Sensitive determination of endogenous hydroxyl radical in live cell by a BODIPY based fluorescent probe

Kepeng Lei,Mingtai Sun,Libo Du,Xiaojie Zhang,Huan Yu,Suhua Wang,Tasawar Hayat,Ahmed Alsaedi

The sensitive and selective fluorescence probe for hydroxyl radical analysis is of significance because hydroxyl radical plays key roles in many physiological and pathological processes. In this work, a novel organic fluorescence molecular probe OHP for hydroxyl radical is synthesized by a two-step route. The probe employs 4-bora-3a,4a-diaza-s-indacene (difluoroboron dipyrromethene, BODIPY) as the fluorophore and possesses relatively high fluorescence quantum yields (77.14%). Hydroxyl radical can rapidly react with the probe and quench the fluorescence in a good linear relationship (R 2 =0.9967). The limit of detection is determined to be as low as 11   nM. In addition, it has been demonstrated that the probe has a good stability against pH and light illumination, low cytotoxicity and high biocompatibility. Cell culture experimental results show that the probe OHP is sensitive and selective for imaging and tracking endogenous hydroxyl radical in live cells.

A novel “Turn-On” fluorescent probe for F− detection in aqueous solution and its application in live-cell imaging

Jian Xu,Shaobo Sun,Qian Li,Ying Yue,Yingdong Li,Shijun Shao

A novel probe incorporating quaternized 4-pyridinium group into a BODIPY molecule was synthesized and studied for the selective detection of fluoride ions (F − ) in aqueous solution. The design was based on a fluoride-specific desilylation reaction and the “Turn-On” fluorescent response of probe 1 to F − was ascribed to the inhibition of photoinduced electron transfer (PET) process. The probe displayed many desired properties such as high specificity, appreciable solubility, desirable response time and low toxicity to mammalian cells. There was a good linearity between the fluorescence intensity and the concentrations of F − in the range of 0.1–1   mM with a detection limit of 0.02   mM. The sensing mechanism was confirmed by the NMR, electrospray ionization mass spectrum, optical spectroscopy and the mechanism of “Turn-On” fluorescent response was also determinated by a density functional theory (DFT) calculation using Gaussian 03 program. Moreover, the probe was successfully applied for the fluorescence imaging of F − in human epithelial lung cancer (A549) cells and alveolar type II (ATII) cells under physiological conditions.

Highly selective and sensitive polymers with fluorescent side groups for the detection of Hg2+ ion

Yiru Jia,Yi Pan,Haiying Wang,Rong Chen,Hua Wang,Xinjian Cheng

A fluorescent monomer of boron dipyrromethene (BODIPY) derivative ( 9AnPD) is designed and synthesized at first, and then it is introduced into polyurethane to obtain polymeric chemosensor with dangling fluorescent side groups. The monomer chemosensor 9AnPD exhibits highly selective properties to Hg 2+ ion in a wide pH range. The color of the monomer changed rapidly upon the addition of Hg 2+ ion. The fluorescence peak shifted and is enhanced about 4-flod. In addition, the monomer chemosensor reaches the detection limit of Hg 2+ is as low as 5 × 10 −6   m. When the fluorescent monomer is introduced to polymers, the sensing properties of the monomer chemosensor 9AnPD still remained. The polymer chemosensors not only exhibit fluorescence shift and color change for identifying Hg 2+ ion as its monomer chemosensor, but also improve the efficiency of detection. The detection limit of polymer chemosensor could reach 5 × 10 −7   m , and the fluorescence enhanced about 6-flod. The glass transition temperature (T g ) of the fluorescent PU is increased with the introduction of 9AnPD . However, it is about 60 °C, and will not lead to any adverse effect for the process. The molecular weights of PUs are lower than which without 9AnPD , but still of the macromolecular level.

