摘要
Cancer treatment techniques often struggle to simultaneously address the issues of significant impact from cancer heterogeneity, poor resistance to drugs, and non-selective drug toxicity. Therefore, cancer treatment strategies based on nanoplatforms with multiple responsive properties have attracted wide attention due to their broad applicability and minimal side effects on normal cells. Herein, we constructed a nano iron-based metal–organic framework loading Pt and coated with polydopamine (Fe-MOF-NH 2 @Pt@PDA) using the solvothermal method for multimodal cancer therapy. The Pt and Fe could leverage the unique weak acidity and overexpression of hydrogen peroxide in the tumor cell to generate reactive oxygen species (ROS) for chemodynamic therapy (CDT). Additionally, Pt and Fe gave its peroxidase-like activity to produce oxygen, improving the hypoxic environment in the tumor microenvironment. Due to Pt, Fe-MOF-NH 2 @Pt@PDA achieved excellent sonodynamic therapy (SDT) under ultrasound (US) stimulation. Under US, more ROS were generated, enhancing the anticancer effect of CDT. Moreover, Fe-MOF-NH 2 @Pt@PDA produced an excellent photothermal effect under 808 nm near-infrared (NIR) irradiation for photothermal therapy (PTT). Furthermore, Fe-MOF-NH 2 @Pt@PDA exhibited good biocompatibility. Taken together, the nano Fe-MOF-NH 2 @Pt@PDA with synergistic CDT, SDT, and PTT combined treatment was a promising biomaterial for non-invasive clinical cancer therapy.
