I. Medium components:

Peptone

Beef paste (powder)

Sodium chloride

Glucose

Yeast powder

Sodium citrate

Dipotassium hydrogen phosphate

5 g/L p-aminobenzoic acid solution

1 mol/L magnesium sulfate solution

4.0 g/L phenol red solution

Penicillinase

Sodium polyanisole sulfonate (SPS)

Distilled water

II. Preparation

In addition to phenol red, penicillinase, the rest of the ingredients are mixed and heated to dissolve, correct pH7.4, then add phenol red, divided into glass bottles with a rubber stopper with a flap, 30 ~ 50mL per bottle, insert a needle into the rubber stopper to ventilate the rubber stopper, after autoclaving at 121 ℃ for 15 min, pull out the needle while it is still hot and incubate at 37 ℃ for 24h for the aseptic test, the qualified person is sealed with an aluminum cap, and add 1.5 ~ 2.5 IU of penicillinase into each bottle when it is used. Add 1.5~2.5 IU of penicillinase into each bottle.

Caveat

Take care to avoid staining and to operate aseptically.
