Bio-remediation are low-cost and environmentally friendly, but microbial enzymes tend to lose their activity when treated antibiotic contaminants due to its high toxicity. In this study, Bacillus megaterium Y-4 was demonstrated to in situ synthesize Pd 0 @cells, and detoxify oxytetracycline (OTC) through enhancing electron transport chain via Pd 0 -dependent F O F 1 -ATPase (proton pumps). Toxicity analyses showed that increasing Pd 0 loading significantly reduced acute and chronic bio-toxicity as well as ecological risk of OTC effluent, as evidenced by the promoting cellular response to oxidative stress with increasing enzymes GSH-Px and IDH activities (4.24 and 1.20-fold). Electrochemical (I-t, LSV, DPV and EDC) showed that the incorporated bio-Pd 0 mainly promoted FDH/Hase-based short chain (gradually took precedence over CoQ-based long chain), diverting OTC extracellular degradation to markedly detoxify OTC to cell. Inhibition (DCCD and TBA) assays and structural equation modelling indicated that bio-Pd 0 upregulated ATP and NADH synthesis. It also acted as transmembrane F O F 1 -ATPase proton pump, which enabled the backward of protons out of the restriction of PMF , eventually maximizing the energy utilization. This study sheds new insights into the detoxification of antibiotics by bio-nanoparticles through proton transmembrane transfer/redistribution.

Biological control agents are beneficial microorganisms that protect plants from disease. However, the effectiveness and viability of microorganisms are often influenced by severe environmental conditions. Encapsulation of microbial cells can protect them from biotic and abiotic stresses. The objectives of this work were to encapsulate Bacillus velezensis ZF481 (ZF481) with chitosan and carrageenan to prepare macrospheres and evaluate their control effect on clubroot disease. Macrospheres with different ratios of chitosan (2 % w/w)/carrageenan (0.3 % w/v) were synthesized by an ionic gelation method, and the bacterial cells of ZF481 were loaded into macrospheres. The maximum swelling ratio (634 %) and maximum survival rate (64.72 ± 2.65 %) were obtained from macrospheres with chitosan: carrageenan ratio of 1: 1. The number of bacteria released from macrospheres into sterile phosphate buffer solution (pH 7.4) reached a release balance after 48 h. The bacteria could be slowly released from macrospheres into the soil. The survival rate of the encapsulated bacteria respectively was 73.33 % and 67.31 % when stored at 4 °C and 25 °C for 80 days, respectively. The control efficiency of ZF481 macrospheres (chitosan: carrageenan, 1: 1) on clubroot disease in pak choi under greenhouse conditions and Chinese cabbage under field conditions was significantly higher than that of ZF481 culture. Moreover, the ZF481 macrospheres also enhanced the plant growth. Collectively, chitosan and carrageenan are ideal wall materials for preparing slow-release bacterial macrospheres to control clubroot. The macrospheres retard the release of B.velezensis ZF481 in soils and significantly improve its biocontrol efficiency on clubroot in pak choi and Chinese cabbage. Thus, this study paves the way for forming formulations against the clubroot disease.

The alkaline protease fromBacillus licheniformisstrain 2709 (AprE 2709) is widely used in Chinese industries but faces stability challenges under high-temperature conditions. This study employed molecular modeling and mutagenesis to identify Asn residues at positions 61, 160, and 211 as key sites affecting the stability of AprE 2709. By leveraging the additive and cooperative effects of mutations, the mutant enzyme AprE 2709 (N61G/N160G/N211G) was engineered, exhibiting enhanced thermostability and catalytic activity. The mutant demonstrated a 2.89-fold increase in half-life at 60 °C and a 1.56-fold improvement in catalytic efficiency compared to the wild-type enzyme. Structural analysis revealed that the improved thermostability was due to altered electrostatic interactions and strengthened hydrophobic contacts. Targeting Asn residues prone to deamidation presents a promising strategy for improving protein heat tolerance. These findings not only enhance our understanding of enzyme stability but also lay a foundation for future research aimed at optimizing alkaline proteases for diverse industrial applications, particularly in high-temperature processes.