Visible-Light-Driven Free Radical Polymerization and 3D Printing with Highly Efficient Carbazole-Substituted Aza-BODIPY Photoinitiators

Chenzhi Yao,Qike Sun,Hui Deng,Gongyang Chen,Heng Xia,Zhengjiang Liu,Timur Borjigin,Jiansong Yin,Jacques Lalevée,Yangyang Xu

Photoinduced radical chemistry is pivotal for synthesizing polymeric materials under light irradiation. In this study, carbazole moieties were incorporated at distinct positions within the aza-boron-dipyrromethene (aza-BODIPY) scaffold, yielding three novel push–pull dyes that function as high-performance photoinitiators for the free radical polymerization of acrylates upon exposure to visible-light LED irradiation. When combined with iodonium salts or amines as co-initiators, these aza-BODIPY derivatives form efficient two-component photoinitiating systems that exhibit outstanding initiation performance. Notably, distinct polymerization kinetics were observed for two benchmark acrylate monomers. Through a combination of experimental investigations and theoretical calculations, a comprehensive photochemical mechanism has been elucidated. Furthermore, these newly developed carbazole-substituted aza-BODIPY-based photoinitiating systems enable the fabrication of high-resolution three-dimensional macroscale architectures via 3D printing. This work advances the fundamental understanding and practical application of visible-light-induced free radical polymerization.

Single molecular-based nanoparticles with aggregation-induced emission characteristics for fluorescence imaging and efficient cancer phototherapy

Jingran Deng,Mengqian Yang,Cheng Li,Genyan Liu,Qi Sun,Xiaogang Luo,Fengshou Wu

Organic nanomaterials have garnered extensive interest in PDT/PTT due to their excellent photo-stability, efficient photothermal and reactive oxygen species generation ability. However, most organic nanomaterials emit very weak in the aggregated state because of the aggregation-caused quenching phenomenon, which limits their application in bioimaging. In this study, a donor-acceptor-donor (D-A-D) type BODIPY-based compound (BODIPY-TPA) was synthesized by connecting triphenylamine (TPA) with BODIPY structure. To further improve the biocompatibility of the compound, a polyethylene glycol chain (PEG) was introduced into the BODIPY core. BODIPY-TPA could transform into the related nanoparticles (BODIPY-TPA NPs) by the self-assembly process. Remarkably, BODIPY-TPA NPs showed strong near-infrared (NIR) fluorescence because of its aggregation-induced emission (AIE) property, which may be ascribed to the twisted structure of TPA. BODIPY-TPA NPs could simultaneously generate singlet oxygen ( 1 O 2 ) and heat under the irradiation of a single laser (635 nm). The photothermal conversion efficiency (PCE) of BODIPY-TPA NPs was calculated to be 20.7%. The subcellular localization experiment indicated the lysosome-targeting properties of BODIPY-TPA NPs in A549 cells. The cytotoxicity of BODIPY-TPA NPs was studied by CCK-8 assay and AM/PI staining on A549 cells. The half maximal inhibitory concentration (IC 50 ) of BODIY-TPA NPs was calculated to be 28.45 μg/mL. This study revealed BODIPY-TPA NPs could potentially be used in bioimaging and photodynamic/photothermal (PDT/PTT) synergistic cancer treatment.

Fluorescent macromolecular chemosensors for highly and selectively detecting of 2, 4, 6-trinitrophenol

Yuanji Xiao,Xinqian Yang,Xinjian Cheng,Shuangyu Xiong,Rong Chen

Among nitroaromatic compounds with similar structures, 2,4,6-trinitrophenol (TNP) exhibits strong biological toxicity and explosion hazard. Therefore, detection of TNP is of great importance. In this work, two novel fluorescent macromolecular chemosensors P1 and P2 were prepared using boron-dipyrromethene (BODIPY) as the fluorophore. After the preparation of small BODIPY molecule M1, another small molecule M2 with two aldehyde groups was synthesized by two consecutive Vilsmeier-Haack reactions. Then, M2 reacts with trans-cyclohexane-1,4-diamine and 1,4-phenylenediamine respectively to obtain two macromolecules P1 and P2 via Schiff Base forming reaction. The results suggested that P1 and P2 could recognize TNP only while other aromatic/nitroaromatic compounds cannot cause significant fluorescence intensity response. When TNP exist, these fluorescent macromolecular chemosensors produce significant fluorescence quenching. At the same time, because the electron-rich Schiff base and the electron-deficient TNP have the structural basis of chemical interaction, these two macromolecular sensors also have excellent sensitivity to TNP, and the excellent recognition limit of detection reach as low as 0.017 μM. High sensitivity and selectivity have been applied to test TNP in lake water samples as well.

