Malathion is an organophosphorus pesticide which could remain in agricultural products and exert irreversible harmful effects on human health. Hence, strict monitoring of malathion contents is very significant. Here, a highly sensitive fluorescent aptasensor was developed for the determination of malathion, the system was based on a cationic polymer-mediated fluorescence ‘turn-off’. In this system, malathion-specific aptamers were bound to cationic polymer through electrostatic interactions . To produce fluorescence resonance energy transfer (FRET), negatively charged upconversion fluorescent nanoparticles (UCNPs) and cationic-polymer encapsulated gold nanoparticles (GNPs) were combined. This combination resulted in fluorescence quenching , and the degree of quenching was correlated with the concentration of malathion. Under optimum conditions, the fluorescence intensities were observed to decrease linearly with the rising concentration of the malathion from 0.01 to 1 μM with a detection limit of 1.42 nM. Furthermore, the developed sensor possessed good selective recognition ability for malathion and was successfully used to detect malathion in adulterated tap water and matcha samples with high accuracy.

Hydrogen sulfide (H2S), a crucial endogenous gasotransmitter, plays a significant role in monitoring pathological and physiological processes as well as in the realm of food safety control. The detection of H2S poses numerous challenges due to the complexity of biological fluids and food samples and the high demands for biocompatible probe molecules. To address these challenges, we have developed an innovative silica nanoparticle-based Förster resonance energy transfer (FRET) system with pyrene (Py) and pyronine (Pyr) embedded as energy donors and acceptors (Pyr@Py-SiO2 NPs). This system not only enhances the biocompatibility of fluorescent probes but also effectively mitigates interference from complex samples in the detection of endogenous H2S through its dual-mode FRET response. Our study reveals that pyrene and pyronine units embedded within the silica nanoparticles facilitate efficient energy transfer, enabling both colorimetric and ratiometric dual-mode optical detection of H2S. This approach is highly sensitive (detection limit of 71.5 nM), rapid (response time of ≤10 s), and uninfluenced by common anions and biological thiols. Moreover, we have successfully applied this system to the detection of H2S in various practical scenarios, including human urine, onion slices, and food spoilage. This work provides an effective and practical method for monitoring H2S in complex biological systems and real-world food samples.

Alpha-synuclein (α-Syn) is a key protein of Parkinson’s disease (PD). Oligomers formed by misfolding and aggregation of α-Syn can cause many pathological phenomena and aggravate the development of PD. Therefore, sensitive and accurate detection of oligomers is essential to understanding the pathology of PD and beneficial to screening and developing new drugs against PD. Here, we demonstrated a simple and sensitive method to detect the early aggregation of α-Syn via Förster resonance energy transfer (FRET) technology. We performed systematic investigations of the FRET sensitizations, efficiencies, and donor-to-acceptor distances during α-Syn aggregation, which was proved to be more sensitive to reflect small distance changes in the early stage of α-Syn aggregation, especially for α-Syn oligomers. The FRET assays were also applied to study the influence of Ser129 phosphorylation (pS129) on the aggregation rate of α-Syn. Our results showed that pS129 modification promotes α-Syn aggregation and enhances the ability of preformed fibrils to induce monomer aggregation. pS129 also increased the cytotoxicity of α-Syn. These results are of great significance for a better understanding of the pathological mechanisms of PD and future PD drug development.

Diagnosis of pancreatic cancer (PC) at early stage remains
a challenge
due to the limited sensitivity and specificity of current biomarkers.
Here, we develop nanostructured magnetic particles (NMPs) exhibiting
highly sensitive and specific circulating tumor cells (CTCs) quantification
enabled by Förster resonance energy transfer (FRET), and demonstrate
precise diagnosis of PC from benign pancreatic tumors (BPT) and healthy
donors (HD). The NMPs are modified with aptamer, which can preferably
bind to cells with higher specificity and guarantee effective substitution
of acceptors. Compared with smooth magnetic particles (SMPs), NMPs
provide more binding sites for CTCs, which significantly enhances
the fluorescent signal and detection limit. An ultralow detection
limit (1 cell/mL) is achieved using this unique capture and detection
platform. Clinical validation demonstrates the superior diagnostic
performance of this CTCs-based approach, with 91.7% sensitivity in
detecting PC patients and 96.6% specificity. Receiver operating characteristic
(ROC) analysis reveals an area under the curve (AUC) of 0.957, outperforming
conventional biomarkers (CA19–9:0.905; CEA: 0.747; CA125:0.625).
Our strategy represents an important methodological progress for tumor
liquid biopsy, showing promise for clinical diagnosis, monitoring,
and evaluation.

