Glutamate dehydrogenase (GDH) catalyzes the reaction of NH₄⁺, α-ketoglutarate, and NADH to generate glutamate and NAD⁺. NADH exhibits a characteristic absorbance peak at 340 nm. By measuring the change in absorbance at this wavelength, the content of α-ketoglutarate can be quantitatively determined.
