Laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) has been widely used for in situ microanalysis of elemental concentrations in solid samples, but its application is hampered by a lack of matrix-matched and homogeneous reference materials (RMs). In this work, a new method for preparing matrix-matched and homogeneous LA-ICP-MS reference materials has been developed. A multiple rare earth element (REE) doped CaWO4 single crystal (CaW-1) was successfully prepared by the Czochralski technique. The matrix composition and crystal structure were confirmed by X-ray diffraction (XRD) and electron microprobe analysis (EMPA), which indicates that these crystals have a matrix composition of CaWO4 and are matrix-matched with natural scheelite. The homogeneity of REE distribution was systematically studied by seven methods and the results demonstrated that REEs are distributed homogeneously in the CaWO4 single crystal with slight fluctuation (RSDs: ca. 4%). It can be concluded that the synthesized CaWO4 single crystal meets both crucial requirements, matrix-matched and homogeneous distribution, for an LA-ICP-MS reference material, and therefore it was used to quantitatively calibrate REE concentrations in natural scheelite. The results showed that acceptable analytical errors (<8%) were obtained, which were more desirable than those obtained with non-matrix-matched NIST SRM 610 as the RM. It robustly validates the ability of the developed method for preparing CaWO4 single crystals as possible LA-ICP-MS reference materials to calibrate REE concentrations in natural scheelite.

Developing quantification and characterization methodology for metallic nanoparticles (MNPs) and their ionic component in complex matrix are crucial for the evaluation of their environmental behavior and health risks to humans. In this study, reversed phase high performance liquid chromatography combined ICP-MS was established for the characterization of MNPs in complex matrix. The ionic component could be separated from NPs with the optimized parameters of aqueous mobile phase . Good linear relationship between average diameter and retention time of NPs was obtained using HPLC-ICP-MS and the size smaller than 40 nm could be determined with this method, the detected results were in accordance with TEM results. The low detection limit of AuNPs and Au(Ⅲ) (both in sub-μg/L level) showed that this method was promising for the characterization of AuNPs and Au(Ⅲ) in environmental water. The mass concentration of ionic Au(Ⅲ) in environmental water could be detected using the proposed HPLC-ICP-MS and the concentration of AuNPs was obtained by subtracting the Au(Ⅲ) concentration from the total Au (The concentration of total Au was detected by ICP-MS after microwave digestion). Furthermore this proposed HPLC-ICP-MS method and single particle-ICPMS (SP-ICP-MS) was used for the analysis of the Ag speciation in commercial antibacterial products.

In this work, we developed an immunoassay based on tyramide signal amplification (TSA) and gold nanoparticles (Au NPs) labeling for highly sensitive detection of alpha fetoprotein (AFP) by inductively coupled plasma mass spectrometry (ICP-MS). AFP was captured by anti-AFP1 coating on the 96-well plate and labeled by anti-AFP2-horseradish peroxidase (HRP), in which the HRP can catalyze the deposition of biotinylated tyramine on the nearby protein. Then the streptavidin (SA)-Au NPs was labeled on the deposited biotinylated tyramine as the intensive signal probe for ICP-MS measurement. Under the optimal experimental conditions, the limit of detection of the developed method for AFP was 1.85 pg/mL and the linear range was 0.005–2 ng/mL. The relative standard deviation for seven replicate detections of 0.01 ng/mL AFP was 5.2%. The proposed method was successfully applied to the detection of AFP in human serum with good recoveries. This strategy is highly sensitive and easy to operate, and can be extended to the sensitive detection of other biomolecules in human serum.