Background Bisphenol A (BPA) is a common monomer in polycarbonate plastics and epoxy resins that can migrate into food and water under extreme conditions, posing significant health risks including endocrine disruption, infertility, and metabolic disorders. Conventional detection techniques such as HPLC, HPLC-MS, and ELISA offer high sensitivity but suffer from costly instrumentation, complex pretreatment, and limited field applicability. Aptamer-based biosensors provide an attractive alternative due to their high specificity, low cost, and ease of use. Dual-mode platforms integrating colorimetric and fluorescent signals can overcome the limitations of single-mode detection Results Here, we developed an electrostatic–FRET coupled dual-mode aptasensor for BPA detection using cysteamine-modified gold nanoparticles (CS-AuNPs) and a FAM-labeled aptamer. In the absence of BPA, FAM-aptamer adsorbed onto CS-AuNPs, leading to fluorescence quenching and colloidal stability. Upon BPA binding, aptamer conformational change suppressed electrostatic adsorption, resulting in fluorescence recovery and nanoparticle aggregation with a visible color shift. Under optimized conditions, the fluorescence mode displayed a linear response from 10–1000 nM with a detection limit of 1.19 nM, while the colorimetric mode showed a range of 25–1000 nM with a detection limit of 2.22 nM. The dual-mode system demonstrated high selectivity against BPA analogues and reliable recoveries (92.7–109.2%) in spiked tap water, confirming its accuracy and robustness in real sample analysis Significance This study presents a cost-effective and visually responsive aptasensor that integrates electrostatic adsorption and FRET for sensitive BPA detection. By combining fluorescence and colorimetric modes, the platform achieves complementary readouts, reducing false positives and enhancing detection reliability. The work highlights the potential of positively charged AuNPs in multimodal biosensing, offering a promising tool for environmental monitoring and food safety assessment.

Major stumbling blocks in the recognition and detection of virus are the unstable biological recognition element or the complex detection means. Here a fluorescent sensor based on virus-molecular imprinted polymers (virus-MIPs) was designed for specific recognition and highly sensitive detection of Japanese encephalitis virus (JEV). The virus-MIPs were anchored on the surface of silica microspheres modified by fluorescent dye , pyrene-1-carboxaldehyde (PC). The fluorescence intensity of PC can be enhanced by the principle of fluorescence resonance energy transfer (FRET), where virus acted as energy donor and PC acted as energy acceptor. The enhanced fluorescence intensity was proportional to the concentration of virus in the range of 24–960 pM, with a limit of detection (LOD, 3σ) of 9.6 pM, and the relative standard deviation was 1.99%. In additional, the specificity study confirmed the resultant MIPs has high-selectivity for JEV. This sensor would become a new key for the detection of virus because of its high sensitive, simple operation, high stability and low cost.

