Principle

Mitosis is the process of equal cell proliferation and is the primary mode of somatic cell division . During mitosis, each chromosome in the cell makes a copy and is then distributed to the daughter cells, so that the two daughter cells contain the same chromosomes as the mother cell in terms of number, form, and nature. mitosis exists in a wide variety of vigorously growing plant tissues. Meristematic tissue → fixation → dissociation → staining → pressing → observation (interphase, early, middle, late, terminal)

Move

1. Take: Garlic, onion root tips, thermostat.
2. Pretreatment: drug treatment (colchicine) or freezing (refrigerator).
3. Fixation: water washing → hydrochloric acid 60°C 8-10'dissociation → water washing, Carnot's fixative 12-24 hours → preservation in 70% alcohol.
4. Dissociation: Constant temperature water bath, 1N hydrochloric acid or 0.5% pectinase and cellulase.5. Staining and pressing: staining solution (modified phenol magenta or magenta acetate), slides and coverslips, cut the hyphal area → absorb water → one drop of staining solution, 15 minutes → division.

6. Chromosome observation: microscope.7. Permanent seal (demonstration): xylene, neutral gum.
