概述
Indirubin, also known as isoindigotin, is an inhibitor of cyclin-dependent kinases (CDK1/cyclin B (IC50 = 10μM), CDK2/cyclin A (IC50 = 2.2μM), CDK2/cyclin E (IC50 = 7.5μM), CDK4/cyclin D1 (IC50 = 12μM), CDK5/p35 (IC50 = 5.5μM)) and of GSK-3β (IC50 = 600nM). Studies investigating functional isoindigos show that they have the ability to disrupt cell growth and proliferation. The compound is a potent aryl hydrocarbon receptor ligand and is found in human urine and fetal bovine serum. Synthesized isoindigo-based oligothiophenes are used to study heterojunction solar cells. Shown to inhibit allergic contact dermatitis by regulating T helper mediated immune system in a DNCB-induced model. An inhibitor of GSK-3β and cyclin-dependent kinases.

Indirubin (NSC 105327) is a potent cyclin-dependent kinases and GSK-3β inhibitor with IC50 of about 5 μM and 0.6 μM.
In vitro
Indirubin is the active ingredient of Danggui Longhui Wan, a mixture of plants that is used in traditional Chinese medicine to treat chronic diseases. Indirubin inhibits CDKs activity with IC50 of 2.2 - 10 μM resulting in cell cycle arrest in the G2/M phase. Indirubin also inhibits GSK-3β with an IC50 of 0.6 μM, attenuating CDK5- and GSK-3β-mediated tau phosphorylation, a process over-active in Alzheimer disease states. It also suppresses tumor necrosis factor (TNF)-induced NF-κB activation in a dose- and time-dependent manner. Indirubin also suppresses the NF-κB activation induced by various inflammatory agents and carcinogens. Indirubin blocks the phosphorylation and degradation of IκBα through the inhibition of activation of IκBα kinase and phosphorylation and nuclear translocation of p65.
