Glycerol-3-phosphate (G3P) is oxidized by glycerol-3-phosphate oxidase (GPO) to produce hydrogen peroxide (H₂O₂). H₂O₂ then reacts with 4-aminoantipyrine and other components to form a red quinone compound, which exhibits a characteristic absorption peak at 510 nm. The activity of glycerol-3-phosphate oxidase (GPO) is determined by measuring the change in absorbance at this wavelength.
