Triple-negative breast cancer (TNBC) presents significant therapeutic challenges due to its aggressive nature and lack of molecular targets. To address this challenge, we developed a pH/thermo-responsive nanoplatform based on epigallocatechin-3-gallate (EGCG) modified lipid-coated polydopamine (TEDP), capable of loading doxorubicin (DOX). EGCG, a bioactive polyphenol with high affinity for laminin receptors overexpressed in cancer cells, served both as a targeting ligand and immune microenvironment remodeling regent, enabling selective uptake and remodeling immunosuppression cells. Thermosensitive EGCG modified DOX loaded PDA (TEDP) nanoparticles exhibited spherical morphology (∼140 nm), good colloidal stability, and efficient photothermal conversion under near-infrared irradiation. Under a trigger temperature and pH 6.0, TEDP exhibited pH/temperature-triggered drug release, achieving 70% precise drug release within 4 h, thereby enabling spatiotemporally controlled drug release. Both in vitro and in vivo experiments demonstrated that TEDP nanoparticles could significantly enhance targeting, ensuring precise drug release at the tumor site while minimizing off-target toxicity of DOX. In a TNBC tumor model, the tumor suppression efficacy of TEDP treatment exceeded that of chemotherapy or photothermal therapy (PTT) monotherapy by 3.2-fold and 1.7-fold, respectively. The findings highlighted that TEDP nanoparticles generated synergistic enhancement effects through chemotherapy, PTT and anti-tumor immune responses activating. The multimodal nanoplatform would hold great promise in TNBC therapy, offering a strategy to overcome the limitations of conventional monotherapies.

The synthesis of Selenium nanoparticles (Se NPs) using plant extracts has been widely recognized for many advantages. In this study, Epigallocatechin gallate (EGCG) was used as the reducing agent for sodium selenite to obtain Epigallocatechin gallate-Selenium nanoparticles (EGCG-Se NPs) whose hypoglycemic potential was evaluated. The Ultraviolet-visible(UV-Vis) spectra, Fourier Transform Infrared Spectroscopy (FTIR) profiles, and X-ray Photoelectron Spectroscopy (XPS) analysis of EGCG-Se NPs all confirmed the Se-O coordination bonds between Se NPs and EGCG, which is conducive to improving the stability of the molecular structure of the complex. The ellipsoidal structure (20–90 nm) and amorphous state of EGCG-Se NPs were characterized via X-ray Diffraction (XRD) and Scanning Electron Microscopy (SEM). Dynamic light scattering (DLS) and zeta potential analyses confirm that EGCG-SeNPs exhibit sustained colloidal stability under physiologically relevant buffer conditions (pH 7.2–7.4). The EGCG-Se NPs have a significant dose-responsive inhibitory effect on α-amylase (6.1–15.9%) and α-glucosidase (2.8–5.7%). In vitro studies have demonstrated the potential of this complex for both the supplementation of selenium and blood glucose regulation, warranting further in vivo validation.

Background: Delayed wound healing, a common problem in patients with diabetes mellitus (DM), is associated with impaired keratinocyte autophagy. Epigallocatechin gallate (EGCG), a catechin, has been proven to promote diabetic wound healing. This study aims to explore the therapeutic mechanism of EGCG on diabetic wound healing. Methods: High glucose (HG)-induced keratinocytes and streptozotocin (STZ)-induced DM rats were prepared and intervened with EGCG to examine its therapeutic effects in in vivo and in vitro settings. The AMPK inhibitor, Compound C, was utilized to determine whether EGCG exerted its therapeutic effects through the AMPK/ULK1 pathway. Results: In vitro, EGCG improved HG-induced autophagy impairment in keratinocytes by increasing LC3II/LC3I, Becline1, and ATG5 levels and decreasing p62 level. Mechanically, EGCG activated the AMPK/ULK1 pathway, thereby promoting keratinocyte autophagy through the phosphorylation of AMPK and ULK1. Notably, EGCG promoted the proliferation, migration, synthesis and release of C-C motif chemokine ligand 2 (CCL2) in HG-treated keratinocytes. Furthermore, EGCG indirectly promoted the activation of fibroblasts, as evidenced by increased alpha-smooth muscle actin (α-SMA) and Collagen I levels. In vivo, EGCG promoted wound healing in DM rats, primarily by reducing inflammatory infiltration and increasing granulation tissue to promote wound epithelialization. Besides, EGCG promoted ATG5, KRT10, KRT14, TGF-β1, Collagen I, and α-SMA expressions in the neonatal epithelial tissues of DM rats. However, the use of Compound C reversed the effects of EGCG. Conclusions: These findings indicated that EGCG restored keratinocyte autophagy to promote diabetic wound healing through the AMPK/ULK1 pathway.

