JF635‑T5 (xHTL-JF635-T5) is an exchangeable HaloTag ligand (xHTL) built on the Janelia Fluor 635 silicon rhodamine scaffold. It binds reversibly to HT7 mutant HaloTag, unlike conventional irreversible covalent HaloTag ligands. λex/λem = 635 nm / 652 nm, highly fluorogenic and cell-permeable. Continuous dye exchange reduces photobleaching, enabling long time-lapse, sequential multi-target labeling, PAINT and MINFLUX super-resolution imaging. It can be multiplexed with other xHTLs, suitable for live/fixed-cell STED, confocal and single-molecule imaging of HaloTag fusion proteins including cytoskeletal, membrane and organellar proteins.
