal model:
Newborn pigs up to 7 days of age
Reagents:
Sodium pentobarbital;
② 20% meconium saline.

Move

The basic process of establishing an animal model of neonatal meconium aspiration syndrome can be divided into the following steps:

A, 3% sodium pentobarbital 1 mL/kg was injected intraperitoneally for anesthesia, and the pulmonary artery, common carotid artery, and external jugular vein were intubated.

B. Tracheotomy and intubation, mechanical ventilation to maintain the normal level of partial pressure of arterial oxygen ( PO2 ), and intratracheal injection of 20% meconium saline 4-6 mL/kg at a uniform rate.

C. If the animals showed obvious cyanosis (severe hypoxemia: PO2<60 mmHg), the modeling was successful.
