概述

L-丙氨酰-L-谷氨酰胺（Alanyl-glutamine，Ala-Glu），又名丙氨酰谷氨酰胺、丙谷二肽，是一种细胞培养添加剂，可直接替代细胞培养基中的L-谷氨酰胺。L-谷氨酰胺（Glutamine）是细胞培养中所必需的一种营养元素，但其在溶液中不稳定，会自发降解生成氨和焦谷氨酸，其中氨对细胞有害；而L-丙氨酰-L-谷氨酰胺在水溶液中十分的稳定，不会自发的降解。

使用说明：

1、本产品按1:100加入细胞培养基即可用于大多数细胞的培养，也可以根据细胞类型和培养基进行适当的调整，但L-丙氨酰-L-谷氨酰胺一般的终浓度为1-4mM。

2、虽然大多数细胞无须进行驯化即可用等浓度的L-丙氨酰-L-谷氨酰胺直接替代L-谷氨酰胺，但如果发现L-谷氨酰胺更换成L-丙氨酰-L-谷氨酰胺后会造成细胞生长减缓，建议在新培养条件期间的第一和第二代细胞使用3:1浓度的L-丙氨酰-L-谷氨酰胺和L-谷氨酰胺，例如，如果L-谷氨酰胺的最终浓度是4mM，则新的培养条件是3mM L-丙氨酰-L-谷氨酰胺和1mM L-谷氨酰胺的混合物。待细胞生长正常之后，再完全使用L-丙氨酰-L-谷氨酰胺进行培养。

注意事项：

1、本产品已经过滤除菌。

2、使用本产品时应注意无菌操作，避免污染。

3、本产品为200mM浓缩液，请根据需要稀释后使用。

4、肽酶的释放和二肽的消化需要一定的时间，因此细胞的延滞期会略微延长，不会造成细胞生长不良，请您放心使用。

5、本产品仅用于科研或进一步研究使用，不用于诊断和治疗。

L‑Alanyl‑L‑glutamine (Ala‑Glu), also known as alanyl‑glutamine or alanyl‑glutamine dipeptide, is a cell culture additive that can directly replace L‑glutamine in cell culture media. L‑Glutamine is an essential nutrient in cell culture, but it is unstable in solution and spontaneously degrades to form ammonia and pyrrolidone carboxylic acid, with ammonia being harmful to cells. In contrast, L‑alanyl‑L‑glutamine is very stable in aqueous solution and does not degrade spontaneously.

Instructions for Use

1.This product can be added to cell culture medium at a 1:100 ratio for the cultivation of most cell types. The amount may be adjusted appropriately according to the specific cell type and medium, but the typical final concentration of L‑alanyl‑L‑glutamine is 1‑4 mM.

2.Although most cells can directly switch from L‑glutamine to an equivalent concentration of L‑alanyl‑L‑glutamine without acclimation, if cell growth is observed to slow down after the substitution, it is recommended to use a mixture of L‑alanyl‑L‑glutamine and L‑glutamine at a 3:1 ratio during the first and second passages under the new culture conditions. For example, if the final concentration of L‑glutamine was 4 mM, the new culture condition would contain 3 mM L‑alanyl‑L‑glutamine and 1 mM L‑glutamine. Once normal cell growth resumes, full replacement with L‑alanyl‑L‑glutamine can be used.

Notes

1.This product has been filter‑sterilized.

2.Aseptic techniques should be followed when using this product to avoid contamination.

3.This product is supplied as a 200 mM stock solution. Please dilute as needed before use.

4.The release of peptidases and digestion of the dipeptide require some time, which may slightly prolong the lag phase of the cells. This does not adversely affect cell growth, and the product can be used with confidence.

5.This product is intended for research or further study only and is not for diagnostic or therapeutic use.