Mycotoxins are fatal threats in food safety due to their strong carcinogenesis and toxicity, thus requiring highly sensitive detections . Herein, different-sized quantum dots (QDs) were used to construct a Förster resonance energy transfer (FRET) based immunosensor for sensitive detection of aflatoxin B 1 (AFB 1 ) in rice grains. To avoid irregular aggregation between two kinds of QDs, monovalent monoclonal antibody (mAb)-labeled red QDs (~0.84 anti-AFB 1 mAbs per QD) and multivalent hapten-labeled green QDs (~6.8 AFB 1 per QD) were designed as acceptor and donor, respectively. The anti-AFB 1 mAbs and AFB 1 interactions promoted one or more acceptors bound with a multivalent AFB 1 -labeled donor, resulting in energy transfer from the green QDs to the red QDs. Various parameters that influence the immunoassay including reactant ratio of donor to acceptor, buffer pH value, buffer ionic strength and immunoreaction time were systematically investigated and optimized. With optimal conditions, the obtained energy transfer efficiency is proportional to the logarithm of AFB 1 concentration in a range over 0.19–16 pM (0.06–5 ng/mL), while offering a limit of detection of 0.13 pM (0.04 ng/mL) in rice extracts. The recovery rates of the intra-assay for spiked samples at AFB 1 concentrations of 0.1, 1.0, and 5.0 ng/mL were 83.27%±3.27%, 97.36%±4.55% and 83.04%±4.94%, respectively, and those for the inter-assay were 81.28%±6.11%, 95.97%±7.07%, and 82.78%±5.99%, respectively. Statistical analysis using t -test had no significant difference between the proposed FRET-based immunoassay and the commercial enzyme-linked immunosorbent assay kit.

Fluorescent probes based on fluorescence resonance energy transfer (FRET) are highly promising for diverse bioapplications. The key to constructing FRET probes is to confine the donor and acceptor within a sufficiently close distance. However, the commonly used covalent linkage often requires elaborate design and complex organic synthesis, and sometimes causes changes in the fluorescence properties of the donor and acceptor. Inspired by the binding between small molecules and protein in nature, herein, we propose a protein-mediated strategy to fabricate FRET probe. In such protein-mediated FRET (P-FRET) probe, protein acts as a carrier to simultaneously confine donor and acceptor in its cavity. As a proof of concept, we use bovine serum albumin (BSA) as a model protein, coumarin derivative as a donor and hydroxyl radical (·OH)-responsive dye fluorescein as an acceptor. Through a series of investigations, including binding parameters, fluorescence properties and detection performance, we prove that the construction of P-FRET probe is simple and feasible and the detection is sensitive. Our P-FRET strategy will provide new insights for the design of FRET probes.

Heparanase (HPA) is a multifaceted endo -β-glucuronidase, and its dysregulation facilitates cancer metastasis. Developing techniques for fast and sensitively monitoring HPA enzymatic activity is crucial for searching for molecular therapies targeting HPA. Herein, we developed a novel fluorescence resonance energy transfer (FRET)-based nanoprobe AuNCs-LMWH-AuNRs, with AuNCs@GSH-cys and AuNRs/end-NH 2 /side-SiO 2 attached to the non-reducing terminus and reducing terminus of low molecular weight heparin (LMWH), respectively. AuNCs@GSH-cys exhibited an absolute quantum yield of 1.1%. The absorption spectra of AuNRs/end-NH 2 /side-SiO 2 (825 nm for maximum longitudinal absorption) and the emission spectra of AuNCs@GSH-cys (824 nm for maximum emission) were precisely overlapping, further enhancing the efficiency of FRET. In the presence of HPA, the LMWH nanoprobe exhibited an ultrasensitive response with excitation/emission wavelength (lambda (ex) = 560 nm, lambda (em) = 824 nm). The probe presented a wide linear dynamic detection range (LDR) of 0.125 ng/μL - 0.01 μg/μL in vitro with a limit of detection (LODs) of 82.15 pM (0.43 pg/μL). The excellent selectivity and good fluorescence turn-on efficiency of the probe made it possible for one-step detection of cellular heparanase activity. High throughput screening of HPA inhibitors also can be accomplished using the highly efficient LMWH nanoprobe.

