摘要
Tumor relapse and cancerometastasis frequently occur post tumor treatments, leading to poor prognosis with high mortality. Unfortunately, therapy-exacerbated inflammation is among the chief causes of poor prognosis, especially for photothermal therapy (PTT). To overcome this mismatch between therapy and prognosis, we design a built-in Pt/polydopamine(PDA) nanoconstruction (PNP) that makes the anti-inflammation potency and therapy outcome positively inter-linked by virtue of their temperature dependency. PDA not only works as the PTT agent but also provides a self-reductive nano-substrate (∼150 nm) for the in-situ generation of ultrasmall Pt nanodots (∼4 nm). The embedding of Pt nanodots affects insignificantly the PTT potency of PDA while importing the enzyme-like ROS-scavenging function to defend inflammation in a therapy-enhanced manner. Specifically, PNP exhibits the increasing anti-inflammation effect with the elevated temperature caused by PTT, which enables the anti-inflammation pathway to match perfectly PTT modality. In vivo results indicate that PNP leads to better antitumor performance and effective inhibition of tumor relapse and cancerometastasis. This study provides new ideas for systematical tumor treatments with improved prognosis by coordinating anti-inflammation and antitumor actions harmoniously.