Abstract Enantiopure alcohols have received much attention due to their widespread use as pharmaceutical intermediates. In the asymmetric biosynthesis of enantiopure alcohols, the excellent performance of carbonyl reductase makes it be the best choice as the biocatalysts. In this work, an alkali-tolerant carbonyl reductase ( Bs CR, encoded by yueD ) from Bacillus subtilis (strain 168) was obtained through gene mining, and successfully heterologously expressed in Escherichia coli with pET-32a. Bs CR showed excellent alkali resistance and even can keep more than 70% of its peak activity after incubation in Tris–HCl buffer at pH 9.0 for 40 h. The Michaelis constants and maximal velocity of the Bs CR to NADPH (A) and ethyl 4-chloroacetoacetate (B) are \(K_{m}^{A}\) = 5.390 × 10 −2 mmol/L, \(K_{m}^{B}\) = 1.855 mmol/L, and \(V_{max}\) = 147.3 μmol·min −1 ·mg −1 , respectively. Applying the E. coli BL21(DE3)/pET-32a-yueD to catalyze asymmetric reduction of ethyl 4-chloroacetoacetate and acetophenone, the yield of S -CHBE reached 89.9% and S -1-phenyl ethanol reached 66.7%, and e.e. of both products reached more than 99%. This work provides a novel CR for asymmetric reduction. Graphic Abstract A carbonyl reductase ( Bs CR) and its gene were identified through gene mining, and overexpressed in Escherichia coli BL21(DE3) for whole-cell biocatalytic asymmetric reduction

PurposeTo screen endophytic Bacillus producing volatile organic compounds (VOCs) with antifungal activity, and to explore their biocontrol properties toward the growth and pathogenicity of Curvularia lunata. Methods Two-sealed-base-plate assays were used to estimate the antifungal activities of Bacillus strains against C. lunata. Conjoint analysis of solid-phase microextraction gas chromatography-mass spectrometry and antagonistic experiments were used to identify the VOCs responsible for the antifungal activity. Effects of individual synthetic VOCs were analyzed along with reactive oxygen species (ROS) accumulation in C. lunata conidia. After exposure to individual VOCs, conidia were also sprayed onto maize leaves to evaluate their pathogenicity. Expression levels of virulence-related genes in C. lunata mycelium following exposure to VOCs were analyzed using quantitative real-time PCR.Results Among the ten endophytic Bacillus strains and two plant growth-promoting rhizobacterial (PGPR) strains, only B. subtilis strain DZSY21 strongly inhibited the growth of C. lunata by producing VOCs. 2-Methylbutyric acid, 2-heptanone, and isopentyl acetate produced by strain DZSY21 showed inhibitory effects on the mycelia growth and conidial sporulation of C. lunata. 2-Heptanone and isopentyl acetate also repressed the germination of conidia and the expression levels of virulence-related genes in C. lunata mycelium. Moreover, isopentyl acetate strongly enhanced the accumulation of intracellular ROS in conidia. The disease indexes of maize leaves sprayed with VOC-treated C. lunata conidia were reduced from 60.52 to 26.64%.Conclusion Endophytic B. subtilis strain DZSY21 displayed the potential to control C. lunata by producing VOCs, especially 2-heptanone and isopentyl acetate.

The bacterium, Bacillus amyloliquefaciens Pc3, was previously isolated from Antarctic seawater and has been found to show antagonistic activity against the fungus, Rhizoctonia solani ACCC 36316, which causes a severe disease known as Sclerotinia sclerotiorum in rapeseed plants. Bacillus lipopeptides had been widely used as biocontrol agents for plant diseases. In this study, we isolated 11 lipopeptide compounds from B. amyloliquefaciens Pc3 culture broth via reversed-phase high-performance liquid chromatography (RP-HPLC) and used matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF-MS) to identify these as iturin A (C 14 , C 15 , C 16 , C 17 ), fengycin B (C 14 , C 15 , C 16 , C 17 ), and surfactin (C 14 , C 15 , C 16 ). We further found that the addition of exogenous alkanoic acids, including myristic acid, pentadecanoic acid, palmitic acid, heptadecanoic acid, octadecanoic acid, and nonadecanoic acid, to the bacterial growth media could promote lipopeptide production and enhance the antifungal activities of crude lipopeptide extracts from B. amyloliquefaciens Pc3. In addition, the transcriptional levels of three lipopeptide synthesis genes, ituD , fenA , and srfA-A , and two fatty acid metabolism–related genes, FabI , which encodes enoyl-ACP reductase, and FadB , which encodes enoyl-CoA hydratase, were found to be upregulated in cells grown with exogenous alkanoic acids. Among the six alkanoic acids tested, those with odd carbon chain lengths had a greater effect on lipopeptide production, antifungal activity, and target gene upregulation than those with even carbon chain lengths. These results provide a practical approach for the efficient production of lipopeptides in Bacillus amyloliquefaciens Pc3.