Introduction Preservation of porcine semen is essential for artificial insemination and genetic improvement in pig breeding programs. However, the overproduction of reactive oxygen species (ROS) and lower levels of protein phosphorylation emerge as two challenges during semen preservation. Inspired by the innate ligand-receptor binding biofunction of dopamine, herein, a dual-task nano-protectant that combines ROS-scavenging and protein phosphorylation-regulating properties via incorporating the natural antioxidant epigallocatechin gallate (EGCG) into polydopamine nanoparticles (EGCG@PDA NPs) was proposed to enhance the quality of pig semen during storage at 4 ℃. The results suggested that EGCG@PDA NPs significantly maintained sperm motility, acrosome integrity and mitochondrial membrane potential, extending semen storage time from 3 days to 10 days. Furthermore, EGCG@PDA NPs effectively scavenged excess ROS and inhibited ROS-mediated sperm apoptosis through the extracellular regulated protein kinases (ERK) signaling pathway. Intriguingly, EGCG@PDA NPs could degrade into ultrasmall particles (< 10 nm) in the semen or H 2 O 2 systems. These particles could target and activate the dopamine D2 receptor (D2DR) on membrane surface of sperm midpiece, thereby enhancing protein phosphorylation via the downstream cyclic adenosine monophosphate/protein kinase A (cAMP/PKA) signaling pathway, ultimately improving sperm motility parameters. This study presents a novel nano-strategy to boost the quality of pig semen, offering significant implications for the pig industry. Graphical

This study aimed to quantify the bitter–astringent interactive perceived intensity (BAIPI) of green tea based on the Weber–Fechner law by two-alternative forced choice test. Caffeine, (-)-epigallocatechin-3-gallate (EGCG), and Quercetin-3-o-rutinoside (Que-rut) were identified as the key bitter–astringent compounds. Then, BAIPI values were assessed based on the concentration ratios and ranges of caffeine, EGCG, and Que-rut in green tea according to the Weber–Fechner law. The BAIPI values were further fitted to a logarithmic curve:yBAIPIvalue=a×lnxconcentration of caffeine−b−c+IVinitialBAIPIvalue−1. The constant termsa,b, andcwere derived from the ratio of caffeine to EGCG, as the slope and X-intercept of the curves correlated exponentially with this ratio. The initial BAIPI value (IV) was calculated as:IVinitialBAIPIvalue=18.7960−0.3956q(theratioofcaffeinetoQue−rut). Validation showed a standard error of 1.5865 between estimated and actual BAIPI values. This method enables the estimation of bitter–astringent intensity in green tea by analyzing the concentrations of caffeine, EGCG, and Que-rut.

Cigarettes consumption is popular all over the world. In the meantime, green tea is widely accepted for their health benefits. Epigallocatechin-3-gallate (EGCG) is the most abundant catechin in green tea. This study aimed to investigate the effects of cigarette smoke (CS) exposure on human prostate cells and rat prostate, as well as the potential protective functions of EGCG. The results suggested that chronic CS exposure stimulated prostatic epithelial-mesenchymal transition (EMT), collagen deposition and fibrosis. Down-regulation of the Hedgehog (HH) signaling pathway helped to ameliorate CS-induced prostatic deficits. Meanwhile, EGCG could exhibit strong anti-fibrosis effects on prostate. Its underlying mechanisms were related to modulate EMT and decrease the activity of HH signal pathway.

