Effect of Dibutyl Phthalate on the Tolerance and Lipid Accumulation in the Green Microalgae Chlorella vulgaris

Duan Kaili,Cui Meng,Wu Yanni,Huang Xueyong,Xue Ahui,Deng Xunan,Luo Liping

In the present study, Chlorella vulgaris were cultured in the presence of the common plasticizer dibutyl phthalate (DBP) with different concentrations for 10 days. The cell density, DBP concentrations, neutral lipid concentrations, and lipid morphology in C. vulgaris were studied using optical microscopy, gas chromatography (GC), fluorescence spectrophotometry, and laser scanning confocal microscopy (LSCM). We observed that the neutral lipid contents and cell density of C. vulgaris were negatively influenced by DBP of high concentrations (50 and 100 mg/L), but significantly stimulated by DBP of low concentrations (5, 10, and 20 mg/L). Lipid bodies were destroyed into pieces by DBP of high concentrations (50 and 100 mg/L), but were slightly suppressed by DBP at low concentrations (5, 10, and 20 mg/L). Chlorella vulgaris treated with DBP (50 mg/L) for 2 days showed the highest removal efficiency (31.69%). The results suggested that C. vulgaris could be used in practice to remove DBP and has the potential of being oleaginous microalgae in DBP contaminated water.

Surface-engineered chlorella alleviated hypoxic tumor microenvironment for enhanced chemotherapy and immunotherapy of first-line drugs

Cheng Gao,Cheryl H.T. Kwong,Qingfu Wang,Hiotong Kam,Jianwen Wei,Qian Chen,Jian Zhang,Simon M.Y. Lee,Dayong Gu,Ruibing Wang

A large number of chemotherapeutic and immunotherapeutic agents have been shown to be less effective when exposed to a hypoxic solid tumor. The tumor hypoxia microenvironment contributes to multidrug resistance and immunosuppression, and current delivery of oxygen or enzyme to alleviate hypoxic microenvironment is often limited by the oxygen supplying capacity of material-based carriers and short window of oxygen production. Herein, macrophage membrane coated chlorella (M-Chl) is constructed for targeted delivery to the solid tumor and sustainable oxygen production via photosynthesis, which provides a new general strategy to overcome tumor hypoxia and improve chemotherapy and immunotherapy. The camouflaged strategy via macrophage membrane coating enhances tumor accumulation and retention of Chl due to the inflammatory homing effects of macrophage membrane, which contributes to long term oxygen production for at least 6 days with one dose of Chl, resulting in efficient downregulation of tumor HIF-1α. In vivo antitumor therapy in mice shows that M-Chl enhances the chemotherapeutic efficacy of doxorubicin via inhibiting hypoxia-mediated overexpression of drug efflux proteins, and also improves the poor immunotherapeutic performance of T cell activation agent, anti-CTLA-4 antibody, via downregulating hypoxia-mediated immunosuppressive proteins. Thus, M-Chl may serve as an important adjuvant for enhancing the therapeutic efficacy of clinical antitumor drugs against solid tumors.

Mercury Reduction, Uptake, and Species Transformation by Freshwater Alga Chlorella vulgaris under Sunlit and Dark Conditions

Xujun Liang,Nali Zhu,Alexander Johs,Hongmei Chen,Dale A. Pelletier,Lijie Zhang,Xixiang Yin,Yuxi Gao,Jiating Zhao,Baohua Gu

As a major entry point of mercury (Hg) to aquatic food webs, algae play an important role in taking up and transforming Hg species in aquatic ecosystems. However, little is known how and to what extent Hg reduction, uptake, and species transformations are mediated by algal cells and their exudates, algal organic matter (AOM), under either sunlit or dark conditions. Here, using Chlorella vulgaris (CV) as one of the most prevalent freshwater model algal species, we show that solar irradiation could enhance the reduction of mercuric Hg(II) to elemental Hg(0) by both CV cells and AOM. AOM reduced more Hg(II) than algal cells themselves due to cell surface adsorption and uptake of Hg(II) inside the cells under solar irradiation. Synchrotron radiation X-ray absorption near-edge spectroscopy (SR-XANES) analyses indicate that sunlight facilitated the transformation of Hg to less bioavailable species, such as β-HgS and Hg-phytochelatins, compared to Hg(Cysteine)2-like species formed in algal cells in the dark. These findings highlight important functional roles and potential mechanisms of algae in Hg reduction and immobilization under varying lighting conditions and how these processes may modulate Hg cycling and bioavailability in the aquatic environment.

