Principle

The basic principle of the single sugar fermentation test is that various bacteria contain different enzymes to decompose sugar (glycosides and alcohols), and their ability to decompose sugar (glycosides and alcohols) varies. For a certain kind of sugar, some bacteria do not decompose, some decompose only acid production, some decompose both acid production and gas production, according to which can be used to identify bacteria.

Observe the results: Because some bacteria can decompose some sugar to produce acid, so the pH in the medium drops to below 7.0, and the color of the medium changes from red to yellow under the phenol red indicator. Acid producer to "+", if at the same time produce gas, the medium in the small inverted tube bubbles appear, this is the production of acid and gas production, to "⊕", do not decompose, then the indicator does not change color, with "-" said. No decomposition, the indicator does not change color and is indicated by "-".

Caveat

The small conduits contained in the sugar fermentation tubes when prepared should be free of air bubbles before inoculation with bacteria, otherwise they should not be inoculated with bacteria.
