Chromosome preparation experiments in the salivary gland of Drosophila melanogaster

Summary

Techniques for stripping salivary glands of larvae such as Drosophila and methods for making salivary chromosome specimens, and observations for characterizing multilineage chromosomes.

Operation method

observation method

Principle

Drosophila salivary gland chromosomes are in a state of homologous chromosome pairing in the somatic cell, due to multiple replication without separation, thus forming a huge chromosome with 1,000-4,000 chromosome filaments, also known as multilineage chromosomes. In this experiment, the salivary gland of a third instar larva of Drosophila was dissected and a chromosome slide specimen was pressed to observe the characteristics of multilineage chromosomes.

Materials and Instruments

Drosophila

Dissecting needle Binocular dissecting microscope Microscope Slide Coverslip

Move

1. Experimental material: third instar larvae of Drosophila melanogaster.
2. Experimental method: Drosophila salivary glands were stripped with a dissecting needle and binocular dissecting microscope, stained with magenta acetate, stained for 10 minutes and then pressed for observation, and slide specimens were made.