Aim Epigallocatechin-3-gallate (EGCG), the major polyphenol found in green tea, exerts multiple protective effects against cardiovascular diseases, including cardiac hypertrophy. However, the molecular mechanism underlying its anti-hypertrophic effect has not been clarified. This study revealed that EGCG could inhibit pressure overload-induced cardiac hypertrophy by regulating the PSMB5/Nmnat2/SIRT6-dependent signalling pathway. Methods Quantitative real-time polymerase chain reaction and western blotting were used to determine the expression of mRNA and protein respectively. A fluorometric assay kit was used to determine the activity of SIRT6, a histone deacetylase. Luciferase reporter gene assay and electrophoretic mobility shift assay were employed to measure transcriptional activity and DNA binding activity respectively. Results EGCG could significantly increase Nmnat2 protein expression and enzyme activity in cultured neonatal rat cardiomyocytes stimulated with angiotensin II (Ang II) and heart tissues from rats subjected to abdominal aortic constriction. Nmnat2 knockdown by RNA interference attenuated the inhibitory effect of EGCG on cardiac hypertrophy. EGCG blocked NF-κB DNA binding activity induced by Ang II, which was dependent on Nmnat2 and the subsequent SIRT6 activation. Moreover the activation of PSMB5 (20S proteasome subunit β-5, chymotrypsin-like) was required for EGCG-induced Nmnat2 protein expression. Additionally, we demonstrated that EGCG might interact with PSMB5 and inhibit the activation of the proteasome. Conclusions These findings serve as the first evidence that the effect of EGCG against cardiac hypertrophy may be, at least partially, attributed to the modulation of the PSMB5/Nmnat2-dependent signalling pathway, suggesting the therapeutic potential of EGCG in the prevention and treatment of cardiac hypertrophy.

Zein nanoparticles are commonly used as colloidal emulsifiers to form and stabilize Pickering emulsions. However, the strong surface hydrophobicity of zein nanoparticles limits their widespread application. In this study, composite colloidal emulsifiers were fabricated from zein, sodium caseinate (NaCas), and epigallocatechin gallate (EGCG). Initially, NaCas-EGCG conjugates were formed using either an alkaline or enzymatic method. The enzymatic method led to conjugates containing more EGCG and with a higher thermal stability and surface hydrophilicity. Colloidal emulsifiers were prepared using an antisolvent precipitation method that involved titrating an ethanolic zein solution into an aqueous NaCas-EGCG conjugate solution. The potential application of these emulsifiers for forming and stabilizing high internal phase emulsions (HIPEs) was then explored. The emulsification properties of the zein nanoparticles were improved after they were complexed with NaCas-EGCG conjugates. Pickering HIPEs containing closely packed polygon oil droplets were formed from the colloidal emulsifiers, even at low particle concentrations (0.3% w/v). Overall, our results show that the functional performance of zein nanoparticles can be improved by complexing them with NaCas-EGCG conjugates. The novel colloidal emulsifiers developed in this study may therefore have useful applications in the food and other industries.

In recent years, molecularly imprinted membrane (MIM) has exhibited superiority in industrial separation, drug purification, solid-phase extraction and even medical detection owing to its remarkable selectivity. For catechins, however, the reactivity with free radicals limits the traditional construction of MIM towards such molecules. Here in this work, we have developed a bio-inspired MIM towards Epigallocatechin gallate (EGCG, a representative catechin) by the anchoring/imprinting strategy. The covalent interactions based on boronate-affinity supplied an oriented anchoring effect, while the non-covalent interactions from l -cysteine promoted the formation of imprinted sites that are highly matched with template. Mild and efficient imprinting was carried out by self-polymerization of dopamine to avoid the destruction of templates. As a result, remarkable selectivity coefficients (2.70 and 4.58), permselectivity coefficients (more than 3.19 and 3.41) and separation factors (5.18 and 7.00) were achieved on the EGCG-imprinted membrane (EIM), accompanied by the impressive imprinting factor (10.14) and superior regeneration performance (only 4.4% of the decline in five cycles). For deep insight into selective recognition and separation, microscopic and visualized mechanisms of site formation and specific rebinding was proposed by molecular dynamics simulations. Approaches with such an anchoring/imprinting strategy will enrich the avant-garde molecular imprinting strategies, and also expand membrane-based applications.

In this study, soybean peptide-succinic acid-modified cyclodextrin (SPT-SACD) nanoparticles (NPs) were successfully fabricated by combining SPT and SACD using an antisolvent precipitation approach. The effects of the average molecular weight of SPT and the SPT/SACD mass ratio on the structure and properties of the SACD-SPT NPs were investigated. Under optimal conditions, the SPT/SACD mass ratio was 2:1, and the SPT average molecular weight was 300 Da. SPT-SACD NPs were prepared under these conditions were spherical and had good uniformity. The particle sizes by DLS of SPT 1 (300 Da) /SACD and SPT 2 (500 Da) /SACD were in the range of 250–400 nm. The interaction between α-amylase and SPT-SACD NPs was investigated using ultraviolet visible (UV–Vis) absorption, fluorescence, and circular dichroism (CD) spectroscopy. The results of the fluorescence spectra and CD spectroscopy suggested that the presence of SPT-SACD NPs changed the microenvironment of the aromatic amino acid residues, which leads to the change of enzyme protein structure. The SPT-SACD NPs statically quenched the intrinsic fluorescence of the α-amylase by forming a complex with the enzyme. Moreover, the SPT-SACD NPs significantly improved the inhibitory effect of EGCG on α-amylase. The semi-inhibitory concentration (IC 50 ) decreased from 0.324 to 0.248 mg/mL. This study provides an improved understanding of the interaction mechanism between polypeptide-cyclodextrin complexes and digestive enzymes, which may facilitate the design of functional foods.