A phenolic-UDP-glycosyltransferase Bs-PUGT from Bacillus subtilis PI18 was cloned and expressed in Escherichia coli BL21 (DE3). The purified Bs-PUGT could catalyze the glycosylation of tyrosol, 4-hydroxybenzyl alcohol, 2-hydroxybenzyl alcohol, caffeic acid, cinnamic alcohol, ferulic acid, and so on. This enzyme showed a high activity and stability over a broad pH range and was sensitive to temperature. Studies on the kinetic parameters indicated that the affinity of Bs-PUGT to UDP-G ( K m) and its catalytic efficiency ( K cat) increased by 1.5-fold and 1.7-fold, respectively, with the addition of 10 mM Ca 2+ . The most effective glycosylation of caffeic acid catalyzed by whole-cell E. coli /Bs-PUGT was achieved with a molar yield of 78.3% in a system with pH 8.0, 30 °C, 25 g/L sucrose, 10 mM Ca 2+ , and 0.5 g/L substrate concentration. The addition of Ca 2+ increased the molar yield of caffeic acid glucosides and shortened the reaction. This work proposes a strategy for the efficient glycosylation of phenolic compounds by microbe-derived glycosyltransferase assisted by metal ions.

Optimized postbiotics achieved 88.5% AFB 1 degradation with Cu 2+ or Fe 2+ . • Postbiotics attack furan and lactone rings of AFB1 to yield C 17 H 12 O 5 and C 16 H 12 O 5 . • Postbiotics reduced AFB 1 in peanut meal to below safety standard. • Postbiotics alleviated hepatic injury and inflammation in mice. • Postbiotics promoted the colonization of beneficial gut microbials. Probiotics and postbiotics offer a promising approach to decontaminate widely occurring aflatoxin B 1 (AFB 1 ). This study identified and optimized the postbiotics from Bacillus velezensis 906, systematically exploring the degradation products and pathways and evaluating the degradation efficacy and in vivo safety. The optimized postbiotics degraded 77.6% of AFB 1 in vitro , which increased to 88.5% with Cu 2+ supplementation. The degradation process involves lactone ring cleavage and furan ring modification to yield C 17 H 12 O 5 and C 16 H 12 O 5 . In moldy peanut meal, treatment merely with postbiotics and probiotics inhibited fungal growth and reduced AFB 1 from 100.86 μg/kg to 37.54 μg/kg and 45.81 μg/kg, while preserving nutritional quality. Oral gavage of moldy peanut meal treated with postbiotics significantly reduced serum alanine aminotransferase (ALT) by 29%, malondialdehyde (MDA) by 43.60% and interleukin-1β (IL-1β) by 59.8%, and normalized liver pathology and the gut microbiota ratio. In summary, B. velezensis 906 can efficiently degrade AFB 1 , ensuring the safety of moldy peanut meal for consumption while preserving its quality and nutritional value, providing an effective and safe strategy for the mycotoxin bioremediation in food and feed matrices. Bacteria-Fungi Interactions play a crucial role in soil nutrient cycling and plant disease suppression. Bacillus and Trichoderma exhibit antagonism when inoculated on laboratory media, global soil sample analysis reveals a positive correlation between these two genera in addition to enhanced plant-pathogen Fusarium oxysporum suppression and plant growth promotion. Here, we assess cross-kingdom interactions within artificial model communities of Bacillus velezensis and Trichoderma guizhouense. Transcriptomic profiling revealed that in the presence of fungi, the key stress sigma factor of B. velezensis activates expression of biosynthetic genes for antimicrobial secondary metabolite production. Among these, surfactin induces T22azaphilone production in T. guizhouense that hinders oxidative stress. Both surfactin and T22azaphilone contribute to Bacillus and Trichoderma maintenance in soil in the presence of F. oxysporum. Finally, F. oxysporum-secreted fusaric acid temporarily inhibits B. velezensis growth whereas it is efficiently degraded by T. guizhouense. These metabolite-mediated interactions reveal how competing soil microorganisms could form effective alliances that ultimately enhance plant protection against soil-borne pathogens.

