Assay principle: HMGCS catalyzes the reaction of acetyl-CoA and acetoacetyl-CoA to produce hydroxymethylglutaryl-CoA along with CoASH, which converts DTNB into the yellow-colored TNB. The product exhibits a characteristic absorbance at 412 nm.
