Nanoflower Structured HA-MnO2-TCPP Nanoplatform for MRI-Guided Synergistic Sonodynamic and Chemodynamic Tumor Therapy

Xinyi Guo,Mengmeng Wang,Danyang Qu,Hao Cui,Jiajing Gong,Qinglin Shen,Yang Han,Xingqi Pan,Zhenbao Liu,Meilin Shi

Sonodynamic therapy (SDT) offers a noninvasive strategy for tumor ablation through ultrasound-activated sonosensitizers that generate reactive oxygen species (ROS); however, its therapeutic efficacy is often limited by tumor hypoxia. Herein, we report the rational design of a multifunctional theranostic nanoplatform, HA-MnO2-TCPP nanoflowers, that integrates active tumor targeting, dual ROS amplification, and magnetic resonance imaging (MRI) guidance for synergistic sonodynamic and chemodynamic therapy. The MnO2 nanoflowers were loaded with the sonosensitizer 4-carboxyphenyl porphyrin (TCPP) and surface-modified with hyaluronic acid (HA). This design enables CD44 receptor-mediated tumor targeting and tumor microenvironment-responsive degradation. Upon exposure to the mildly acidic and reductive tumor microenvironment, MnO2 is reduced to Mn2+, leading to glutathione depletion, in situ oxygen generation, and alleviation of hypoxia, while the released Mn2+ catalyzes Fenton-like reactions to generate highly toxic hydroxyl radicals (•OH) and simultaneously serves as an efficient T1-weighted MRI contrast agent. Meanwhile, TCPP enhances singlet oxygen (1O2) generation upon ultrasound activation, enabling effective sonodynamic therapy. Through the coordinated activation of SDT and CDT under MRI guidance, HA-MnO2-TCPP nanoparticles achieve precise and efficient tumor suppression without reliance on photothermal mechanisms. This work provides a robust paradigm for tumor microenvironment-responsive nanotheranostics and highlights the potential of ultrasound-activated strategies for precision cancer therapy.

Fe³ ⁺-mediated formation and synergistic mechanism of PDA-Fe³ ⁺-HA ternary composite underwater lubrication film

Chaobao Wang,Binlu Zhang,Jiale Chen,Yuanke Cai,Zihao Sun

Water-based lubricants are environmentally sustainable, but their inherently low viscosity is not conducive to the formation of stable lubricating films. To address this challenge, we propose a biomimetic strategy that integrates polydopamine (PDA) adhesion and hyaluronic acid (HA) hydration lubrication via Fe³ ⁺ coordination. The PDA-Fe³ ⁺-HA (PFH) ternary composite was prepared stepwise: Fe³ ⁺ coordinated with PDA catechol groups and interacted with HA chains through ionic crosslinking and chelation, thereby anchoring HA onto PDA. Tribological tests of UHMWPE/tin bronze tribopairs lubricated with the optimal PFH composite (mass ratio) showed that, compared with pure water, the average friction coefficient and wear rate decreased by up to 43.93% and 60.28%, respectively. The lubrication mechanism involves three synergistic effects: (i) Fe³ ⁺-enhanced PDA anchoring forms a stable adhesive layer; (ii) surface-exposed HA chains provide a hydrated low-shear lubrication layer; (iii) Fe³ ⁺ coordination bridges PDA and HA, integrating adhesion and lubrication functions. This bioinspired design offers an eco-friendly lubricant additive strategy. Download: Download high-res image (247KB) Download: Download full-size image

Antibacterial Antimicrobial Peptide Grafted HA/SF/Alg Wound Dressing Containing AIEgens for Infected Wound Treating

Yize Lin,Yan Tan,Rong Deng,Li Gong,Xiaoshan Feng,Zhongqi Cai,Yanxian He,Longbao Feng,Biao Cheng,Yi Chen