Antioxidative Effect of Chlorella Pyrenoidosa Protein Hydrolysates and Their Application in Krill Oil-in-Water Emulsions

Yujia Liu,Yuli Qi,Qi Wang,Fawen Yin,Honglei Zhan,Han Wang,Bingnan Liu,Yoshimasa Nakamura,Jihui Wang

Chlorella pyrenoidosais an excellent source of protein, and in this research, we assessed the antioxidant and emulsifying effects of Chlorella protein hydrolysate (CPH) using neutral proteases and alkaline proteases, as well as the properties of CPH-derived krill oil-in-water (O/W) emulsions. The CPHs exhibited the ability to scavenge several kinds of free radicals, including 1,1-diphenyl-2-picrylhydrazyl (DPPH), O2−, hydroxyl, and ABTS. Additionally, the CPHs (5 mg/mL) scavenged approximately 100% of the DPPH and ABTS. The CPHs showed similar emulsifying activities to Tween 20 and excellent foaming activities (max FS 74%), which helped to stabilize the krill oil-in-water emulsion. Less than 10 mg/mL CPHs was able to form fresh krill oil-in-water emulsions; moreover, the CPHs (5 mg/mL) in a krill O/W emulsion were homogenous, opaque, and stable for at least 30 days. Based on their inhibitory effects on the peroxide value (POV) and thiobarbituric acid reactive substances (TRABS), the CPHs were found to be able to inhibit lipid oxidation in both emulsifying systems and krill O/W emulsions. Thus, the CPHs could improve superoxide dismutase (SOD) activities by 5- or 10-fold and decrease the high reactive oxygen species (ROS) level caused by the addition of H2O2in vitro. In conclusion, health-promoting CPHs could be applied in krill oil-in-water emulsions as both emulsifiers and antioxidants, which could help to improve the oxidative and physical stability of emulsions. Keywords:Chlorella pyrenoidosaprotein hydrolysate;antioxidant activity;krill oil-in-water emulsion;lipid oxidation

Effects of environment-relevant concentrations of antibiotics on seawater Chlorella sp. biofilm in artificial mariculture effluent

Chong Yu,Chi Li,Ying Zhang,Xiang Du,Jing-Han Wang,Zhan-You Chi,Qian Zhang

Attached cultivation of microalgae using mariculture effluent could simultaneously realize cost-effective biomass production and effluent polishing, and the obtained microalgal biofilm could be readily applied to aquatic animals that feed directly on biofilms. However, the widespread existence of antibiotics in mariculture effluent may have inhibitory effects on microalgae. Existing studies regarding the influences of antibiotics on microalgae were overwhelmingly mostly conducted in suspended freshwater cultures, and the explored antibiotic concentrations were several orders of magnitudes higher than realist effluent, rendering limited guidance for practical applications. This study, therefore, studied three commonly observed antibiotics in mariculture effluent, i.e. sulfamethoxazole (SMX), tetracycline (TL), and clarithromycin (CLA), and explored their effects on attached seawater Chlorella sp. growth, nutrients removal, and antioxidative properties at environment-relevant concentrations (0.1–200 μg/L). Hormesis was shared among the three antibiotics, and highest biofilm density (14.6 g/m 2 ) was observed under 0.1 μg/L SMX. SMX displayed strongest chlorophyll impairment, while SMX, TL, and CLA respectively favored lipid (up to 42.4 %), protein (up to 39.6 %), and polysaccharides (up to 65.4 %) accumulation. Highest correlation between biofilm density and extracellular polymeric substances (EPS) contents as well as highest total nitrogen (TN) removal were observed under TL exposure, while total phosphorus (TP) removal among all groups met the strictest local discharge standard for mariculture effluent (TP < 0.2 mg/L). Antioxidative responses revealed least stress towards attached seawater Chlorella sp. under TL treatment, while best acclimation under SMX exposure. Potential coping mechanisms of this microalgae under each antibiotic exposure were proposed as well. This study expands the knowledge of microalgae biofilm under environment-relevant concentrations of antibiotics.