Polymeric fluorescent probes for the detection and removal of 4-nitrophenol

Fan Fan,Jiao He,Junyu Chen,Xinjian Cheng

4-Nitrophenol (4-NP) poses a threat to the environment and human health. Developing a method to efficiently detect and separate 4-NP is of great significance. In this work, boron-dipyrromethane (BODIPY) phosphor was first prepared. Then, a small molecule fluorescent probe (BD-Ac) with an acetylene group was prepared through a Schiff base formation reaction. At the same time, the reversible addition-fragmentation chain transfer (RAFT) reagents (CTA-1, CTA-2) were prepared and polymerized with ethyl 2- (hydroxymethyl) acrylate and tert-butyl vinylcarbamate to obtain three non-fluorescent macromolecules P-01, P-02, and P-03 containing amino groups (−NH 2 ). The three polymers P-01, P-02, and P-03 were combined with the small molecule fluorescent probe (BD-Ac) through the alkyne-amine click reaction to obtain three fluorescent macromolecules P-1, P-2, and P-3. Meanwhile, through gel permeation chromatography (GPC) testing, it was confirmed that they have a good narrow distribution, with PDI values reaching 1.08, 1.05, and 1.06 respectively. Due to the existence of excited-state proton transfer (ESPT), 4-NP can quench the fluorescence of three fluorescent macromolecules. Limit of detection (LOD) reached 2.8 × 10 −7  mol/L, 3.4 × 10 −7  mol/L, and 4.8 × 10 −7  mol/L respectively. Not only can it identify 4-NP, but it can also achieve efficient separation of 4-NP. The separation efficiencies are 67.9 %, 68.9 %, and 60.3 % respectively. The as-prepared polymeric fluorescence probes might find application in the analytical and environmental sciences.

pH-Responsive Pluronic F127–Lenvatinib-Encapsulated Halogenated Boron-Dipyrromethene Nanoparticles for Combined Photodynamic Therapy and Chemotherapy of Liver Cancer

Jingjing Zong,Hao Peng,Xin Qing,Zhe Fan,Wenjing Xu,Xuanlong Du,Ruihua Shi,Yewei Zhang

Combination therapy such as photodynamic therapy (PDT)-enhanced chemotherapy is regarded as a promising strategy for cancer treatment. Boron-dipyrromethene (BODIPY), as close relatives of porphyrins, was widely used in PDT. However, poor water solubility, rapid metabolism by the body and lack of targeting limits its clinical application. Lenvatinib, as the first-line drug for molecular-targeted therapy of liver cancer, restricted its clinical application for its side effects. Herein, to achieve the synergy between PDT and chemotherapy, we synthesized two halogenated BODIPY, BDPBr2 and BDPCl2, which were prepared into self-assembly nanoparticles with lenvatinib, and were encapsulated with Pluronic F127 through the nanoprecipitation method, namely, LBPNPs (LBBr2 NPs and LBCl2 NPs). The fluorescence quantum yields of LBPNPs were 0.73 and 0.71, respectively. The calculated loading rates of lenvatinib for LBBr2 NPs and LBCl2 NPs were 11.8 and 10.2%, respectively. LBPNPs can be hydrolyzed under weakly acidic conditions (pH 5.0) to generate reactive oxygen species (ROS), and the release rate of lenvatinib reached 88.5 and 82.4%. Additionally, LBPNPs can be effectively taken up by Hep3B and Huh7 liver cancer cells, releasing halogenated BODIPY and lenvatinib in the acidic environment of tumor cells to enhance the targeting performance of chemotherapeutics. Compared with free lenvatinib and separate halogenated BODIPY, LBPNPs can inhibit tumor growth more effectively through pH-responsive chemo/photodynamic synergistic therapy and significantly promote the cascade of caspase apoptotic protease. This study shows that LBPNPs can be a promising nanotheranostic agent for synergetic chemo/photodynamic liver cancer therapy.