Chronic wounds are characterized with excessive biofluid and persistent infection. Therefore, there is an urgent desire to develop a multifunctional wound dressing that can meet the extreme requirements including effective antibacterial and powerful wound microenvironment regulation and protection function to promote wounds heal quickly. In this study, a multifunctional composite dressing (HA-AMP/SF/Alg/Rb-BG-AIEgens) was synthesized by combining a mesoporous bioactive glass framework loaded with AIEgens (Rb-BG-AIEgens) with cross-linked antimicrobial peptide grafted hyaluronic acid (HA-AMP), sodium alginate (Alg), and silk fibroin (SF). It is important to note that the Rb-BG-AIEgens can achieve real-time and sensitive bacterial detection. HA-AMP can achieve broad spectrum antibacterial and avoid the residue of drug-resistant bacteria. The HA-AMP/SF/Alg/Rb-BG-AIEgens dressing can up-regulate related proliferative proteins, thereby promoting regeneration of tissue and the rapid healing of chronic wounds. With good biocompatibility and antibacterial ability, HA-AMP/SF/Alg/Rb-BG-AIEgens dressing has great potential to become a next generation wound dressing for clinical biological fluid management and chronic bacterial infection treatment.

Functionally graded triply periodic minimal surface scaffold of HA-Al2O3 via Vat photopolymerization 3D printing

Qiuyu Liu,Qinchao Tang,Zhifeng Huang,Zhiwei Li,Xuye Wang,Pin Wen,Yi Bai,Fei Chen

Hydroxyapatite (HA) is widely used in scaffolds due to its high similarity in the composition to natural bone. However, the weak and fragile characteristics of synthetic HA limit its application. The strength of HA can be increased by the incorporation of alumina (Al 2 O 3 ) to obtain composites with extraordinary compressive strength and biocompatibility. HA-Al 2 O 3 composites with different ratios were prepared by Vat photopolymerization 3D printing, and their shrinkage, porosity, phase composition and biocompatibility were systematically investigated. In addition, the Diamond structure of the scaffold was chosen to achieve the trade-off relationship between compressive strength and porosity. The results suggest that the HA-Al 2 O 3 composite with 20 vol% HA possesses the maximum compressive strength and 60 vol% HA has optimal biocompatibility. A compositional gradient scaffold from 60 vol% HA and 20 vol% HA was proposed to satisfy different needs of the different parts in scaffold for biocompatibility and compressive strength. The HA-Al 2 O 3 gradient scaffold achieves a preferable compressive strength of 13.7 MPa while possessing gradient biocompatibility.

Oral delivery of Silybin via HA-DOCA-functionalized zein nanocolloids for amelioration of liver fibrosis

Lin Wen,Jijiao Wu,Yuxin Zhao,Zihao Sun,Chuipeng Liang,Xiaolian Liu,Jia Luo,Xiaofang Li

Silybin (SIL), a natural flavonoid with recognized anti-hepatic fibrosis activity, suffers from poor aqueous solubility and low oral bioavailability, which restrict its clinical utility. To address these limitations, we developed a novel oral delivery system, termed HDZN/SIL, based on hyaluronic acid–deoxycholic acid (HA–DOCA)-modified zein nanoparticles loaded with SIL. This system was fabricated via an anti-solvent precipitation method. Under optimized conditions, specifically at a zein-to-SIL mass ratio of 10:1, the resulting nanoparticles displayed a uniform spherical morphology with a smooth surface and an average diameter of 127.9 ± 8.2 nm. The formulation also demonstrated high drug loading (97.55 ± 1.02 %) and excellent colloidal stability. Following oral administration, HDZN/SIL achieved effective liver-targeted delivery. In a mouse model of CCl₄-induced hepatic fibrosis, HDZN/SIL treatment resulted in significantly enhanced therapeutic efficacy compared to free SIL, along with an improved safety profile. These findings support HDZN/SIL as a promising strategy to advance the clinical application of SIL.

Solar Hydrogen Production by (HA)PbX4 (X = I, Br) 2D Perovskite Crystals Doped with MoS2

Lili Gao,Ke Hao,Ping Hu,Sheng Huang,Fan Yang,Deng Li,Shengzhong (Frank) Liu

Halide perovskites have gained significant attention in the field of photovoltaics and photocatalysis due to their exceptional optoelectronic properties. However, the exploration of other classes of 2D perovskite materials is limited. In this work, the photocatalytic performance of (HA)PbX 4 2D crystals embedded with MoS 2 in their bulk structure is investigated. The (HA)PbX 4 @MoS 2 composites are directly synthesized in a (HA)PbX 4 crystal solution. Notably, the (HA)PbI 4 @MoS 2 composite exhibits a remarkable 2.94-fold increase in H 2 evolution activity. Furthermore, the composite demonstrated stability in a (HA)PbX 4 (HA = histamine, X = I, Br, Cl)-saturated aqueous HX solution. The photogenerated electrons in (HA)PbX 4 are efficiently transferred to the MoS 2 sites, facilitating the reduction of protons to H 2 . In this study, the potential of hybrid 2D perovskite–MoS 2 composites is highlighted as promising visible-light photocatalysts for aqueous solution applications.