Combined toxic effects of TiO2 nanoparticles and organochlorines on Chlorella pyrenoidosa in karst area natural waters

Jun Zhang,Xujiao Xie,Qing Li,Ji Wang,Shuai Zhang

With the discharge of nanoparticles (NPs) into the environment, NPs can interact with coexisting organic pollutants, resulting in combined toxic effects. In order to more realistically evaluate the potential toxic effects of NPs and coexisting pollutants on aquatic organisms. We evaluated the combined toxicities of TiO 2 nanoparticles (TiO 2 NPs) and three different organochlorines(OCs)-pentachlorobenzene (PeCB), 3,3′,4,4′-tetrachlorobiphenyl (PCB-77) and atrazine to algae ( Chlorella pyrenoidosa ) in three karst natural waters. The results indicate that the individual toxicities of TiO 2 NPs and OCs in natural waters were less than those of OECD medium, and the combined toxicities were different from but generally similar to those of OECD medium. The individual and combined toxicities were the largest in UW. The correlation analysis showed that the toxicities of TiO 2 NPs and OCs were mainly related to TOC, ionic strength, Ca 2+ and Mg 2+ in natural water. The binary combined toxicities of PeCB and atrazine with TiO 2 NPs to algae were synergistic. The binary combined toxicity of TiO 2 NPs and PCB-77 to algae was antagonistic. The presence of TiO 2 NPs increased the algae-accumulations of OCs. PeCB and atrazine all increased the algae-accumulations of TiO 2 NPs, while PCB-77 showed the opposite result. The above results indicated that due to the influence of different hydrochemical properties in karst natural waters, there were differences between TiO 2 NPs and OCs in their toxic effects, structural and functional damage, and bioaccumulation.

Glycerolipid remodeling is crucial for acclimation to naphthenic acid exposure in Chlorella pyrenoidosa and Chlamydomonas reinhardtii

Miao Yang,Jingyi Ma,Sihui Yu,Kunpeng Xie,Weidong Zhang,Endong Zhang,Qingzhi Wang,Xi Xie,Zheng Gong

Naphthenic acids (NAs) serve as natural components of petroleum, and have turned into essential pollutants in wastewater discharged from various petroleum industries . Some NAs could elicit toxicity towards multiple organisms. Microalgae from culture collections and the indigenous algal strains have been tested for their capacity to tolerate and biodegrade NAs. It is necessary to comprehensively assess the physiological response of microalgae to NA exposure, especially the glycerolipid metabolism. In this study, the representative algal strain for wastewater treatment , Chlorella pyrenoidosa and the model green alga , Chlamydomonas reinhardtii , were used to evaluate their responses to a model NA, cyclohexanecarboxylic acid (CHCA), involving growth, photosynthetic activity , cell morphology and glycerolipid metabolism. The results showed that, C. pyrenoidosa cells were not affected in growth performance and cell structure during 9 days of CHCA exposure. However, the growth of C. reinhardtii was retarded, and the surface of the enlarged cells got damaged within 3 days of CHCA treatment. Their photosynthetic activity both remained stable when subjected to CHCA stress. More importantly, the fatty acyl profiles of total lipids, membrane lipids and neutral lipids were all remodeled and C18:3n3 in the two algae serves as the potential biomarker in response to CHCA treatment. Thus, C. pyrenoidosa presented more superior tolerance to CHCA exposure relative to C. reinhardtii , and they both reconstructed the glycerolipid components for acclimation to CHCA stress. Overall, glycerolipid remodeling is crucial for green microalgae to accommodate the NAs-contaminated environment. This study offers valuable insights into the potential tolerance and physiological response of green algae to NA stress, especially in glycerolipid remodeling, which will benefit improved monitoring of NAs pollution in aquatic ecosystems and effective treatment of NA-contaminated wastewater.

