Lactose is decomposed into galactose and glucose by β-galactosidase. The galactose is then decomposed by a complex enzyme system containing galactose dehydrogenase, simultaneously reducing NAD⁺ to NADH. The contents of lactose and galactose are calculated respectively by measuring the increase of NADH at 340 nm.