Bioengineered functional scaffolds co-loaded with osteonectin and HA-internalizing osteoblasts for boosting bone regeneration

Wei Yang,Qin Zou,Chenxin Wang,Yuankun Ren,Rui Zhang,Mingyue Lin,Xueqi Zhang,Yu Liu,Boyang Wang,Wei Li,Li Jiang,Xiyu Li

Traditional bone repair scaffolds often struggle to meet the demands of complex bone defect regeneration due to insufficient early osteogenic stimulation or osteogenesis. In this study, we proposed a synergistic strategy to combine PHA (PCL/HA) scaffolds with HA-internalized osteoblasts (OHA) and osteonectin (ON), where OHA provides fast calcium and phosphate ion release via lysosomal degradation, and ON facilitates calcium phosphate–collagen binding to jointly enhance the early bone regeneration. This “OHA + ON” dual-pathway synergy helps to overcome the limitations of poor early osteogenesis or insufficient Ca/P ion release. The results demonstrated that the PHA + ON+OHA combination was much superior to PHA or PHA + ON scaffolds in terms of the osteogenic gene expression, osteoblast proliferation and mineralization in vitro , and the osteogenic capability in subcutaneous model and femoral defect model in vivo . These findings suggest that the “OHA + ON” synergistic strategy realizes an early pro-osteogenesis, optimizes the osteogenic process, and elevates the new bone quality, offering a viable approach for future clinical repair of complex bone defects.

Synergistic anti-inflammatory and osteogenic n-HA/resveratrol/chitosan composite microspheres for osteoporotic bone regeneration

Limei Li,Mali Yu,Yao Li,Qing Li,Hongcai Yang,Meng Zheng,Yi Han,Di Lu,Sheng Lu,Li Gui

The development of functional materials for osteoporosis is ultimately required for bone remodeling. However, grafts were accompanied by increasing pro-inflammatory cytokines that impaired bone formation. In this work, nano-hydroxyapatite (n-HA)/resveratrol (Res)/chitosan (CS) composite microspheres were designed to create a beneficial microenvironment and help improve the osteogenesis by local sustained release of Res. Study of in vitro release confirmed the feasibility of n-HA/Res/CS microspheres for controlled Res release. Notably, microspheres had anti-inflammatory activity evidenced by the decreased expression of pro-inflammatory cytokines TNF-α, IL-1β and iNOS in RAW264.7 cells in a dose dependent manner. Further, enhanced adhesion and proliferation of BMSCs seeded onto microspheres demonstrated that composite microspheres were conducive to cell growth. The ability to enhance osteo-differentiation was supported by up-regulation of Runx2, ALP, Col-1 and OCN, and substantial mineralization in osteogenic medium. When implanted into bone defects in the osteoporotic rat femoral condyles, enhanced entochondrostosis and bone regeneration suggested that the n-HA/Res/CS composite microspheres were more favorable for impaired fracture healing. The results indicated that optimized n-HA/Res/CS composite microspheres could serve as promising multifunctional fillers for osteoporotic bone defect/fracture treatment.

Fabrication of HA/PEI-functionalized carbon dots for tumor targeting, intracellular imaging and gene delivery

M. Zhang,X. Zhao,Z. Fang,Y. Niu,J. Lou,Y. Wu,S. Zou,S. Xia,M. Sun,F. Du

Carbon quantum dots (CDs) as emerging carbon nano-materials have attracted tremendous attention in biomedical fields due to unique properties. In this study, hyaluronate (HA) and polyethylenimine (PEI) functionalized carbon dots (HP-CDs) were synthesized by a facile bottom-up method for tumor targeting and gene delivery. After HA modification, the HP-CDs exhibited superior dispersibility in water and good biocompatibility and were internalized readily into the cytoplasm of cancer via HA-receptor mediated endocytosis. Meanwhile, the HP-CDs with PEI functionalization were shown to have excellent gene condensation capability via electrostatic attraction and protective capacity by preventing nuclease degradation. By virtue of good photoluminescence properties, the HP-CDs as a gene carrier were successfully applied to intracellular imaging and gene delivery. Taken together, the resultant HP-CDs displayed great potential in tumor targeting, intracellular imaging and gene delivery.

