概述
本产品专门用于冠状病毒、流感病毒、手足口病毒、麻疹风疹病毒等各种病毒及相关样本的采集、运输、保存和免提取核酸释放。它能快速灭活病毒的活性，同时还能保护病毒的核酸在常温条件下1周不降解，方便样品的运输和异地检测。
本产品具有以下特点：
1. 能即刻灭活病毒，使其快速失去活性。避免其对后续操作人员的第二次感染。
2. 含有特殊的核酸保护剂和稳定剂，保护液中的拭子样本常温下保存长达1周，方便运输和存储。
3. 可用于各种拭子的采集保存和运输，包括口腔拭子、咽喉拭子、鼻腔拭子、阴道拭子等等。
4. 用本产品保存的病毒样本无需进行核酸提取操作，可直接进行PCR、qPCR、NGS、SNP等多种操作，简单方便，节省时间。
使用方法：
注意：尽管本产品含有胍盐类病毒灭活剂，但不能排除离心管外在取样时污染了病毒，因此操作人员必须按处理传染性样品的方法进行相应的防护，必须在合乎国家规定的实验室进行所有操作。本产品具有抑菌性，但不能排除分装到采样管中时不被污染，分装前后需进行适当的灭菌处理。
1. 管内装量：1ml（一拖一），3ml（一拖五），5ml（一拖十）
2. 采样前，在含有病毒保存液的采样管的标签上标注相关样品信息。
3. 根据不同的采样要求，用采样拭子在相应的部位和器官采样。具体采样方法如下：
a) 鼻拭子：将拭子头轻轻插入鼻道内鼻腭处，停留片刻后缓慢转动退出。
b) 咽拭子：用拭子擦拭双侧咽扁桃体及咽后壁后退出。
c) 尿道采样：对男性，用拭子插入尿道约2cm处旋转，静止数秒后退出。对女性，抹去宫颈口粘液，用拭子插入宫颈管1-2cm处后退出。
d) 其他拭子采样：本手册暂不详述。
4. 迅速将拭子放入含有病毒保存液的采样管中，将拭子在样品管中旋转15秒。
5. 将采样拭子在折断处折断，丢弃把柄端，旋紧管盖。
6. 新鲜采集的标本应在4°C-25°C下内运送至实验室，可在常温条件下保存1周。
7. 用本产品保存的病毒样本无需进行常规的核酸提取操作，可以直接用做PCR、RT-PCR、qPCR、qRT-PCR、NGS、SNP扩增的模板，与市面上的核酸扩增试剂兼容。
（注：作为直接扩增模板，样本含量不得超过PCR反应总体系的10%，如若是20μL的反应体系，样本含量最多添加2μL。）
8. 未用完的部分模板可以放-20°C长期保存。
This product is specially designed for the collection, transportation, preservation and extraction-free nucleic acid release of specimens containing coronaviruses, influenza viruses, hand-foot-and-mouth disease viruses, measles and rubella viruses, and other viruses. It rapidly inactivates viral activity while protecting viral nucleic acids from degradation for up to one week at ambient temperature, facilitating sample transportation and off-site testing.
Product Features:
1. It achieves immediate viral inactivation to rapidly abolish viral activity and prevent secondary infection of subsequent operators.
2. Formulated with specialized nucleic acid protectants and stabilizers. Swab specimens in the preservation solution can be stored at ambient temperature for up to one week to simplify transportation and storage.
3. Suitable for collection, preservation and transportation of various swab specimens, including oral swabs, throat swabs, nasal swabs, vaginal swabs, etc.
4. Viral specimens preserved with this product require no nucleic acid extraction. The samples can be directly used for multiple assays such as PCR, qPCR, NGS and SNP analysis. The workflow is simple and time-saving.
Instructions for Use:
Note: Although this product contains guanidine-based viral inactivators, potential viral contamination on the outer surface of centrifuge tubes during sampling cannot be excluded. Operators must wear appropriate personal protective equipment as required for handling infectious specimens, and all operations shall be carried out in laboratories complying with national regulations. The product has bacteriostatic properties, yet contamination may still occur during aliquoting into sampling tubes. Adequate sterilization shall be performed before and after aliquoting.
1. Filling volume: 1 mL (1 sample per tube), 3 mL (pool of 5 samples), 5 mL (pool of 10 samples).
2. Label the sampling tube pre-filled with viral preservation solution with relevant sample information prior to sampling.
3. Collect specimens from corresponding anatomical sites using sampling swabs according to sampling requirements. Specific sampling procedures are described below:
a) Nasal swab: Gently insert the swab tip into the nasopharynx, hold briefly, then slowly rotate and withdraw.
b) Throat swab: Swab both palatine tonsils and posterior pharyngeal wall with the swab, then withdraw.
c) Urethral sampling: For males, insert the swab approximately 2 cm into the urethra, rotate, hold for several seconds and withdraw. For females, remove mucus from the cervical orifice, insert the swab 1–2 cm into the cervical canal and withdraw.
d) Other swab sampling: Not detailed in this manual.
4. Immediately place the swab into the sampling tube containing viral preservation solution, and rotate the swab inside the tube for 15 seconds.
5. Break the swab at the pre-scored breakpoint, discard the handle segment, and tightly screw on the tube cap.
6. Freshly collected specimens shall be transported to the laboratory at 4°C–25°C and can be stored at ambient temperature for one week.
7. Viral specimens preserved with this product do not require conventional nucleic acid extraction and can be directly used as templates for PCR, RT-PCR, qPCR, qRT-PCR, NGS and SNP amplification. It is compatible with commercially available nucleic acid amplification reagents.
(Note: When used as a direct amplification template, the sample volume must not exceed 10% of the total PCR reaction volume. For a 20 μL reaction system, the maximum sample input is 2 μL.)
8. Unused template can be stored long-term at -20°C.
