Glucose oxidase catalyzes the oxidation of glucose to generate H₂O₂; peroxidase subsequently catalyzes the oxidation of o‑dianisidine by H₂O₂ to produce a colored product. This product exhibits a characteristic absorption peak at 500 nm. By measuring the change in absorbance at this wavelength, the activity of glucose oxidase can be quantitatively determined.