Numerous studies propose that epigallocatechin-3-gallate (EGCG), an abundant polyphenol in green tea, has anti-cancer properties. However, its mechanism of action in breast cancer remains unclear. This study investigated the capacity of EGCG to suppress breast cancer cell growth in vitro and in vivo, characterizing the underlying mechanisms, focusing on the effect of EGCG on glucose metabolism. EGCG reduced breast cancer 4T1 cell growth in a concentration- (10–320 μM) and time- (12–48 h) dependent manner. EGCG induced breast cancer apoptotic cell death at 24 h, as evidenced by annexin V/PI, caspase 3, caspase 8 and caspase 9 activation. Furthermore, EGCG affected the expression of 16 apoptosis-related genes, and promoted mitochondrial depolarization. EGCG induced autophagy concentration-dependently in 4T1 cells by modulating the levels of the autophagy-related proteins Beclin1, ATG5 and LC3B. Moreover, EGCG affected glucose, lactate and ATP levels. Mechanistically, EGCG significantly inhibited the activities and mRNA levels of the glycolytic enzymes hexokinase (HK), phosphofructokinase (PFK), and lactic dehydrogenase (LDH), and to a lesser extent the activity of pyruvate kinase (PK). In addition, EGCG decreased the expression of hypoxia-inducible factor 1α (HIF1α) and glucose transporter 1 (GLUT1), critical players in regulating glycolysis. In vivo, EGCG reduced breast tumor weight in a dose-dependent manner, reduced glucose and lactic acid levels and reduced the expression of the vascular endothelial growth factor (VEGF). In conclusion, EGCG exerts an anti-tumor effect through the inhibition of key enzymes that participate in the glycolytic pathway and the suppression of glucose metabolism.

Background: Prostate cancer (PCa) is characterized by a unique metabolic dependency on the hyperactivated tricarboxylic acid (TCA) cycle, creating a distinct vulnerability to cuproptosis-a copper-dependent form of regulated cell death. However, metabolic plasticity allows tumor cells to evade single-pathway inhibition through compensatory glycolysis, limiting the efficacy of copper-based therapies. Therefore, there is an urgent need to develop integrated nanoplatforms capable of orchestrating a dual-pathway metabolic blockade to overcome this therapeutic resistance. Results: Herein, we engineered a self-assembling copper-epigallocatechin gallate (EGCG) nanoreactor (Cu-EQ NP) designed to execute a synchronized dual-metabolic assault. The Cu-EQ NPs leverage the tumor microenvironment to release copper ions that trigger TCA-dependent cuproptosis, while the EGCG component simultaneously inhibits key glycolytic enzymes to prevent metabolic escape. Mechanistically, this lethality is self-amplified by EGCG-quinone-mediated depletion of glutathione (GSH) and the downregulation of the ATPase copper-transporting beta (ATP7B) efflux pump, leading to irreversible intracellular copper accumulation. Furthermore, we demonstrate that this metabolic collapse is highly immunogenic. In a syngeneic RM-1 mouse model, Cu-EQ NP-induced immunogenic cell death (ICD) released damage-associated molecular patterns (DAMPs) that successfully transformed the immunologically "cold" tumor microenvironment. This remodeling was characterized by enhanced dendritic cell maturation, M2-to-M1 macrophage repolarization, and robust infiltration of cytotoxic CD8 + T cells, resulting in potent tumor regression. Conclusions: The Cu-EQ nanoreactor represents a precision nanomedicine strategy that converts the specific metabolic vulnerabilities of PCa into a fatal weakness. By integrating dual-metabolic disruption with robust immune activation, this platform offers a promising therapeutic paradigm for overcoming resistance in advanced prostate cancer.