Engineering NK cell membrane-camouflaged copper-BODIPY nanoassembly to synergize dual-phototherapy and cuproptosis for enhanced antitumor immunity

Xuya Yu,Yushi Zhang,Zhuona Rong,Xueping Li,Lingpeng Luo,Lu Zhang,Liyu Sun,Yayue Liu,Wen Li,Lin Mei,Feng Lv,Xiaocong Pang,Qingqing Xiong

Cuproptosis, a copper-dependent form of programmed cell death, has emerged as a promising strategy to activate anti-tumor immunity through immunogenic cell death (ICD). However, its efficacy is often hindered by insufficient copper accumulation and glutathione-mediated copper chelation in tumor cells. Herein, we present an NK cell membrane-camouflaged copper‑boron dipyrrolylmethyl (BODIPY) coordinated nanoassembly to facilitate the tumor-targeted delivery of copper and integrate dual-wavelength phototherapy to potentiate cuproptosis and enhance antitumor immunity for synergistic cancer therapy. The nanoplatform enables 658 nm laser-triggered photodynamic therapy (PDT) to generate reactive oxygen species and 1064 nm laser-induced mild photothermal therapy (PTT) to produce localized hyperthermia. The combined PDT/PTT treatment not only produces direct cytotoxicity against tumor cells, but also depletes intracellular glutathione through oxidative stress, preventing copper chelation to amplify cuproptosis. This multimodal approach triggers robust ICD, driving dendritic cell maturation, macrophage polarization, tumor-infiltrating lymphocyte activation and pro-inflammatory cytokines secretion. Consequently, the immunosuppressive tumor microenvironment is remodeled, eliciting a strong and durable anti-tumor immune response that effectively inhibits tumor growth and prevents tumor recurrence and metastasis. Overall, this study proposes a novel strategy to potentiate cuproptosis-mediated ICD for synergistic cancer therapy.

Fluorinated Hypoxia-Responsive Aza-BODIPY for NIR-II FL/19F MR/PA Imaging and Phototherapy of Lung Cancer

Anfeng Li,Fang Wang,Mou Jiang,Yu Li,Xin Zhou,Zhong-Xing Jiang

Tumor hypoxia limits the efficacy of photodynamic therapy (PDT), necessitating photosensitizers with hypoxia-adaptive therapy and imaging. Here, we present a fluorinated N -oxide aza-BODIPY ( OFBD ) nanoemulsion ( OFBD-NP ) for hypoxia-responsive, multimodal imaging-guided phototherapy. OFBD generates robust singlet oxygen under normoxia for PDT, but is reduced by CYP450 enzymes in hypoxic cells to photothermal-potent FBD , enabling switchable PDT/PTT. Co-assembly with fluorinated oil enhances oxygen delivery, boosts 19 F MRI sensitivity, and promotes J -aggregation, shifting fluorescence into the NIR-II window for deep-tissue imaging. The redox conversion also activates photoacoustic signals, enabling responsive tri-modal imaging (NIR-II FLI/ 19 F MRI/PAI). OFBD-NP shows potent cytotoxicity in vitro under both normoxic and hypoxic conditions via apoptosis. In vivo, it selectively accumulates in tumors, offers high-contrast imaging, and leads to complete tumor regression in subcutaneous A549 models after laser irradiation, without systemic toxicity. This work demonstrates a smart nanoplatform that integrates deep-tissue imaging and hypoxia-triggered therapeutic switching, addressing major limitations of conventional photosensitizers in cancer imaging and phototherapy.