Beneficial plant-associated microbes such as Bacillus species have important potential uses as bioinoculants that reduce dependence on agricultural chemicals. These microbes produce diverse metabolites that enhance plant resilience, but the functions of these compounds and the associated plant hormone pathways remain unclear. Using Bacillus velezensis ES-120, a novel strain isolated from Chenopodium album L., we demonstrate that distinct classes of ES-120 metabolites play different roles. Using Arabidopsis thaliana , we show that non-volatile soluble metabolites produced by ES-120, including lipopeptides identified by reversed-phase high-performance liquid chromatography, exhibit biocontrol activity by inhibiting multiple soil-borne pathogens. In contrast, volatile organic compounds (VOCs) emitted by ES-120 promote plant growth, likely by modulating auxin and brassinosteroid (BR) signalling pathways, with effects sustained across diverse conditions. Headspace solid-phase microextraction coupled with gas chromatography–mass spectrometry identified multiple VOCs produced by ES-120. Individual compounds showed limited activity, suggesting that growth promotion may depend on interactions among multiple VOCs or on an undetected low-abundance compound. In greenhouse trials, ES-120 reduced wilt in tomato ( Solanum lycopersicum ) and increased the biomass of Chinese cabbage ( Brassica rapa var. chinensis ). Thus, B. velezensis ES-120 functions as a dual-purpose bioinoculant: its soluble metabolites suppress soil-borne pathogens and its VOCs enhance plant growth via auxin and BR pathways, highlighting its potential for sustainable agriculture.

Introduction: Surfactins, a class of lipopeptide biosurfactants secreted by plant growth-promoting rhizobacteria (PGPR), have garnered significant attention due to their dual functionality in promoting plant growth and controlling plant diseases. Their potential as biopesticides is underscored by their unique physicochemical properties and biological activities. However, the practical application of surfactin is currently limited by its low yield in natural strains. Methods: This study aimed to optimize the culture conditions for Bacillus velezensis BN, a strain with exceptional biocontrol properties, to enhance its surfactin yield. Critical factors, including nitrogen sources and amino acid supplementation, were systematically investigated to determine their impact on surfactin production. Results: The study revealed that nitrogen sources and amino acid supplementation were pivotal factors influencing surfactin yield. Compared to the baseline, these factors resulted in a remarkable 5.94-fold increase in surfactin production. Furthermore, a positive correlation was established between surfactin yield and biocontrol efficacy. Enhanced surfactin yield was associated with improved antifungal activity, biofilm formation, and rhizosphere colonization capacity of B. velezensis BN on potato plantlets.Discussion: These findings provide novel insights into the practical application of surfactin and establish a scientific foundation for the development of innovative and eco-friendly antifungal agents suitable for agricultural use. The results demonstrate that optimizing culture conditions can significantly enhance surfactin yield and biocontrol efficacy, thereby highlighting the potential for sustainable agricultural practices.

Phospholipase D (PLD) is a valuable enzyme in industrial processes for converting phosphatidylcholine (PC) to phosphatidylserine (PS). In this study, a strain of Bacillus cereus was isolated from soil and identified through 16S rRNA sequencing. To enhance PLD activity, various mutagenesis strategies—including chemical treatment, irradiation, and their combinations—were employed, resulting in four high-activity positive mutants (C-7, I-12, CI 7–12, and IC 13–14). Among these, the CI 7–12 mutant exhibited a significantly higher enzymatic activity, showing a 3.12-fold increase (312.2%) compared to the wild-type strain. Fermentation conditions were optimied using response surface methodology (RSM), achieving a PLD activity of 35 U/mL. The enzyme demonstrated stability over a wide temperature range (30–60 °C) and pH range (6–10), with a half-life of 128 days. Kinetic analysis revealed a V max of 20.04 μmol/h and a K m of 7.13 μmol/mL, indicating efficient activity. In bioconversion experiments, the PLD-enriched fermentation broth catalyzed the conversion of PC to PS, achieving a 53.0% conversion rate and a 92.3% selectivity for PS in a two-phase system. These findings expand the potential sources of PLD and underscore its applicability for PS production in biotechnological applications.
