Principle

Meiosis is a specialized form of cell division that occurs during gamete formation, in which the chromosomes are replicated only once while the cell divides twice in succession, so that a diploid sex mother cell forms into four haploid sex cells.

I. Materials and methods
1. Materials: onion or broad bean buds, locust spermatheca onion buds, microscope, Carnot's fixative, phenol magenta staining solution.
2. Fixation: Carnoy's fixative, preserved in 70% ethanol.
3. Anther or spermatheca extraction
4. Staining and pressing: staining solution (modified phenol magenta or magenta acetate), slides, coverslips.
5. Chromosome observation: microscope
6. Sealing: Sealing wax, Eupral gum.
II. Steps
1. Taking: Spring inflorescences, young green and full, with smooth involucres (at the peak of division).
2. Fixation: Carnot's fixative for 4-24 hours → 70% preservation in alcohol.

4. Staining: (dispersion) → one drop of staining solution, 10 minutes, pressing, microscopic examination.