Water contamination caused by heavy metals represents an urgent global issue. Cu 2+ , a potential trace heavy metal pollutant, can accumulate in the human body through the food chain, leading to excessive levels that give rise to diverse health complications. Hence, in this investigation, a novel and efficacious fluorescent probe named Eu-BTB was developed for the detection of Cu 2+ , employing 1,3,5-triphenyl(4-carboxyphenyl) (H 3 BTB) as the ligand and Eu 3+ as the metallic framework. The probe demonstrates exceptional fluorescence characteristics. The interaction between the probe ligand BTB and Eu 3+ triggers an antenna effect, heightening the emission efficiency of Eu 3+ while preserving its intrinsic emission. The introduction of Cu 2+ competes with BTB for binding, thus quelling the antenna effect and inducing a fluorescence alteration. Within the concentration range of 0.05–10 μM, the fluorescence intensity-to-Cu 2+ concentration ratio exhibits a robust linear correlation, with a remarkably low detection limit of 10 nM and a rapid response time of 3 min. The fluorescent probe has been effectively deployed for the detection of copper ions in water across diverse environmental conditions, with the obtained outcomes being validated via the conventional approach of inductively coupled plasma mass spectrometry (ICP-MS). The Eu-BTB probe showcases the advantages of simplicity, swiftness, and broad applicability, thus affirming its potential for the prompt and accurate detection of Cu 2+ in diverse environmental water samples.

Background Tin and lead contamination is a global threat to marine ecosystems considering their species-specific toxicity, bioavailability and mobility. Hence simultaneous measurement of multiple tin and lead compounds at μg L −1 to pg L −1 levels in environmental water is always an indispensable but challengeable task. High performance liquid chromatography coupled with inductively coupled plasma mass spectrometry (HPLC-ICP-MS) is one of the most widely used choices for this purpose because of good sensitivity, strong separation power and good compatibility. Previous HPLC-ICP-MS methods based on a single elemental speciation strategy are low-efficiency and sensitivity-insufficient for a large set of unstable samples and interaction of multiple metal(loid)s down to ng L −1 levels. Results In this study, we developed a sensitive, efficient and environment-friendly analytical method for accurate quantification of inorganic and organic species of tin and lead simultaneously based on HPLC-ICP-MS with online integration of solid phase extraction (SPE). By using graphene oxide modified silica conditioned with 1 mM benzoic acid to enrich tin and lead species from 10 mL sample, detection limits were improved to 2–8 pg per liter due to satisfactory enrichment factors (522−2848 folds). The SPE-HPLC-ICP-MS method was applicable to quantification of ultra-trace tin and lead species at pg L −1 levels in uncontaminated seawater . Tributyltin was the only tin species detected at subnanograms per liter levels while Pb(II) was the only lead species detected at several nanograms per liter in thirteen coastal seawater samples collected in Hangzhou Bay, indicating light contamination of tin and lead. Significance Overall, the proposed SPE-HPLC-ICP-MS method is highly sensitive, efficient and environment-friendly that are fairly suitable to routine speciation analysis of tin and lead in environmental, food, and biological samples.

A new method of solidified floating organic drop microextraction (SFODME) combined with electrothermal vaporization (ETV)-inductively coupled plasma mass spectrometry (ICP-MS) was developed for the determination of trace heavy metals in environmental water samples with sodium diethyldithiocarbamate (DDTC) as both chelating reagent in SFODME and chemical modifier in ETV. The factors affecting the microextraction efficiency were studied in detail and the optimal extraction conditions were established. Under the optimal conditions, the limits of detection (LODs) for SFODME-ETV-ICP-MS determination of Co, Pd, Cd, Hg, Pb and Bi were found to be 0.0060, 0.0091, 0.0020, 0.0041, 0.0170 and 0.0041 ng mL −1 , respectively, with the relative standard deviations (RSDs) of 2.8–10.0% ( c = 0.5 ng mL −1 , n = 7). The developed method was successfully applied to the analysis of six target metals in Yangtze River and East Lake water samples with recoveries ranging from 77.7 to 119.1%. To validate the accuracy of the method, a certified reference material of Environmental Water (GSBZ50009-88) was analyzed and the determined values were in good agreement with the certified values.