Laser fabrication of Ag-HA nanocomposites on Ti6Al4V implant for enhancing bioactivity and antibacterial capability

Xiangmei Liu,H.C. Man

For titanium alloy implants, both surface bioactivity and antibacterial infection are the two critical factors in determining the success of clinical implantation of these metallic implants. In the present work, a novel nanocomposite layer of nano-silver-containing hydroxyapatite (Ag-HA) was prepared on the surface of biomedical Ti6Al4V by laser processing. Analysis using SEM, EDS and XRD shows the formation of an Ag-HA layer of about 200   μm fusion bonded to the substrate. Mineralization tests in simulated body fluid (SBF) showed that laser fabricated Ag-HA nanocomposite layer favors the deposition of apatite on the surface of the implants. Antibacterial tests confirmed that all Ag-HA nanocomposite layers can kill bacteria while a higher Ag content would lower the cytocompatibility of these coatings. Cell viability decreases when the Ag content reaches 5% in these coatings, due to the larger amount of Ag leached out, as confirmed by ion release evaluation. Our results reveal that laser fabricated Ag-HA nanocomposite coatings containing 2% Ag show both excellent cytocompatibility and antibacterial capability.

Fabrication and cell viability of injectable n-HA/chitosan composite microspheres for bone tissue engineering

Bin Cai,Qin Zou,Yi Zuo,Limei Li,Boyuan Yang,Yubao Li

Composite microspheres based on nano-hydroxyapatite (n-HA)/chitosan (CS) with different weight ratios were fabricated by an emulsion method and using natural vanillin as a cross-linking agent. FT-IR, XRD, TGA, SEM and laser particle size analysis were used to characterize their physico-chemical properties. Live-dead cell staining and MTT assay were used to evaluate cell viability and cytocompatibility. The results showed that the rod-like n-HA crystals could be incorporated into the CS matrix in a relatively high level up to 29.57 wt%. The encapsulation efficiency of n-HA in the composite microspheres was between 29.50–73.93%. The composite microspheres with a diameter of 10.9–55.0 μm and good swelling properties should be easy to inject into irregular bone defects. The studies of in vitro biodegradation and cell culture using MG63 cells demonstrated that the vanillin-crosslinked n-HA/CS microspheres could be degraded in a sustained and controlled behavior while supporting cell growth, thus acting as worthy candidates for a multifunctional injectable matrix in bone tissue engineering.

Enhanced healing activity of burn wound infection by a dextran-HA hydrogel enriched with sanguinarine

Qiyu Zhu,Ming Jiang,Qiang Liu,Shina Yan,Longbao Feng,Yong Lan,Guiqiu Shan,Wei Xue,Rui Guo

Burn wounds are associated with a series of risks, such as infection and pathologic scar tissue formation, which significantly delay wound healing and lead to complications. In this study, we successfully fabricated a dextran-hyaluronic acid (Dex-HA) hydrogel enriched with sanguinarine (SA) incorporated into gelatin microspheres (GMs), which had high porosity, good swelling ratio, enhanced NIH-3T3 fibroblast cell proliferation, and sustained SA release profile. The in vitro degradation results indicate that the SA/GMs/Dex-HA hydrogel can be degraded. The in vitro antibacterial tests showed that the SA/GMs/Dex-HA hydrogel can inhibit methicillin-resistant Staphylococcus aureus (MRSA) and Escherichia coli (E. coli). We evaluated the wound-healing effects and antibacterial properties of SA/GMs/Dex-HA hydrogels in a rat full-thickness burn infection model. The hematoxylin–eosin (H&E) and Masson's trichrome staining results of the SA/GMs/Dex-HA hydrogel showed that it improved re-epithelialization and enhanced extracellular matrix remodeling, and immunohistochemistry results showed that the expression of TGF-β1 and TNF-α was decreased, while the TGF-β3 expression was increased. Our findings suggest that the SA/GMs/Dex-HA hydrogel provides a potential way for infected burn treatment with high-quality and efficient scar inhibition.