Hydrogen peroxide (H 2 O 2 ) is considered a significant biomarker in various diseases and could induce deleterious health problems at irregular physiological concentrations. Therefore, developing a simple, efficient biocompatible nanoprobe for trace amount H 2 O 2 detection with high sensitivity and specificity is of great help for early diagnosis and therapeutics. Herein, we designed amphiphilic poly(styrene-co-maleic anhydride) (PMSA)-encapsulated nanoclusters composed of upconversion nanoparticles (UCNPs) and manganese dioxide nanoparticles (MnO 2 NPs) at a specific ratio to produce a near-infrared (NIR) excited luminescent nanoprobe for H 2 O 2 detection. Our results revealed that the MnO 2 NPs tended to experience catalytic decomposition when exposed to H 2 O 2 , while the UCNPs were retained inside the PSMA encapsulation, causing recovery of the UCNP emission band at 470 nm in accordance with H 2 O 2 concentration. This luminescence recovery was linearly dependent on H 2 O 2 concentrations, yielding a limit of detection (LOD) of 20 nM. The easy-to-interpret H 2 O 2 nanoprobe also proved high selectivity in the presence of other interfering substances, and biocompatibility and water-dispersibility, making it an ideal candidate for real-time detection of disease-related H 2 O 2 in living organisms. Background The early breast cancer diagnosis is a great challenge for the treatment success. Traditional FRET biosensors often rely on a one-to-one output pattern, meaning that each target biomarker generates a single fluorescence signal, limiting both the analytical efficiency and diagnostic value. Furthermore, the use of single biomarker for early diagnosis has substantial drawbacks, as it often leads to incomplete or misleading results. The simultaneous detection of miR-155 and miR-105 offers a more robust and clinically relevant diagnostic tool, reducing the likelihood of false positives and allowing for a more accurate assessment of breast cancer at an early stage. Results In this work, a hand in hand catalytic hairpin assembly (CHA) centered FÖrster resonance energy transfer (HCA-FRET) biosensor is designed, which allows two-in-one analysis of breast cancer related two biomarkers. In this system, miR-155 and miR-105, which are involved in regulating the malignant proliferation of breast cancer, are designed as two inputs as the AND DNA logic gate. Hand in hand CHA amplification circuit is effectively triggered under dual miRNA (miR-155 and miR-105), and the resulting significantly high FRET detection signal is measured which satisfies the AND logic gate function for the simultaneous detection of dual miRNA allied with breast cancer. HCA-FRET biosensor is successfully applied to detect miR-155 (0.01–1000 nM) and miR-105 (0.01–1000 nM) with limit of detections as 0.02 and 0.05 pM, respectively. HCA-FRET has excellent selectivity for recognizing single base mismatches and interfering miRNAs, and shows good stability in different matrices. The FRET detection signal recorded from the serum samples of breast cancer patients is found much greater than that of the healthy controls, which effectively distinguishes breast cancer patients from the healthy persons. Significance The study reports a simple, unique HCA-FRET biosensor which undergo dual target miR-155 and miR-105 biomarkers detection enabling early breast cancer diagnosis. Compared with the traditional single marker detection or two signal output analysis methods, this strategy allows more accurate diagnosis of breast cancer patients from normal people. More importantly, through testing the clinical serum samples, we found that this method is expected to be used to distinguish breast cancer patients from normal people. Therefore, the proposed method has great potential applications for nucleic acid based clinical diagnostics.

The effective detection of CA15-3 is of great significance for the postoperative monitoring and early diagnosis of breast cancer. Here, a fluorescent biosensor based on the fluorescence resonance energy transfer (FRET) between the prismatic hollow Metal-polydopamine frameworks (PHMPF) and 6-carboxyfluorescein (FAM)-labeled protein aptamer (FAM-Apt) was established to detect CA15-3. Characterization of SEM, TEM, EDS, FT-IR, XRD and XPS confirmed that the synthesized PHMPF was a polyhedral prismatic hollow organometallic frame containing the elements of C, N, O, and Co. PHMPF can quench the fluorescent group modified on the aptamer efficiently. Under optimized conditions, the proposed fluorescent biosensor can detect CA15-3 low to 2.56 × 10 −5 U mL −1 . In addition, the excellent selectivity and spiked experiments indicate the proposed biosensor owns potential applications for the clinical diagnosis of tumor markers in body fluids.