Different responses of Chlorella vulgaris to silver nanoparticles and silver ions under modulation of nitric oxide

Zhou Gaoxiang,Xu Limei,Wang Haoyu,Sun Aoxue,Wang Yong,Li Xiaochen,Jiang Ruixue

Silver nanoparticles (Ag-NPs) are widely used in daily life because of their antibacterial properties. A fraction of Ag-NPs are released into the ecosystem during their production and utilization. The toxicity of Ag-NPs has been reported. However, it is still disputed whether the toxicity is mainly due to the released silver ions (Ag + ). In addition, few studies have reported the response of algae to metal nanoparticles under modulation of nitric oxide (NO). In this study, Chlorella vulgaris ( C. vulgaris ) was used as a model organism to study the toxic effects of Ag-NPs and Ag + released from Ag-NPs on algae under the modulation of NO. The results showed that the biomass inhibition rate of Ag-NPs (44.84%) to C. vulgaris was higher than that of Ag + (7.84%). Compared with Ag + , Ag-NPs induced more severe damage to photosynthetic pigments, photosynthetic system II (PSII) performance, and lipid peroxidation. More serious damage to cell permeability led to higher internalization of Ag under Ag-NPs stress. Application of exogenous NO reduced the inhibition ratio of photosynthetic pigments and chlorophyll autofluorescence. Further, NO reduced the MDA levels by scavenging reactive oxygen species induced by Ag-NPs. NO modulated the secretion of extracellular polymers and hampered the internalization of Ag. All these results showed that NO alleviates the toxicity of Ag-NPs to C. vulgari s. However, NO did not improve the toxic effects of Ag + . Our results provide new insights into the toxicity mechanism of Ag-NPs to algae modulated by the signal molecule NO.

Glucose as a mitigator of cadmium-induced toxicity in chlorella sp.: Insights from cell division and photosynthesis

Bocheng Su,Jianke Huang,Enci Ye,Xiwen Xue,Jindong Zhao,Zi Ye

Glucose increased Chlorella biomass by counteracting Cd inhibition. • Cd 2+ disrupts cell wall degradation and photosynthetic systems. • Glucose co-treatment restored the cell wall degradation and chlorophyll homeostasis. Cadmium (Cd) pollution is a growing environmental challenge worldwide, impairing cellular integrity and transcriptional regulation across species and undermining ecosystem stability. This study investigates the effects of Cd 2 + stress on Chlorella pyrenoidosa in terms of growth, cell division, and photosynthetic function, and evaluates the alleviating role of soluble organic carbon supplementation, using glucose as a representative source. Results indicate that Cd 2+ exposure markedly suppresses algal growth, disrupts photosynthetic systems, reduces chlorophyll content, diminishes key photosynthetic metrics, and interferes with cell wall degradation. Biomass and specific growth rate declined from 0.6 g L −1 and 0.49 d −1 in controls to 0.38 g L −1 and 0.42 d −1 under Cd 2+ stress, but recovered to 0.95 g L −1 and 0.61 d −1 with glucose addition. Glucose supplementation substantially counteracted these inhibitory effects, partially reviving photosynthetic activity through enhanced QA-to-QB electron transfer and stabilization of porphyrin metabolism. Transcriptomic profiling revealed that Cd 2+ downregulated genes involved in cellulose degradation while upregulating those associated with lignin biosynthesis; glucose treatment restored the expression of lignin-related genes to near-control levels. Collectively, our findings demonstrate that glucose alleviates Cd 2+ toxicity in C. pyrenoidosa by restoring photosynthetic competence and cell division capacity, thereby elucidating key metabolic pathways underlying heavy-metal detoxification. Enhancing removal performance of ciprofloxacin hydrochloride by Chlorella vulgaris-bacteria consortium: Insights of microbial responses and stoichiometry

Yongrui Pi,Yan Li,Wenpeng Jia,Yongzheng Tang

The microalgae-bacteria consortia (MBC) system has demonstrated significant potential in enhancing the removal efficiency of antibiotics from wastewater. This study systematically evaluated the treatment of ciprofloxacin hydrochloride (CIP) wastewater using pure microalgae ( Chlorella vulgaris ) (PM), pure bacteria (PB), and the MBC formed by the combination of PM and PB. The MBC system achieved an impressive CIP removal rate of 94.6 ± 0.6 % at an initial concentration of 10 mg/L. As the initial concentration of CIP increased from 10 to 80 mg/L, the biodegradation rate declined from 0.42 to 0.13 d - ¹ , with the half-life of CIP significantly increasing from 1.66 to 5.23 days. The MBC notably enhanced CIP removal efficiency by promoting microbial metabolic activity and increasing the concentration of photosynthetic pigments. The secretion of CIP-degrading enzymes to mitigate intracellular oxidative stress indicates the superior tolerance and degradation capacity of the MBC system towards CIP. Acinetobacter johnsonii was identified as the primary contributor to the CIP biodegradation. The molar yield coefficient (MYC) for the bacteria is nearly 2-times fold of that for C. vulgaris based on the growth-process equations of cells on CIP. Collectively, these findings provide valuable insights into the mechanisms by which MBC outperforms PM or PB in the treatment of CIP-contaminated wastewater, thereby contributing to advancements in sustainable wastewater management practices.