Background Molybdenum (Mo) isotope has been widely used to trace various redox processes in earth science, however, conventional separation methods, typically depend on toxic hydrofluoric acid (HF), redox reagents (e.g., hydrogen peroxide (H 2 O 2 ) and ascorbic acid (HAsc)), or multi-column purification steps, which introduce either safety hazards or operational complexity. In this study, a safe and efficient single-column procedure without use of either HF or redox reagents was developed for Mo separation from various samples for high precision Mo isotope measurements. Results This method takes advantage of high selective adsorption of Mo on an in-house TOPO resin and only utilize the diluted HCl - HNO 3 media, achieving an effective matrix removal, a high Mo recovery (>93 %) and low procedural blanks (<0.50 ng). Repeated analyses of NIST SRM 3134, Mo metal, and seawater CASS-6 yielded an external reproducibility better than ±0.06 ‰ (2SD). Mn-doping experiments revealed that Mn/Mo ratios exceeding 0.50 induced significant polyatomic interference, causing measurable negative shifts (>0.10 ‰) in δ 98/95 Mo NIST SRM 3134 values. The method was validated using 11 geological reference materials, showing good agreement with published values. Additionally, the δ 98/95 Mo NIST SRM 3134 values of five biological reference materials (e.g., green tea, human hair, pig liver, and fetal bovine serum) were reported for the first time, exhibiting a significant isotopic variation of 1.44 ‰. Significance and novelty Compared with anion/cation-exchange resins or extraction resins, this novel procedure based on in-house TOPO resin depends on dilute HCl - HNO 3 media and effectively purifies Mo from the matrix, with good performance and operational safety. Additionally, the δ 98/95 Mo NIST SRM 3134 values for biological reference materials provide valuable references for quality assurance and inter-laboratory comparisons in emerging applications of Mo isotopes in biological systems. Overall, this study provided a simpler and safer alternative method for high-precise determination of Mo isotope in various samples.

Methamphetamine (METH) is one of the most highly addictive illicit drugs abused all over the world. Much evidence indicates that METH abuse leads to major toxicity, medical consequences, and even severe public health consequences. Existing studies usually focus on the pathomechanism of METH-induced toxicity; therefore, data on metabolites and elements correlating with particular toxicity remain scarce. The objective of the present study is to develop appropriate analytical procedures to identify the differential metabolic and elemental biomarkers on METH-induced hepatic injury to rats. The rats were administrated with METH (15 mg/mL/kg, two times per day) via intraperitoneal (i.p.) injections for four consecutive days. The alanine aminotransferase and aspartate aminotransferase activity levels of in the rat serum of the METH group increase significantly compared with those of the control group, suggesting obvious hepatic injury. The results are further confirmed by the histopathological microscopic observation. A total of 18 small molecular metabolites and 19 elements are selected to perform the simultaneous quantification based on the combination of liquid chromatography coupled with tandem mass spectrometry and inductively coupled plasma mass spectrometry. Sample preparation was optimized to cover all the analytes. Both methods are optimized and validated according to developed guidelines such as limits of detection, limits of quantification, linearity, precision, and recovery. All the obtained data are within the satisfactory range. The normalized data were processed according to the partial least squares discrimination analysis (PLS-DA) model. Five differential metabolic and six elemental markers are identified in rat plasma based on the variable importance in projection (VIP) (> 1) and t test results. Overall, the results obtained in this study demonstrate the developed methods are suitable for simultaneous determination of metabolic and elemental markers in the hepatic injury to rats induced by METH.

A simple and fast method of low-density solvent based dispersive liquid-liquid microextraction (LDS-DLLME) followed by high-performance liquid chromatography-inductively coupled plasma mass spectrometry (HPLC-ICP-MS) has been developed for the speciation analysis of organoarsenic and inorganic arsenic in water samples. The low-density solvent (octanol) was given as the organic phase and injected into the aqueous sample (donor phase) with methanol as the disperser. With As (V), As (III), p-APAA, 4-HPAA, ROX and PAA as target species, factors of LDS-DLLME including pH value, anionic carriers, elution conditions and extractant, were studied in detail. Besides, volumes of solvents were further optimised by response surface methodology. Under the optimal conditions, the limits of detection for four phenylarsenics and arsenate were in the range of 0.001–0.039 μg L−1. The relative standard deviations (RSDs) were 3.6–9.4% and the enrichment factors varied from 6.2 to 70.8. The proposed method of LDS-DLLME-HPLC-ICP-MS was satisfactorily applied to the determination of six arsenic compounds in water samples with recoveries of 81.8–111.7% for the spiked lake water samples.