A BODIPY-based nanotheranostic suppresses osteosarcoma via dual photodynamic/photothermal action by disruption of YAP1/β-catenin Axis

Zhewen Deng,Mengyuan Zhang,Senqiang Zhu,Yunlong Zhang,Haitao Liao,Jiaqi Li,Peiyu Yan,Qi Zhang,Rui Liu,Bo Ma

Osteosarcoma (OS), a highly aggressive bone cancer with a dismal prognosis in the metastatic setting, urgently demands novel therapeutic modalities. While phototherapy offers a promising, minimally invasive strategy, its clinical efficacy is often hampered by the poor aqueous solubility of photosensitizers and the development of multidrug resistance. In response, we constructed the dual-modal nanoplatform BDP-NPs by leveraging a multifunctional F127/TPGS mixed micelle to encapsulate a cationic BODIPY photosensitizer, thereby creating a system engineered for enhanced tumor accumulation and to overcome multidrug resistance. Benefiting from their uniform size, colloidal stability, and hypoxia-responsive properties, the BDP-NPs demonstrate potent reactive oxygen species generation and high photothermal conversion efficiency under 525 nm laser irradiation. This dual-modal action induces severe mitochondrial dysfunction, thereby triggering apoptosis in both 3D tumor spheroids and tumor tissues. The synergistic phototherapy elicited profound mitochondrial-targeted cytotoxicity, inhibited clonogenic survival, and significantly suppressed tumor growth in human osteosarcoma models, all while maintaining a favorable biosafety profile. Mechanistic analyses revealed that this synergistic phototherapy plays a pivotal role in suppressing the YAP1/β-catenin axis, thereby reversing the epithelial-mesenchymal transition (EMT) program and consequently inhibiting OS cell proliferation, migration and invasion. This study not only establishes BDP-NPs as a potent and biocompatible nanotherapeutic for OS but also presents a groundbreaking phototherapeutic strategy to disrupt the YAP1/β-catenin axis-a key signaling network driving tumorigenesis and malignant progression. This approach provides a robust conceptual foundation for devising next-generation phototherapeutics that precisely target oncogenic signaling in aggressive cancers.

Pure-phase CsPbBr3 nanowires with higher charge transfer achieved by BODIPY as co-ligands and their efficient photocatalysis

Xiaoxia Feng,Yanli Qi,Juandi Cheng,Bo Wang,Jiacheng Liu

BODIPY-derived primary amine salts (BDPBr-nC) are innovatively adopted as co-ligands to one-pot fabricate high-purity CsPbBr 3 perovskite nanowires. • Bromine-rich BDPBr-nC interacts electrostatically with surface uncoordinated Pb 2+ to regulate nanowire growth, purify crystal phase, and passivate surface defects; short-chain BDPBr-3C further reduces steric hindrance and boosts charge transfer efficiency. • The optimized CsPbBr 3 -BDPBr-3C achieves efficient photocatalytic radical-tandem cyclization benefiting from matched energy level alignment, providing a viable surface ligand engineering strategy for high-efficiency perovskite photocatalysts. Lead halide perovskite nanowires (NWs) demonstrate considerable potential in photocatalysis due to their strong visible light absorption, tunable bandgap, and high photoconductivity. However, traditional ligands often desorb from the perovskite NW surface, generating the impure phase and abundant surface defects that severely hinder the separation and utilization of photogenerated carriers, thereby limiting catalytic efficiency. Herein, a series of BODIPY-based primary amine salts with varying carbon chain lengths (denoted as BDPBr-nC (n = 3, 4, 5, 6)) as co-ligands were employed to one-pot synthesize pure phase CsPbBr 3 NWs (CsPbBr 3 -BDPBr-nC). The enriched Br environment in BDPBr-nC enables electrostatic interaction with uncoordinated Pb 2+ on the CsPbBr 3 surface, effectively promoting the NWs growth and driving a phase transition from mixed phase to pure phase. This structural improvement facilitates efficient separation of photo-generated carriers in CsPbBr 3 NWs. Moreover, the shorter carbon chain in BDPBr-3C provides more favorable steric effects and enhanced conductivity, leading to superior defect passivation and charge transfer capabilities. Therefore, with CsPbBr 3 -BDPBr-nC as a representative catalyst, we achieved efficient photocatalytic radical-tandem cyclization of N-phenylmaleimide and N-phenylglycine in 1, 2-dichloroethane. Favorable energy level alignment among BDPBr-nC, CsPbBr 3 NWs, and the reactants promotes efficient electron transfer, resulting in excellent photocatalytic efficiency. This work highlights the promising potential of surface ligand engineering in enhancing both the stability and photocatalytic performance of perovskite nanowires.
