Pseudocomplex ciliated columnar epithelium morphologic observation experiments

1. Staining: HE

2. Visualization by the naked eye:

The specimen is a transverse section of the trachea with a thin layer of purplish-blue tissue on the luminal surface, i.e., pseudociliated columnar epithelium. Be careful not to mistake the "C-shaped" blue cartilaginous ring in the section for epithelium.

3. Low magnification observation:

The cells of pseudo-ciliated columnar epithelium are unequal in height, so the corresponding nuclei are staggered, resembling a complex layer. The basement membrane of this kind of epithelium is more obvious. Choose the site with more cup-shaped cells and turn to high magnification observation.

4. High magnification observation:

Identify the various cells of this epithelium.

(1) Cylindrical cells: the main cells of this epithelium, numerous, with a wide free end, reaching the luminal surface, with a row of fine and neat cilia on the cell surface. The nucleus is ovoid, located in the wider part of the cell.

(2) Cup-shaped cells: fewer in number, scattered among other cells. It is shaped like a tall wine glass, the free surface reaches the luminal surface, the top of the cell is larger, stained light blue or vacuolated (due to the dissolution of the mucogenic particles); the bottom is thin and narrow, and there is a deeply colored, triangular nucleus inside.

(3) Cone-shaped cells: located at the base of the epithelial cells, the cells are cone-shaped with unclear boundaries. The nucleus is round and small.

(4) Studded cells: pointed at both ends, thicker in the middle, the nucleus is ovoid, but narrower than the nucleus of columnar cells. The cell boundaries are unclear and not easily distinguishable.