Radionuclide uranium has both a chemical and radioactive toxicity, leading to severe nephrotoxicity as it predominantly deposits itself in the kidneys after entering into human bodies. It crosses renal cell membranes, accumulates in mitochondria and causes mitochondrial oxidative damage and dysfunction. In this study, a mitochondria-targeted heptamethine indocyanine small molecule chelator modified with gallic acid (IR-82) is synthesized for uranium detoxication. Both gallic acid and sulfonic acid, as two hydrophilic endings, make IR-82, being excreted feasibly through kidneys. Gallic acid with polyphenol groups has a steady metal chelation effect and potent antioxidant ability, which may facilitate IR-82-alleviated uranium nephrotoxicity simultaneously by enhancing uranium decorporation from the kidneys and reducing mitochondrial oxidative damage. Cell viability assays demonstrate that IR-82 can significantly improve the cell viability of uranium-exposed human renal (HK-2) cells. It is also demonstrated to accumulate in mitochondria and reduce mitochondrial ROS and total intracellular ROS, as well as intracellular uranium content. In vivo imaging experiments in mice show that IR-82 could be excreted out through kidneys. ICP-MS tests further reveal that IR-82 can efficiently decrease the uranium deposition in mouse kidneys. IR-82 treatment improves the animal survival rate and renal function of experimental mice after high-dose uranium exposure. Collectively, our study may evidence that the development of uranium decorporation agents with kidney–mitochondrion dual targeting abilities is a promising strategy for attenuating uranium-induced nephrotoxicity.

A novel Schiff base-chitosan-grafted multiwalled carbon nanotubes (S-CS-MWCNTs) solid-phase extraction adsorbent was synthesized by covalently grafting a Schiff base-chitosan (S-CS) onto the surfaces of oxidized MWCNTs. The adsorbent was characterized by Fourier-transform infrared spectroscopy, transmission electron microscopy, and thermal gravimetric analysis. The results showed that S-CS was successfully grafted onto the surfaces of MWCNTs. A method was developed for the determination of heavy metals, namely V(V), Cr(VI), Cu(II), As(V) and Pb(II) in biological and environmental samples by inductively coupled plasma mass spectrometry coupled with preconcentration with S-CS-MWCNTs. The parameters influencing preconcentration of target ions, such as the pH of the sample solution, the flow rate of sample loading, the eluent concentration, and eluent volume, were investigated and optimized. Under the optimal conditions, the enrichment factors of V(V), Cr(VI), Cu(II), As(V), and Pb(II) reached 111, 95, 60, 52, and 128, respectively, and the detection limits were as low as 1.3–3.8 ng L −1 . The developed method was successfully applied to the determination of trace-metal ions in herring, spinach, river water, and tap water with good recoveries ranging from 91.0% to 105.0%.

This study aimed at developing an accurate method for determining the levels of total arsenic (As) and arsenic species in edible shrimp with the collision/reaction cell (CRC) in the O2 mass-shift mode to remove interferences. The microwave-assisted extraction method demonstrated satisfactory efficiency (86.9%), and As species were separated rapidly within 11 min when the optimized chromatographic conditions were adopted. During the study, the limits of detection, precision, and spike recovery for six arsenic species were in the ranges from 0.010 to 0.028 μg/L, 1.89 to 3.14%, and 96.9 to 103.2%, respectively. The analytical methods were successfully applied to determine the concentrations of total arsenic and As species in eight shrimp samples. The results showed that arsenobetaine was the predominant As species, and inorganic arsenic contents were below 0.5 mg/kg in the shrimp samples analyzed. The health risk assessment indicated that the shrimp samples detected posed no particular risk of As to local consumers.
