Gelatin Adhesive, also known as Haupt’s Adhesive, is a common histological adhesive for attaching sections to glass slides. It is composed of gelatin, glycerol and phenol, and is suitable for adhering paraffin sections, frozen sections and fungal fruiting‑body sections. After sectioning, confirm the section quality by microscopic examination, then use this adhesive to firmly attach sections onto glass slides. Paraffin is then removed (this step is not required for frozen sections), followed by subsequent procedures such as staining and dehydration. Slides prepared with this reagent can be stored at 4 ℃ for six months. For research use only; not intended for clinical diagnosis or other applications.
Procedure (For reference only)
1. Gelatin adhesive tends to gelatinize at low temperature. Warm it in a water‑bath at 25‑37 ℃ until fully dissolved before use.
2. Apply gelatin adhesive onto the surface of glass slides and allow it to stand for an appropriate period of time (slides may also be coated by immersion).
3. Take out the slides and dry at 37 ℃ for 1‑2 h until visually completely dry. Drying can be accelerated by placing slides in a fume hood for several minutes. For some experiments, a drying time of 3 h or longer is required.
Precautions
1. Gelatin can be digested and absorbed by some types of cells; excessive gelatin may cause cytotoxicity.
2. When coating slides by immersion, ensure slides are fully submerged in the liquid. Incomplete coating may result in sample detachment.
3. Avoid dust contamination during the drying process.
4. For personal safety, wear a lab coat and disposable gloves during handling.
5. It is recommended to use up the reagent promptly after opening to prevent compromised experimental performance.
