摘要

When traditional T 1 and T 2 contrast agents coexist, the magnetic coupling effect leads to signal interference, which severely limits the synergistic enhancement potential of dual-mode magnetic resonance imaging (MRI). In this study, we introduced an innovative “dispersion-reaggregation” dynamic regulation nano-platform (SPMN) activated by hypoxia and acidity, which achieved T 1 -T 2 dual-signal synergistic enhancement through a two-step distance coordination mechanism. Upon hypoxia-induced dissociation of the nanocarriers, Mn 2+ (the T 1 contrast agent) and SPIO (the T 2 contrast agent) were released. This initial separation prevented signal cancelation due to the magnetic coupling effect. Through dynamic regulation of spatial distance, interference between T 1 and T 2 signals was mitigated, thereby accomplishing the first step of distance coordination. Thereafter, the acidic microenvironment facilitated the aggregation of SPIO via molybdenum (VI)-based polyoxometalate (POM)-mediated protonation and hydrogen bonding, which further increased the distance from Mn 2+ . Notably, the saturation magnetization of SPIO aggregates significantly exceeded that of individual SPIO particles. Consequently, the aggregation of dispersed SPIO enhanced the inhomogeneity of the magnetic field, thereby improving the T 2 signal and achieving the second step of distance coordination, termed “regathering”. In conclusion, SPMN demonstrated substantial dual-modality MRI enhancement and held considerable promise for MRI-guided tumor diagnosis.