Molecular insights into how organophosphate esters regulate conventional contaminants removal by Chlorella sorokiniana for synthetic wastewater treatment

Di Zhou,Liandong Zhu

Organophosphate esters (OPEs) are typical types of emerging contaminants that are widely presented in wastewater. While they can serve as phosphorus (P) sources for microalgae growth, their toxicity disrupts metabolic processes, posing challenges to microalgae-based bioremediation. This study elucidated the regulatory effects of OPEs on the removal of ammonia (NH 4 + -N) and phosphate (PO 4 3- -P) from synthetic wastewater by Chlorella sorokiniana , as well as the concurrent OPEs degradation, while unveiling the molecular mechanism of these interactions. OPEs exposure (1 and 10 mg/L) initially inhibited nutrient assimilation, reducing NH₄⁺-N and PO₄³⁻-P removal by 21.78 %-46.59 % and 7.46 %-26.07 %, respectively. The suppression was primarily attributed to oxidative stress-induced DNA damage, leading to G2/S cell cycle arrest, and metabolic disruptions particularly in crucial nitrogen assimilation enzymes (GS, GDH) and genes ( IMPL2, IPPK ) related to Pi supply strategy adjustment. Meanwhile, OPEs continuously impaired heterotrophy (NaAc matabolism), restricting energy supply for nutrient uptake. Despite this initial inhibition, microalgae exhibited adaptive recovery through photosynthetic regulation and EPS secretion. Enhanced linear electron transfer (LET) and Calvin cycle facilitated photoautotrophy, while EPS played a crucial role in OPEs adsorption and degradation via photosensitization and biosorption, achieving 41.53 %-54.50 % OPEs removal without additional energy input. Transcriptome analysis, combined with KEGG-based pathway annotation, was performed to elucidate the molecular mechanisms underlying these physiological responses. This study unveils a novel stress adaptation mechanism in microalgae, demonstrating their ability to mitigate OPEs toxicity and restore nutrient removal efficiency. The findings provide new insights into the synergistic bioremediation of emerging and conventional pollutants, offering a sustainable strategy for low-carbon wastewater treatment.

UV254 triggers chloroacetic acids degradation to generate degrading products for Chlorella sorokiniana growth: mechanism and performance evaluation

Wei Wang,Qirui Wu,Hanzhi Wang,Mingxiang Qv,Dian Dai,Xiaoman He,Ivan P. Pozdnyakov,Li-Zhi Huang,Liandong Zhu

This study investigated the UV 254 irradiation degradation of chloroacetic acids (CAAs), specifically monochloroacetic acid (MCAA), dichloroacetic acid (DCAA), and trichloroacetic acid (TCAA), and evaluated the ecological impact of their degradation products on Chlorella sorokiniana (CS). The degradation followed first-order kinetics, with dechlorination efficiencies of 91.2% for MCAA, 92.9% for DCAA, and 92.4% for TCAA. TCAA was degraded fastest due to its higher chlorine content. Initial pH (3–10) had minimal influence, whereas lower concentrations (0.25   mM) led to slower degradation. Hydroxyl radicals (•OH) were identified as the dominant reactive species via electron paramagnetic resonance analysis. Dechlorination generated Cl⁻ and HCl, resulting in acidification and increased toxicity. Total organic carbon (TOC) analysis indicated nearly complete mineralization. Untreated CAAs significantly inhibited microalgal growth and photosynthesis, while post-degradation pH adjustment (to pH > 4) mitigated toxicity and enabled microalgal assimilation, with CS culture pH further increasing to 9–10, thereby facilitating self-buffered bioremediation. These findings confirm the effectiveness of UV 254 degradation on CAAs and provide valuable guidance for ecological safety in aquatic ecosystem management.
